Sweetener and Flavor Enhancer Food Additives in Industrial Food Products Marketed in Dakar: Frequency and Diversity

Sweeteners and flavor enhancers are food additives widely used in industry, respectively, to add sweetness and flavor to foods. However, the presence of these substances is often criticized by consumers for their effects on health. What’s more, some scientific studies link these substances to certain pathologies. To guarantee food safety, competent authorities should have food standards based on risk analysis using consistent, reliable data. However, in developing countries, such data is often weak or non-existent. The aim of this study is therefore to carry out a pilot survey to establish the profile of sweeteners and flavour enhancers present in industrial food products marketed in Senegal. The methodology consisted of sampling various food products sold on the Senegalese market, based on analysis of labels containing information on ingredients, including additives. The investigation involved nine stores, one supermarket and 5 mini-markets in Dakar. The results showed the presence of 6 taste enhancers in food products, the most frequent being sodium L-monoglutamate (E621), inosinate (E631) and disodium guanylate (E627). Solid broths are the foodstuffs with the highest number of taste exhalters. As for sweeteners, 12 substances were identified, the most frequent being acesulfame potassium (E950), aspartame (E951), sucralose (E955) and saccharin (E954). Given the potential health risks associated with the consumption of these food additives, their control and monitoring on the market should be a priority for the competent authorities.

Macrocycle-based glycoclusters, on account of their promising anti-adhesive properties against bacteria,are potential therapeutic alternatives to classic antibiotics through the much less explored anti-adhesive strategy. In this study, a series of constitutionally-pure pentavalent glycoclusters was prepared by conjugating assorted azido-carbohydrates onto a penta-propargyl rim-differentiated pillar[5]arene(RD-P[5])scaffold through Cu(I)-catalyzed azide–alkyne cycloaddition “click” reactions. Their binding towards therapeutically relevant bacterial lectins, such as Lec A and Lec B from Pseudomonas aeruginosa and concanavalin A(Con A), were evaluated subsequently by isothermal titration calorimetric studies. Most of these isomerfree RD-P[5] pentavalent glycoclusters, except the fucosylated ones, display good affinities to ***, the dissociation constants observed are similar to those displayed by an analogous pentavalent glycocluster consisting of four P[5] constitutional isomers, in which the RD-P[5] component merely accounts for 7% in the mixture. Our results revealed that high constitutional purity is not essential for achieving effective multivalent interactions between P[5]-based glycoclusters and lectins, presumably as a result of the conformationally labile nature of the P[5] scaffold. This information provides valuable design principles for low-cost and facile syntheses of glycosylated P[5]s for biomedical applications.

In Silico Docking of Rhodanine derivatives and 3D-QSAR Study to Identify Potent Prostate Cancer Inhibitors

The 3VHE protein is considered as a potential target for the treatment of prostate cancer. In order to find new 3VHE inhibitors, pharmacophore models based on the molecular structure of rhodanine derivatives and a three-dimensional quantitative structure-activity relationship model (3D-QSAR) have been developed and validated by different methods. The 3D-QSAR model was evaluated for its predictive performance on a diverse test set containing 18 prostate cancer inhibitors. It presents very interesting internal and external statistical validation parameters (SD = 0.081;R2 = 0.903;Q2 = 0.869;;F = 247.2). This result suggests that the 3D-QSAR combinatorial model can be used to search for new 3VHE inhibitors and predict their potential activity. Based on the combinatorial pharmacophore model, a virtual screening of the Enamine database was performed. Compounds selected after virtual screening were subjected to molecular docking protocols (HTVS, SP, XP and IFD). Twenty new active compounds have been identified and their absorption, distribution, metabolism and excretion (ADME) property calculated using Schr?dinger’s Qikprop module. These results suggest that these new compounds could constitute new chemical starting points for further structural optimization of 3VHE inhibitors.

Green coal or bio-coal is coal produced with rich biodegradable materials, elaborated from agricultural and household residues with a high percentage of carbon. This green charcoal (fuel briquettes) is an alternative to charcoal. Well known for its contribution to greenhouse gas emissions, charcoal is one of the causes of tree felling. The valorization of waste by the manufacture of biofuels could be an alternative to the use of charcoal. The general objective of the present study is the valorization of nine biomasses from Togo as raw materials. Specifically, physico-chemical characteristics such as dehydration, acidity, and conductivity were determined. Information on the structure and composition of the biomass was found. These data on the nature of the biomass were found through the use of Fourier Transform Infrared (FTIR) and Thermogravimetry (TGA). The promising results inform on the nature of the analyzed samples and allow the selection of the best biomass which would give an important thermal conductivity for the manufacture of the briquettes, but also of the binders to be used according to the physico-chemical characteristics like the pH.

