The evolution of properties of crosslinked polyethylene(XLPE)-based insulating mate-rials under radio-chemical ageing conditions is presented and discussed.In total,seven different polymer compounds were tested,characterised by an increasing quantity of additives,for example,antioxidants(AOs)and fillers as flame retardants.Samples were analysed using different testing techniques,that is,Fourier Transform Infra-Red and dielectric spectroscopy.This allowed a broad characterisation of the polymer,bringing to good correlations between the arising of new chemical groups caused by ageing and the modifications on the complex permittivity.Phenol-based AOs are found to be the most efficient in terms of protection against oxidation.On the contrary,flame retardants are concluded not to have any impact on the material ageing.Acquired data were then treated using the principal component analysis.The data treatment successfully distinguished the material composition(with and without additives)and the ageing environments.The results confirm the relevance of this analysis in understanding the impact of irradiation on the chemical and functional properties of the polymer materials studied.

Dendrites are specialized neuronal compartments that sense, integrate and transfer information in the neural network. Their development is tightly controlled and abnormal dendrite morphogenesis is strongly linked to neurological disorders. While dendritic morphology ranges from relatively simple to extremely complex for a specified neuron, either requires a functional secretory pathway to continually replenish proteins and lipids to meet dendritic growth demands. The Golgi apparatus occupies the center of the secretory pathway and is regulating posttranslational modifications, sorting, transport, and signal transduction, as well as acting as a non-centrosomal microtubule organization center. The neuronal Golgi apparatus shares common features with Golgi in other eukaryotic cell types but also forms distinct structures known as Golgi outposts that specifically localize in dendrites. However, the organization and function of Golgi in dendrite development and its impact on neurological disorders is just emerging and so far lacks a systematic summary. We describe the organization of the Golgi apparatus in neurons, review the current understanding of Golgi function in dendritic morphogenesis, and discuss the current challenges and future directions.

Recent invasions by non-native gobiid fish species that are ongoing in the Western European rivers Rhine and Meuse, will lead to interactions with native benthic fish species. Since both non-native gobiids and native benthic species are bottom dwelling species with a preference for shelter during at least part of their life cycle, it is likely that competition for shelter will occur between these non-native and native species when shelter is a limiting factor. To investigate the importance of this mechanism for species replacements, various habitat choice experiments were conducted between two common native benthic fish species (Cottus perifretumandBarbatula barbatula) and four invasive non-native gobiid species (Proterorhinus semilunaris,Neogobius melanostomus,N. kessleriandN. fluviatilis). The first series of single specimen experiments determined the habitat choice of each individual fish species. In a second series of competition experiments, shifts in habitat choice in comparison with the previously observed habitat choice, were determined when a native benthic fish species co-occurred with non-native gobiid species. NativeC. perifretumdisplayed a significant shift in habitat choice in co-occurrence with the gobiidsN. kessleriorP. semilunaris. C. perifretumwas outcompeted and moved from the available shelter place to less preferred habitat types. During the competition experiments no change in habitat choice ofB. barbatulawas shown. Our study therefore suggests that competition for shelter is likely to occur in rivers invaded byN. kessleriandP. semilunarisat sites where shelter is limiting.

＜正＞We recently identified the 1,2,4-triazole derivative SecinH3 as the first small-molecule inhibitor of a class of cytoplasmic regulatory proteins called cytohesins by using an aptamer-displacement assay.[1]SecinH3 targets the Sec7 domain of cytohesins 1,2,and 3,and inhibits their guanine-nucleotide-exchange activity.Its application in human liver cells,flies,and mice led to impaired insulin signaling in each case.Thus,cytohesins associated with the insulin-receptor complex appear to be essential components of this central cellular signaling pathway.[2]

