Mastitis affects almost all mammals including humans and dairy cows. In the dairy industry, bovine mastitis is a disease with a persistently high incidence, causing serious losses to the health of cows, the quality of dairy products, and the economy of dairy farms. Although local udder infection caused by the invasion of exogenous pathogens into the mammary gland was considered the main cause of mastitis,evidence has been established and continues to grow, showing that nutrition factors and gastrointestinal microbiome(GM) as well as their metabolites are also involved in the development of mammary inflammatory response. Suboptimal nutrition is recognized as a risk factor for increased susceptibility to mastitis in cattle, in particular the negative energy balance. The majority of data regarding nutrition and bovine mastitis involves micronutrients. In addition, the dysbiotic GM can directly trigger or aggravate mastitis through entero-mammary gland pathway. The decreased beneficial commensal bacteria, lowered bacterial diversity, and increased pathogens as well as proinflammatory metabolites are found in both the milk and gastrointestinal tract of mastitic dairy cows. This review discussed the relationship between the nutrition(energy and micronutrient levels) and mastitis, summarized the role of GM and metabolites in regulating mastitis. Meanwhile, several non-antibiotics strategies were provided for the prevention and alleviation of mastitis, including micronutrients, probiotics, short-chain fatty acids, highfiber diet, inulin, and aryl hydrocarbon receptor.

Sulforaphane ameliorates non-alcoholic steatohepatitis by KLF4-mediated macrophage M2 polarization

Non-alcoholic fatty liver disease (NAFLD) has become a global issue and a severe threat to public ***, to date, no approved therapeutic drugs have been developed. Dietary interventions with naturalproducts have shown promise in preventing and treating NAFLD. Sulforaphane (SFN) is a phytocompoundwith antioxidant and anti-inflammatory properties, and previous research has demonstrated that SFN canameliorate hepatic lipid accumulation and inflammation. However, the molecular mechanisms underlying thesebeneficial effects remain unclear. In this study, we confirmed the protective effects of SFN on excessive lipidaccumulation and inflammatory injury in a high-fat, high-fructose diet-induced non-alcoholic steatohepatitis(NASH) mouse model. We found that SFN attenuates the inflammatory injury in a macrophage cell line andthe liver of NASH mice, owing to the promotion of M1-type macrophage polarization toward the M2-type andthe regulation of inflammatory mediators. Further analysis demonstrated that this SFN-induced macrophageM2-type polarization occurs in a Krüppel-like factor 4 (KLF4)-dependent manner. In summary, we uncovereda new mechanism of action underlying SFN activity and provide evidence that dietary intervention with SFNmight be protective against NASH.

Recent advances in probiotics oral delivery systems targeting the inflammatory bowel disease: Types， mechanisms and perspectives

Inflammatory bowel disease (IBD) influences several million people around the globe， with a high prevalence in North America and Europe. Results from the studies about host-gut microbial interactions demonstrated that gut microbiota plays a critical role in the progression of IBD， and probiotics can significantly improve microflora dysbiosis and inflammatory response caused by intestinal pathogens. However， several limitations existed for the probiotics delivered to the intestine in the free form (non-encapsulated)， such as low pH and diverse digestive enzymes in the gastrointestinal tract， etc. To overcome the problems， several probiotic delivery systems were established and verified with effects. Here， the types and applications of probiotics in animal models and clinical studies are firstly reviewed in this paper. Subsequently， various types of probiotic delivery systems are elaborated， containing the well-known microcapsules and hydrogel delivery systems， and the engineered probiotic delivery systems are also introduced. Furthermore， mechanism of action associated with probiotics are illustrated， including maintaining gut microbiota barrier balance， modulating the immune response， and alleviating oxidative stress， etc. Finally， we discussed the relative advantages and disadvantages of different encapsulation methods， as well as future trends for further development of probiotic delivery systems with health benefits.

Parvalbumin-positive retinal ganglion cells(PV+RGCs)are an essential subset of RGCs found in various ***,their role in transmitting visual information remains ***,we characterized PV+RGCs in the retina and explored the functions of the PV+RGC-mediated visual *** applying multiple viral tracing strategies,we investigated the downstream of PV+RGCs across the whole ***,we found that the PV+RGCs provided direct monosynaptic input to PV+excitatory neurons in the superficial layers of the superior colliculus(SC).Ablation or suppression of SC-projecting PV+RGCs abolished or severely impaired the flight response to looming visual stimuli in mice without affecting visual ***,using transcriptome expression profiling of individual cells and immunofluorescence colocalization for RGCs,we found that PV+RGCs are predominant glutamatergic ***,our findings indicate the critical role of PV+RGCs in an innate defensive response and suggest a non-canonical subcortical visual pathway from excitatory PV+RGCs to PV+SC neurons that regulates looming visual *** results provide a potential target for intervening and treating diseases related to this circuit,such as schizophrenia and autism.