Valorization of Griffonia simplicifolia Seed Oil for Biodiesel Production: A Sustainable Alternative

Bio-derived oxygenated hydrocarbons, such as mixtures of fatty acid methyl esters (biodiesel), are promising alternatives for alleviating the adverse effects of fossil fuel consumption on climate change and preventing petroleum resource depletion. However, the selection of a viable feedstock for competitive biodiesel production remains challenging. Recent studies focusing on Griffonia simplicifolia seeds, the sole plant industrially exploited for 5-hydroxy- tryptophan (5-HTP) extraction, have shown that G. simplicifolia seed oil (GSO) can be solvent-extracted directly from ground seeds or the remaining seed cakes obtained after 5-HTP extraction with quantitative yields. This work documents the conversion of GSO into biodiesel through homogeneous base-catalyzed transesterification. The refractive index and density of the obtained methyl ester mixtures decreased with increasing oil-to-methanol molar ratio, reaction temperature, and time. Under specific conditions, 1.43 wt% FFA oil, 63.5C, 60 min, and 1:9 oil/MeOH molar ratio with 1.2 wt% NaOH or 1.3 wt% KOH as catalysts, optimal reaction conditions were reached. There were no significant differences in the potential for diminution of the refractive index and density between the NaOH and KOH catalysts. The predicted fuel properties based on the fatty acid composition determined by GC-MS showed that G. simplicifolia biodiesel exhibited a cetane index of 50.29, volumetric energy density of 34.97 MJ/L, cloud point of -1.03°C, kinematic viscosity of 4.07 mm2/s, and oxidative stability of 0.65 h. Apart from its unfavorable oxidative stability and slightly lower energy density compared to petrodiesel, all other calculated parameters met the current standards. The valorization concept proposed in this study should be integrated into the 5-HTP extraction process, preferably using the remaining dry seed cakes as raw materials to maximize revenue in a bioeconomic and sustainable approach.

Structures, Lipophilicity, Dipole Moments, Acidity and Spectroscopic Properties of Non-Steroidal Anti-Inflammatory Drugs Diclofenac, Bromfenac and Amfenac: A Theoretical Study

This work is a contribution of theoretical chemistry to the classification of some non-steroidal anti-inflammatory drugs (NSAIDs). Indeed, research on the efficacy of NSAIDs has shown that no NSAID is recognized as the most efficient anti-inflammatory drug. We have made a theoretical study of diclofenac, bromfenac and amfenac, in order to compare their efficacy from some physicochemical properties. To do this, we used the DFT and TD-DTF methods at the B3LYP/6-311+G(d, p) level theory. The lipophilicity study shows that diclofenac and bromfenac are very lipophilic. Acidity study shows that diclofenac is more acid than bromfenac and amfenac. The results from molecular orbital and the TD-DFT calculations reveal that for the three NSAIDs, the lowest energy transition is due to the excitation from HOMO to LUMO. The absorption energy corresponding to H→L transition is comparable with the energy gap value. Our findings have shown that bromfenac is more reactive than amfenac, which is more reactive than diclofenac.

Hypoglycaemic effect of Berberis vulgaris *** normal and streptozotocin-induced diabetic rats

Objective:To achieve a primary pharmacological screening contained in the aqueous extract of Berberis vulgaris(***) and to examine the hypoglycaemic effect and biochemical parameters of aqueous and saponins extract on groups of rats rendered diabetic by injection of ***:The phytochemicol tests to detect the presence of different compounds were based on the visual observation of color change or formation of precipitate after the addition of specific *** was induced in rats by intraperitoneal(i.p.) injection of streptozotocin(STZ) at a dose of 65 mg/kg *** fasting blood glucose levels were estimated by glucose oxidase-peroxidase reactive strips(dextrostix,Bayer Diagnostics).Blood samples were taken by cutting the tip of the *** cholesterol and serum triglycerides were estimated by enzymatic DHBS colorimetric ***:Administration of 62.5 and 2S.0 mg/kg of saponins and aqueous extract respectively in normal rats group shows a significant hypoglycemic activity(32.33%and 40.17%respectively) during the first ***,diabetic group treated with saponin extract produced a maximum fall of 73.1%and 76.03%at day 1 and day 21 compared to the diabetics ***,blood glucose levels of the diabetic rats treated with aqueous extract showed decrease of 78.79%on the first day and the effect remains roughly constant during 3 week. Both extracts also declined significantly biochemical parameters(20.77%-49.00%).The control in the loss of body weight was observed in treated diabetic rats as compared to diabetic controls. Conclusions:These results demonstrated significant antidiabetic effects and showed that serum cholesterol and serum triglycerides levels were decreased,significantly,consequently this plant might be of value in diabetes treatment.

The use of Chenopodium ambrosioides in some communities in Togo reveals its importance and its medicinal properties. The plant is often infused for medicinal drinks before use. Thus, through a comparative study, this work focused on the identification of the different phytochemical groups and then on the anti-radical capacities of ethanolic and aqueous extracts of aerial parts (lea- ves and seeds) of Chenopodium ambrosiooides obtained by maceration and infusion. The phytochemical tests, carried out on the various extracts, showed that they contained certain biomolecules such as flavonoids and tannins. Likewise, the presence of terpenoids and terpenes has been detected. However, it is only in aqueous extracts that the alkaloids have been revealed.
