Age-dependent genetic architectures of chicken body weight explored by multidimensional GWAS and molQTL analyses

Chicken body weight(BW) is a critical trait in breeding. Although genetic variants associated with BW have been investigated by genome-wide association studies(GWAS), the contributions of causal variants and their molecular mechanisms remain largely unclear in chickens. In this study, we construct a comprehensive genetic atlas of chicken BW by integrative analysis of 30 age points and 5 quantitative trait loci(QTL) across 27 tissues. We find that chicken growth is a cumulative non-linear process, which can be divided into three distinct stages. Our GWAS analysis reveals that BW-related genetic variations show ordered patterns in these three stages. Genetic variations in chromosome 1 may regulate the overall growth process, likely by modulating the hypothalamus-specific expression of SLC25A30 and retina-specific expression of NEK3. Moreover, genetic variations in chromosome 4 and chromosome 27 may play dominant roles in regulating BW during Stage 2(8-22 weeks) and Stage 3(23-72 weeks), respectively. In summary, our study presents a comprehensive genetic atlas regulating developmental stage-specific changes in chicken BW, thus providing important resources for genomic selection in breeding programs.

Genetic Analysis and Mapping of an Enclosed Panicle Mutant Locus esp1 in Rice (Oryza sativa L.)

A mutant was isolated from the M2 of 60Co-T ray mutagenized male-fertility restorer line Zao-R974 in rice. The mutant showed pleiotropic phenotypes including dwarfism, delayed heading time, short and partially enclosed panicles, short uppermost internode, decreased grain and secondary branch numbers per panicle, and partially degenerated spikelets. The mutant was named as espl （enclosed shorter panicle 1）. Genetic analysis indicated that the mutant phenotype was controlled by a recessive locus. Spraying exogenous GA~ did not rescue the panicle enclosure. Using an F2 and a BC, population of the cross between espl and a japonica cultivar Nipponbare, we mapped the ESP1 locus to a region of-260 kb on chromosome 11. This result provides a basis for further map-based cloning of the ESP1 locus.

Effects of dietary tert-butylhydroquinone on domoic acid metabolism and transcription of detoxification-related liver genes in red sea bream Pagrus major

Domoic acid (DA) is a neuroexcitatory amino acid that is produced by Pseudo-nitzschia during harmful algal blooms (HAB). Accumulation of DA can be transferred through food chain and cause neuronal damage in marine animal and in human. Like other algal toxins, DA was suggested to increase the oxidative stress and increase the detoxification-related gene expression in fish. The widely used food antioxidant, tert-butylhydroquinone (tBHQ), was known to induce a wide range of antioxidative potentials such as elevation of the glutathione levels and glutathione S-transferases (GSTs), via the activation of antioxidant response elements (AREs). In this study, the influences of dietary tBHQ on domoic acid (DA) metabolism and detoxification-related gene transcription were investigated both in vivo and in vitro. Oral administration of tBHQ resulted in significant decreases of DA accumulation of liver tissues in which red sea bream were fed with a single dose of 10 mg DA and 100 mg tBHQ per kg body weight per fish. Real-time PCR further revealed that the mRNA levels of AHR/ARNT/CYP1A1/GSTA1/GSTR were up-regulated in the above liver tissues at 72 h post tBHQ treatment. In consistence, tBHQ exposure also resulted in increased mRNA transcription of GSTA1, GSTA2 and GSTR in cultured red sea bream hepatocytes. Collectively, our findings in this research suggested that the dietary intake of tBHQ accelerated DA metabolism in fish, through mechanisms involving altered transcription of detoxification- related liver genes.

Mapping QTL affecting the vertical distribution and seed set of soybean [Glycine max(L.) Merr.] pods

Effects of Ca addition on microstructure and properties of AZ63 magnesium alloy

Effects of Ca addition on the microstructure and viscosity as well as electrochemical properties of casting AZ63 magnesium alloy were studied. Testing results show that the viscosity value increases with the increase of calcium content, especially at the higher temperature, and there exists such a relation between the ignition temperature and viscosity of magnesium alloy: when the melt viscosity increases, the ignition temperature increases too. The microstructure of AZ63 magnesium alloy is first refined and then coarsened with the increase of Ca addition. By the addition of 0.15% Ca, AZ63 alloy has more negative open-circuit potential(1.624 5V), bigger anode efficiency (55.65%) and lower corrosion rate(0.214 g/(m^2·h)). AZ63 alloy containing some Ca content is a high driving-potential and high-efficiency sacrificial anodes material.

The phylogenetic status of typical Chinese native pigs: analyzed by Asian and European pig mitochondrial genome sequences

China is one of the most diverse countries, which have developed 88 indigenous pig breeds. Several studies showed that pigs were independently domesticated in multiple regions of the world. The purpose of this study was to investigate the origin and evolution of Chinese pigs using complete mitochondrial genomic sequences (mtDNA) from Asian and European domestic pigs and wild boars. Thirty primer pairs were designed to determine the mtDNA sequences of Xiang pig, Large White, Lantang, Jinhua and Pietrain. The phylogenetic status of Chinese native pigs was investigated by comparing the mtDNA sequences of complete coding regions and D-loop regions respectively amongst Asian breeds, European breeds and wild boars. The analyzed results by two cluster methods contributed to the same conclusion that all pigs were classified into two major groups, European clade and Asian clade. It revealed that Chinese pigs were only recently diverged from each other and distinctly different from European pigs. Berkshire was clustered with Asian pigs and Chinese pigs were involved in the development of Berkshire breeding. The Malaysian wild boar had distant genetic relationship with European and Asian pigs. Jinhua and Lanyu pigs had more nucleotide diversity with Chinese pigs although they all belonged to the Asian major clade. Chinese domestic pigs were clustered with wild boars in Yangtze River region and South China.

Identification and Characterization of the Populus AREB/ABF Subfamily

Abscisic acid （ABA） is a major plant hormone that plays an important role in responses to abiotic stresses. The ABA-responsive element binding protein/ABRE-binding factor （AREB/ABF） gene subfamily contains crucial transcription factors in the ABA-mediated signaling pathway. In this study, a total of 14 putative AREB/ABF members were identified in the Populus trichocarpa Torr. ＆ Gray. genome using five AREB/ABF amino acid sequences from Arabidopsis thaliana L. as probes. The 14 putative Populus subfamily members showed high protein similarities, especially in the basic leucine zipper （bZIP） domain region. A neighbor-joining analysis combined with gene structure data revealed homology among the 14 genes. The expression patterns of the Populus AREB/ABF subfamily suggested that the most abundant transcripts of 11 genes occurred in leaf tissues, while two genes were most transcribed in root tissues. Significantly, eight Populus AREB/ABF gene members were upregulated after treatment with 100 t^M exogenous ABA, while the other six members were downregulated. We identified the expression profiles of the subfamily members in Populus tissues and elucidated different response patterns of Populus AREB/ABF members to ABA stress. This study provided insight into the roles of Populus AREB/ABF homologues in plant response to abiotic stresses.
