Background: and Aims: We previously demonstrated that cancer-associated fibroblasts(CAFs) promote tumor growth via recruiting of myeloid-derived suppressor cells(MDSCs). 5-lipoxygenase(5-LO) is highly expressed in myeloid cells and critical for synthesizing leukotriene B4(LTB4), which is involved in tumor progression via activating its receptor Leukotriene B4 receptor type 2(BLT2). In this study, we investigated the potential role of CAFs in regulating MDSC function to enhance cancer stemness, the driving force of the cancer aggressiveness and chemotherapy refractoriness, in highly desmoplastic intrahepatic cholangiocarcinoma(ICC).Approach and Results: RNA-sequencing analysis revealed enriched metabolic pathways but decreased inflammatory pathways in cancer MDSCs compared with blood MDSCs from ICC patients. Co-injection of ICC patient-derived CAFs promoted cancer stemness in an orthotopic ICC model, which was blunted by MDSC depletion. Conditioned media(CM) from CAF-educated MDSCs drastically promoted tumorsphere formation efficiency and stemness marker gene expression in ICC cells. CAF-CM stimulation increased expression and activity of 5-LO in MDSCs while 5-LO inhibitor impaired the stemness-enhancing capacity of MDSCs in vitro and in vivo. Furthermore, CAFs were identified as the major source for IL-6 and IL-33, which mediated hyperactivated 5-LO metabolism in MDSCs. Furthermore, IL-6 and IL-33 mainly expressed by CAFs mediated hyperactivated 5-LO metabolism in *** identify the LTB4-BLT2 axis as the critical downstream metabolite signaling of 5-LO in promoting cancer stemness, as treatment with LTB4 that were elevated in CAF-educated MDSCs or blockade of BLT2 that was preferentially expressed in stem-like ICC cells significantly reduced stemness-enhancing effects of CAF-educated MDSCs. Screening study on new tumor marker periplakin for lung cancer

Objective: The aim of this study was to use lung cancer targeting binding polypeptide ZS-9 to screen cDNA library of human lung cancer and obtain ZS-9 specific ligand to confirm tumor marker of non small-cell lung cancer. Methods: Artificially synthesize biotin labeled peptide ZS-9, anchored ZS-9 in the enzyme label plate coupled by avidin, used ZS-9 as probe to screen cDNA library of human lung cancer, after screening, obtained bacteriophage clone specifically binding with anchored polypeptide ZS-9. Extracted plasmid of bacteriophage and performed sequencing after amplified by PCR. Results: It was demonstrated by bioinformatic analysis on the sequence of ligand binded by lung cancer specific peptide ZS-9 that the ligand was the cytoskeletal protein periplakin on the surface of lung cancer cells, suggesting that periplakin might be a new marker for non-small-cell lung cancer in lung cancer. Conclusion: Use specific lung cancer binding peptide to screen new tumor marker periplakin in lung cancer and further studies on its biologic functions in genesis and development of lung cancer are still needed.

A genetic variant in pseudogene E2F3P1 contributes to prognosis of hepatocellular carcinoma

Certain pseudogenes may regulate their protein-coding cousins by competing for miRNAs and play an active biological role in cancer. However, few studies have focused on the association of genetic variations in pseudogenes with cancer prognosis. We selected six potentially functional single nucleotide polymorphisms （SNPs） in cancerrelated pseudogenes, and performed a case-only study to assess the association between those SNPs and the prognosis of hepatocellular carcinoma （HCC） in 331 HBV-positive HCC patients without surgical treatment. Log-rank test and Cox proportional hazard models were used for survival analysis. We found that the A allele of rs9909601 in E2F3P1 was significantly associated with a better prognosis compared with the G allele [adjusted hazard ratio （HR） = 0.69, 95% confidence interval （CI） = 0.56-0.86, P = 0.001]. Additionally, this protective effect was more predominant for patients without chemotherapy and transcatheter hepatic arterial chemoembolization （TACE） treatment. Interestingly, we also detected a statistically significant multiplicative interaction between genotypes of rs9909601 and chemotherapy or TACE status on HCC survival （P for multiplicative interaction 〈 0.001）. These findings indicate that rs9909601 in the pseudogene E2F3P1 may be a genetic marker for HCC prognosis in Chinese.