Genetic variation is a key factor influencing cytokine production capacity,but which genetic loci regulate cytokine production before and after vaccination,particularly in African population is unknown.Here,we aimed to identify single-nucleotide polymorphisms(SNPs)controlling cytokine responses after microbial stimulation in infants of West-African ancestry,comprising of low-birth-weight neonates randomized to bacillus Calmette-Gue rin(BCG)vaccine-at-birth or to the usual delayed BCG.Genome-wide cytokine cytokine quantitative trait loci(cQTL)mapping revealed 12 independent loci,of which the LINC01082-LINC00917 locus influenced more than half of the cytokine-stimulation pairs assessed.Furthermore,nine distinct cQTLs were found among infants randomized to BCG.Functional validation confirmed that several complement genes affect cytokine response after BCG vaccination.We observed a limited overlap of common cQTLs between the West-African infants and cohorts of Western European individuals.These data reveal strong population-specific genetic effects on cytokine production and may indicate new opportunities for therapeutic intervention and vaccine development in African populations.

Background: Psychosocial factors attract interest in investigating the occurrence of cardiovascular disease. Design This study aimed to examine the impact of critical life events on the development of myocardial infarction in smokers and ex-smokers. We hypothesized that critical life events increase the risk of the disease. Methods: Data were taken from the Cologne Smoking Study (CoSmoS), a retrospective multicentre case-control study that examines which psychosocial factors may lead to a higher risk for smokers and ex-smokers of suffering from a myocardial infarction. Our sample consisted of n = 278 myocardial infarction participants and control participants. Both groups had a history of smoking. Logistic regression was used in the analysis. Results: The study results of the smoking and ex-smoking participants showed that sociodemographic data like gender and age have an effect on the development of myocardial infarction. Physical activity seems to offer protection aganist myocardial infarction. Final, the unexpected result that the experience of at least one critical life event seems to have a positive effect on health and so lowers the risk of myocardial infarction. Conclusions: Socio-demographic data and physical activity have an effect on the development of myocardial infarction. Participants with experience of critical life events appear to be strengthened after the events and possess adequate resources to protect their health.

Anorexia and fasting are host adaptations to acute infection, inducing a metabolic switch towards ketogenesis and the production of ketone bodies, including β-hydroxybutyrate (BHB) 1-6. However, whether ketogenesis metabolically influences the immune response in pulmonary infections remains unclear. Here we report impaired production of BHB in humans with SARS-CoV-2-induced but not influenza-induced acute respiratory distress syndrome (ARDS). CD4^(+) T cell function is impaired in COVID-19 and BHB promotes both survival and production of Interferon-γ from CD4^(+) T cells. Using metabolic tracing analysis, we uncovered that BHB provides an alternative carbon source to fuel oxidative phosphorylation (OXPHOS) and the production of bioenergetic amino acids and glutathione, which is important for maintaining the redox balance. T cells from patients with SARS-CoV-2-induced ARDS were exhausted and skewed towards glycolysis, but can be metabolically reprogrammed by BHB to perform OXPHOS, thereby increasing their functionality.

星形胶质细胞支持正常的脑功能,但在卒中等病理条件下,星形胶质细胞反应性激活,其也可能参与神经退行性改变。反应性星形胶质细胞增生对机体有益或有害,与环境背景有关,但其分子基础仍未得到充分解释。借助RiboTag技术,我们在特异性地短暂性大脑中动脉闭塞(tMCAO)72h后的小鼠星形胶质细胞中,纯化翻译mRNA,构建卒中特异性星形胶质细胞翻译组数据库。我们发现,与对照组脑组织相比,tMCAO后的反应性星形胶质细胞显示与A2表型相关的转录物的富集,这与神经保护相关。星形胶质细胞也会上调大量潜在的神经毒性基因。我们共鉴定了1003个基因和38个转录因子的差异表达,其中Stat3,Sp1和Spi1变化最明显。为了进一步探索Stat3介导的通路对卒中发病机制的影响,我们选择星形胶质细胞特异性条件性缺失Stat3的小鼠制造tMCAO模型,发现这些小鼠的局灶性缺血卒中体积减少,且梗死72h后小鼠的运动结果改善。综上所述,本研究扩展了新兴的星形胶质细胞特异性靶向卒中治疗数据库,并肯定了星形胶质细胞是脑功能的关键保障细胞。
