Background:Nonalcoholic fatty liver disease(NAFLD)is a global health concern with the acid sphingomyelinase(AsM)/ceramide(CE)pathway and the NOD-like receptor family,pyrin domain-containing protein 3(NLRP3)inflammasome identified as pivotal players in lipid disorders and *** study explores the interaction mechanism between the AsM/CE pathway and NLRP3 in NAFLD cell models,aiming to understand the impact of amitriptyline(Ami),an AsM inhibitor,on lipid deposition and hepatocyte injury by regulating the AsM/CE-NLRP3 ***:HepG2 and HL-7702 cells were exposed to free fatty acids(FFAs)to establish the NAFLD *** cells were divided into 5 groups:control group,model group,Ami group,tumor necrosis factoralpha(TNF-α)group,and Ami+TNF-α*** lipid droplets were visualized using Oil Red O staining,and Western blot analysis quantified AsM,NLRP3,and caspase 1 protein *** linked immunosorbent assay(ELIsA)was measured CE and AsM levels,while qRT-PCR assessed mRNA *** apoptotic rate was evaluated by flow cytometry(FCM).Results:Following FFAs incubation,significant increases in AsM and CE levels were observed in HepG2 and HL-7702 cells,accompanied by elevated expression of NLRP3,and caspase 1,and IL-1β.TNF-αtreatment further amplified these *** demonstrated a reduction in lipid deposition,suppressed AsM/CE pathway activation,downregulated NLRP3 and caspase 1 expression,and improved ***,MCC950,a selective inhibitor of the NLRP3,mitigated NLRP3,caspase 1,and IL-1βexpression,alleviating lipid deposition and apoptosis in the NAFLD cell ***:The AsM/CE-NLRP3 pathway in NAFLD cells promotes hepatocyte steatosis,inflammation,and cell *** emerges as a promising therapeutic agent by inhibiting the AsM/CE-NLRP3 pathway,underscoring its potential as a key target for NAFLD treatment.

Recently, bispecific T-cell engagers(BiTEs) and chimeric antigen receptor-modified T cells(CAR-Ts) have been shown to have high therapeutic efficacy in hematological tumors. CD87 is highly expressed in solid tumors with an oncogenic function. To assess their cytotoxic effects on invasive nonfunctioning pituitary adenomas(iNFPAs), we first examined CD87 expression and its effects on the metabolism of iNFPA cells. We generated CD87-specific BiTE and CAR/IL-12 T cells, and their cytotoxic effects on iNFPAs cells and in mouse models were determined. CD87 had high expression in i NFPA tissue and cell samples but was undetected in noncancerous brain samples. CD87×CD3 BiTE and CD87 CAR/IL-12 T-cells showed antigenic specificity and exerted satisfactory cytotoxic effects, decreasing tumor cell proliferation in vitro and reducing existing tumors in experimental mice. Overall, the above findings suggest that CD87 is a promising target for the immunotherapeutic management of iNFPAs using anti-CD87 BiTE and CD87-specific CAR/IL-12 T cells.

BACKGROUND Laparoscopic colorectal cancer surgery increases the risk of incisional hernia(IH)at the tumor extraction *** To investigate the incidence of IH at extraction sites following laparoscopic colo-rectal cancer surgery and identify the risk factors for IH *** This study retrospectively analyzed the data of 1614 patients who underwent la-paroscopic radical colorectal cancer surgery with tumor extraction through the abdominal wall at our center between January 2017 and December ***-rences in the incidence of postoperative IH at different extraction sites and the risk factors for IH incidence were *** Among the 1614 patients who underwent laparoscopic radical colorectal cancer surgery,303(18.8%),923(57.2%),171(10.6%),and 217(13.4%)tumors were ex-tracted through supraumbilical midline,infraumbilical midline,umbilical,and off-midline *** these,52 patients developed IH in the abdominal wall,with an incidence of 3.2%.The incidence of postoperative IH was significantly higher in the off-midline incision group(8.8%)than in the middle incision groups[the supraumbilical midline(2.6%),infraumbilical midline(2.2%),and umbilical incision(2.9%)groups](χ^(2)=24.985;Psis showed that IH occurrence was associated with age,obesity,sex,chronic cough,incision infection,and combined diabetes,anemia,and hypopro-teinemia(Psimilarly,multivariate analysis showed that off-midline incision,age,sex(female),obesity,incision infection,combined chronic cough,and hypoproteinemia were independent risk factors for IH at the site of laparoscopic colorectal cancer surgery(PsION The incidence of postoperative IH differs between extraction sites for laparoscopic colorectal cancer *** infraumbilical midline incision is associated with a lower hernia rate and is thus a suitable tumor extraction site.