Repeated inoculation with rumen fluid accelerates the rumen bacterial transition with no benefit on production performance in postpartum Holstein dairy cows

Background The dairy cow’s postpartum period is characterized by dramatic physiological changes,therefore imposing severe challenges on the animal for maintaining health and milk *** dynamics of the ruminal microbiota are also tremendous and may play a crucial role in lactation *** aim to investigate the potential benefits of early microbial intervention by fresh rumen microbiota transplantation(RMT)and sterile RMT in postpartum dairy *** fistulated peak-lactation dairy cows were selected to be the donors for rumen fluid *** postpartum cows were divided into 3 groups as the transplantation receptors respectively receiving 10 L fresh rumen fluid(FR),10 L sterile rumen fluid(SR),or 10 L saline(CON)during 3 d after *** Production performance,plasma indices,plasma lipidome,ruminal microbiome,and liver transcriptome were *** fresh and sterile RMT,we found that the molar proportion of propionic acid was increased on d 7 in the FR and SR groups and the bacterial composition was also significantly changed when compared with the CON group.A similarity analysis showed that the similarities between the CON group and FR or SR group on d 7 were 48.40%or 47.85%,whereas the similarities between microbiota on d 7 and 21 in the FR and SR groups were 68.34%or 66.85%.Dry matter intake and feed efficiency were not affected by ***β-hydroxybutyrate concentration in the FR group was decreased and significantly different lipids mainly included phosphatidylcholine and lysophosphatidylcholine containing polyunsaturated fatty *** transcriptomics analysis indicated acutephase response pathways were upregulated in the SR *** Our study suggests that RMT can shorten the transition process of the ruminal microbiota of postpartum dairy cows with no benefit on dry matter intake or feed *** with rumen fluid may not be a useful approach to promote the recovery of postpartum dairy cows.

Effects of oat β-glucan–based prebiotic blended formulas on defecation and gut microbiota in Chinese adults: a randomized clinical trial

Digestive health is important for overall well-being， but gut health problems are common in the Chinese population. The objective of this study was to investigate the effects of oat β-glucan–based prebiotic blended formulas in humans on digestive health by assessing defecation frequen-cy， defecation symptoms， stool quality， self-reported digestive feelings， defecation satisfaction， and gut bacteria. A parallel， randomized， double-blind， controlled trial was conducted in 168 eli-gible adults randomized to three groups for 14 days: Intervention Groups A and B received two different doses of oat β-glucan–based prebiotic blended formulas and a Control Group received oat β-glucan mixed with hydroxypropylmethyl-cellulose and microcrystalline cellulose. Partici-pants completed a defecation diary and Bristol stool chart daily and digestive system question-naires weekly. Fecal samples were collected on Day 0 and Day 14 to evaluate gut bacteria using 16S rDNA analysis. Both formulas improved defecation frequency， stool quality， defecation symptoms， defecation satisfaction， and on digestive health of human， compared with baseline (P health in generally healthy Chinese adults. These formulas are a feasible nutritional strategy for digestive health.

Neonicotinoids(NEOs),a large class of organic compounds,are a type of commonly used pesticide for crop *** uptake and accumulation in plants are prerequisites for their intra-and intercellular move-ments,transformation,and *** the molecular mechanisms that underpin NEO uptake by plants is crucial for effective application,which remains ***,we demonstrate that NEOs enter plant cells primarily through the transmembrane symplastic pathway and accumulate mainly in the *** plasma membrane intrinsic proteins discovered in Brassica rapa,BraPIP1;1 and BraPIP2;1,were found to encode aquaporins(AQPs)that are highly permeable to NEOs in different plant species and facilitate NEO subcellular diffusion and *** conserved transport function was further demonstrated in Xenopus laevis oocyte and yeast ***1;1 and BraPIP2;1 gene knockouts and interaction as-says suggested that their proteins can form functional *** of the potential of mean force indicated a negative correlation between NEO uptake and the energy barrier of BraPIP1;1 *** study shows that AQPs transport organic compounds with greater osmolarity than previously thought,providing new insight into the molecular mechanisms of organic compound uptake and facilitating innovations in systemic pesticides.

