The involvement of proline-rich protein Mus musculus predicted gene 4736 in ocular surface functions

AIM: To research the two homologous predicted proline -rich protein genes, Mus musculus predicted gene 4736 (MP4) and proline-rich protein BstNI subfamily 1 (Prb1) which were significantly upregulated in cultured corneal organs when encountering fungal pathogen preparations. This study was to confirm the expression and potential functions of these two genes in ocular surface. METHODS: A Pseudomonas aeruginosa keratitis model was established in Balb/c mice. One day post infection, mRNA level of MP4 was measured using real-time polymerase chain reaction (PCR), and MP4 protein detected by immunohistochemistry (IHC) or Western blot using a customized polyclonal anti -MP4 antibody preparation. Lacrimal glands from normal mice were also subjected to IHC staining for MP4. An online bioinformatics program, BioGPS, was utilized to screen public data to determine other potential locations of MP4. RESULTS: One day after keratitis induction, MP4 was upregulated in the corneas at both mRNA level as measured using real -time PCR and protein levels as measured using Western blot and IHC. BioGPS analysis of public data suggested that the MP4 gene was most abundantly expressed in the lacrimal glands, and IHC revealed that normal murine lacrimal glands were positive for MP4 staining. CONCLUSION: MP4 and Prb1 are closely related with the physiology and pathological processes of the ocular surface. Considering the significance of ocular surface abnormalities like dry eye, we propose that MP4 and Prb1 contribute to homeostasis of ocular surface, and deserve more extensive functional and disease correlation studies.

Association between red blood cell distribution width and the occurrence of cervical carcinoma

Objective:To explore correlation between red blood cell distribution width(RDW)and cervical ***:50 cervical carcinoma patients and 75 healthy controls from the First Affiliated Hospital of Soochow University from June 2020 to December 2020 were involved as the patients group and the control group *** data were collected from routine blood tests and biochemistry examination,the correlation of RDW with hemoglobin(Hb)and c-reactive protein(CRP)was evaluated by Spearman rank correlation,and receiver operating characteristic(ROC)curve was drawn to analyze the diagnostic value of RDW for cervical ***:The levels of RDW,squamous cell carcinoma antigen(SCCA)and carcinoembryonic antigen(CEA)in patients with cervical carcinoma were higher than those in the *** levels of high-density lipoprotein(HDL),Albumin,Prealbumin,Hb,RBC and neuron-specific enolase(NSE)in patients were lower than those in the controls(all P0.05).The AUC of RDW in the diagnosis of cervical carcinoma was *** on Spearman's rank correlation coefficient analysis,RDW was positively correlated with Hb(r=0.434,P<0.05)and CRP(r=0.366,P<0.05)RDW,and was negatively correlated with hematocrit(r=-0.363,P<0.05),in cervical carcinoma group,cervical carcinoma subgroupⅠ-ⅡandⅢ-Ⅳwere higher than the control group(P<0.05).Conclusions:Elevated RDW is associated with the occurrence of cervical carcinoma,and it may have clinical value in the diagnosis of cervical carcinoma and the course of reaction.

To the Editor： Childhood splenectomy exposes patients to increased risk of bacterial infections. Appropriate vaccination and education have helped combat these; however, insufficient attention is paid to viral infections atter splenectomy. Herein, we report a case of cold agglutinin disease （CAD） caused by recurrent viral infections after splenectomy to treat childhood immune thrombocytopenic purpura （1TP）. Antiviral and immunostimulant therapies were administered, resulting in an improved general condition.

Hepatocellular carcinoma(HCC) is a worldwide health problem with limited treatment options and poor prognosis. Inflammation is associated with liver injury and hepatocyte regeneration, which leads to fibrosis, cirrhosis and eventually HCC. Interleukin-1α(IL-1α) is one of the most important inflammatory cytokines involved in the inflammation and tumor development. IL-1α presents as multiple forms in vivo, including precursor, propiece, membrane and secreted forms, and their functions were thought to be different. The role of membrane IL-1α in hepatocellular carcinogenesis is still not clear. Here we examined the functions of membrane IL-1α in murine HCC models. We found that membrane IL-1α potently inhibited HCC tumor growth. Further studies showed membrane IL-1α promoted T and NK cell activation in vivo. IFN-γ production by CD8+ T and NK cells was also increased by membrane IL-1α expression. Moreover, the cytotoxicity of the cytotoxic T lymphocytes(CTL) and NK cells was also enhanced by membrane IL-1α expression. Further in vitro study demonstrated that membrane IL-1α could also activate T cells and NK cells. Depletion both CD8+ T and NK cells suppressed the anti-tumor role of membrane IL-1α. Our studies demonstrated that membrane IL-1α could promote anti-tumor immune responses through activation of T and NK cells. Thus, our finding provides new insights of IL-1α functions during HCC development.