With the annual increases in the morbidity and mortality rates of tumors,the use of biomarkers for early diagnosis and real-time monitoring of tumor cells is of great *** used for tumor cell detection in body fluids include circulating tumor cells,nucleic acids,protein markers,and extracellular *** them,circulating tumor cells,circulating tumor DNA,and exosomes have high potential for the prediction,diagnosis,and prognosis of tumor diseases due to the large amount of valuable information on tumor characteristics and evolution;in addition,in situ monitoring of telomerase and miRNA in living cells has been the topic of extensive research to understand tumor development in real *** techniques,such as enzyme-linked immunosorbent assays,immunoblotting,and mass spectrometry,have been widely used for the detection of these *** them,the detection of tumor cell markers in body fluids based on electrochemical biosensors and fluorescence signal analysis is highly preferred because of its high sensitivity,rapid detection and portable ***,we summarize recent research progress in the detection of tumor cell biomarkers in body fluids using electrochemical and fluorescence biosensors,outline the current research status of in situ fluorescence monitoring and the analysis of tumor markers in living cells,and discuss the technical challenges for their practical clinical application to provide a reference for the development of new tumor marker detection methods.

Expression quantitative trait loci for PVT1contributes to the prognosis of hepatocellular carcinoma

Aim: Plasmacytoma variant translocation 1 (PVT1), a long intergenic non-coding RNA, was overexpressed in liver cancer. A single nucleotide polymorphism (SNP) rs4733586 was identified as an expression quantitative trait loci (eQTL) for PVT1 using bioinformatics analysis. This study was to assess the association of PVT1 eQTL with hepatocellular carcinoma (HCC) prognosis. Methods: A case-only study was performed to assess the association between SNP and HCC overall survival in 331 HCC patients with hepatitis B virus. Cox proportional hazard regression models were conducted for survival analysis with adjustment for age, gender, smoking status, drinking status, Barcelona-Clinic Liver Cancer stages, and chemotherapy or transcatheter hepatic arterial chemoembolization (TACE) status. Results: The variant genotype C allele of rs4733586 was significantly associated with a higher death risk compared with T allele (adjusted hazard ratio = 1.26, 95% confidence intervals = 1.05-1.51,P = 0.012 in the additive model). By stepwise Cox proportional hazard analysis, four variables (age, drinking status, chemotherapy or TACE status, PVT1 eQTL) were remained in the final regression model. In the stratified analysis, no heterogeneity was observed among different subgroups. Conclusion: These findings suggest that eQTL SNP for PVT1 may be susceptibility marker for the HCC overall survival.

Prognostic significance of X-ray cross-complementing gene 1 expression in gastric cancer

Objective： The aim of this study is to identify the prognostic significance of X-ray cross-complementing gene 1 （XRCCI） in patients with gastric cancer undergoing surgery and platinum-based adjuvant chemotherapy. Methods： Immunohistochemistry （IHC） was used to evaluate XRCCI protein expression profiles on surgical specimens of 612 gastric cancer patients. The relationship between XRCC1 expression and existing prognostic factors, platinum-based adjuvant chemotherapy, disease-free survival （DFS） and overall survival （OS） were analyzed. Results： Among 612 patients staged II/III in our study, 182 （29.74%） were evaluated as XRCC1 IHC positive. XRCC1 expression was not significantly related to OS （P=0.347） or DFS （P=0.297）. Compared with surgery only, platinum-based adjuvant chemotherapy significantly improved the OS （P=0.031）. And the patients with negative XRCC1 expression benefited more from platinum-based adjuvant chemotherapy （P=0.049）. Multivariate analysis demonstrated that tumor size, T category, N category, vascular or nerve invasion and platinum-based chemotherapy were good prognostic factors for OS （P〈0.05）. Though XRCCI plays an important role in DNA repair pathways, no significant relationship is found in XRCCI expression and OS among gastric cancer in our study. Conclusions： XRCC1 might be an alternative prognostic marker for the patients of gastric cancer after radical resection. The patients with negative XRCC1 expression can benefit more from platinum-based adjuvant chemotherapy.