Nitrogen dioxide(NO_(2))is a prevalent air pollutant in urban areas,originating from outdoor sources,household gas consumption,and secondhand *** limited evaluation of the disease burden attributable to NO_(2),encompassing different health effects and contributions from various sources,impedes our understanding from a public health *** on modeled NO2 exposure concentrations,their exposure–response relationships with lung cancer,chronic obstructive pulmonary disease,and diabetes mellitus,and baseline disability-adjusted life years(DALYs),we estimated that 1,675(655–2,624)thousand DALYs were attributable to NO2 in urban China in 2019[138(54–216)billion Chinese yuan(CNY)economic losses].The transition from gas to electricity for household cooking was estimated to reduce the attributable economic losses by 35%.This reduction falls within the range of reductions achieved when outdoor air meets the World health Organization interim target 3 and air quality guidelines for annual NO_(2),highlighting the significance of raising awareness of gas as a polluting household energy for *** findings align with global sustainable development initiatives,providing a sustainable solution to promote public health while potentially mitigating climate change.

Standardized ileal digestibility of amino acids in soybean meal fed to non-pregnant and pregnant sows

Background Two studies were designed to determine standard ileal crude protein(CP)and amino acid(AA)digestibility of soybean meal(SBM)from different origins fed to non-pregnant and pregnant *** solvent-extracted SBMs from soybeans produced in the USA,Brazil,and China were *** Exp.1,eight different diets were created:a nitrogen(N)-free diet and 7 experimental diets containing SBM from different origins as the only N *** non-pregnant,multiparous sows were arranged in an 8×8 Latin square design(8 periods and 8 diets).In Exp.2,the diet formula was the same as in *** gestating sows(parity 3)were assigned to 4 different diets in a replicated 4×3 Youden square design(three periods and four diets)in mid-gestation and again in late-gestation *** When fed to non-pregnant and late-gestating sows,the standardized ileal digestibility(SID)of CP and most AAs from different SBM were not significantly different(P>0.05).When fed to mid-gestating sows,the SID values for Arg,His,Lys,Phe,Cys,Gly,Ser,and Tyr in SBM 1 were lower than in SBM 4 and 5(P<0.05),whereas SID for Leu from SBM 5 was higher than in SBM 1 and 4(P<0.05).SID values for Ile,Ala,and Asp from SBM 4 were lower than in SBM 1 and 5(P<0.05).Sows had significantly greater SID values for Lys,Ala,and Asp during mid-gestation when compared with late-gestation stages(P<0.05).Mid-gestating sows had greater SID value for Val and lower SID value for Tyr when compared with non-pregnant and late-gestating sows(P<0.01),whereas non-pregnant sows had significantly greater SID value for Met when compared with gestating sows(P<0.01).Conclusions When fed to mid-gestating sows,the SID values for most AAs varied among SBM *** SID values for Lys,Met,Val,Ala,Asp,and Tyr in SBM were affected by sow gestation *** findings provide a cornerstone for accurate SBM use in sow diets.

Discovery of active compounds and key targets of Thymus quinquecostatus Celak. based on gastrointestinal metabolism and Gut flora-Compound-Target-Pathway network with TOPSIS method

Thymus quinquecostatus Celak.， a traditional aromatic edible plant from Lamiaceae， is widely used as food additive， food condiment， spice， and herbal teas. Its polyphenol-rich fraction （PRF） has been proven to be effective protective effect for cerebral ischemia reperfusion injury （CIRI） in our previous study. In this study， we developed a novel “Gut flora-Compound-Target-Pathway” （GCTP） network based on network pharmacology coupled with gastrointestinal metabolism for screening bio-active components， key targets and gut floras through the classical technique for order preference by similarity to ideal solution （TOPSIS） method. This compensates for the lack of gut floras and gastrointestinal metabolism in network pharmacology. Firstly， four incubation models covering simulated gastric juice， simulated intestinal juice， gut floras of normal and transient middle cerebral artery occlusion （tMCAO） rat in vitro were applied to PRF. The 109 proto-components and 64 metabolites were elucidated by UPLC-QE-Orbitrap-MSn. Then， the key targets of MMP9， PTGS2， FYN， ESR1， APP， and EGFR， and gut floras of Enterococcus avium LY1 were selected. Moreover， the selected key proto-components were rosmarinic acid， daidzein， quercetin， luteolin， apigenin， methyl rosmarinate， kaempferol， luteoloside， and caffeic acid， and the key metabolites were isokaempferide， isorhamnetin， isoquercetin， and mangiferin. Binding of compounds to the key proteins was analyzed by molecular docking， and also verified though a AAPH induced oxidative stress zebrafish model and qRT-PCR assays. This study provides a new idea and a better understanding of PRF for its protective effects on CIRI and its underlying mechanisms.
