Transcriptome Analysis of Heterosis in Survival in the Hybrid Progenies of‘Haida No.1’and Orange-Shelled Lines of the Pacific Oyster Crassostrea gigas

Heterosis has been exploited to enhance the yield and adaptability in various shellfish species;however,the molecular basis of it remains unclear.The Pacific oyster Crassostrea gigas is one of the most economically important aquaculture species,and its productive traits can be improved by hybridization.Here,an intraspecific cross between orange shell(O,10th generation)and‘Haida No.1’(H,13th generation)of C.gigas was performed to assess the heterosis of survival trait.Survival rates of hybrid family(OH)and inbred families(HH and OO)were compared at larval stage,and eyed-pediveliger larvae of three families were subjected to transcriptome analysis.The analysis results of best-parent heterosis and mid-parent heterosis showed that the hybrid family exhi-bited a high heterosis in survival relative to the parental families.The OH-M(OH vs.OO)and OH-P(OH vs.HH)had 425 and 512 dif-ferentially expressed genes(DEGs),respectively.Functional enrichment analysis of these DEGs revealed that the significantly enrich-ed genes function in virion binding,C-type lectin receptor signaling pathway,cellular defense response and other immune-related pro-cesses,which involves perlucin-like protein,CD209 antigen-like protein,ZNFX1,caspase-3 and acan genes.These differentially ex-pressed genes in OH-M and OH-P,together with the immune-related processes mentioned above may play an important role in the larval survival of C.gigas.In addition,three genes(CYP450,fucolectin and perlucin-like)are associated with the orange shell and low survival of maternal oyster OO.These findings provide support for the application of hybrid with superior survival and will facilitate the understanding of heterosis formation in the Pacific oyster.

Single-cell transcriptomic landscape of peripheral blood cells provides insights into adaptation of red-eared sliders(Trachemys scripta elegans)

Red-eared sliders(Trachemys scripta elegans),as one of the 100 most threatening aliens,have stronger immunity than the native species in response to environmental stress.Blood cells are an important component of immunity in the body.However,the blood cell researches of turtle are still in the traditional blood cell classification and morphological structure observation.Furthermore,turtle granulocytes cannot be accurately identified using traditional methods.Single-cell RNA sequencing techniques have been successfully implemented to study cells based on the mRNA expression patterns of each cell.The present study profiled the transcriptomes of peripheral blood cells in red-eared sliders to construct a single-cell transcriptional landscape of the different cell types and explored environmental adaptation mechanism from the perspective of hematology.All 14 transcriptionally distinct clusters(platelets,erythrocytes1,erythrocytes2,CSF1R monocytes,POF1B monocytes,neutrophils,GATA2high basophils,GATA2low basophils,CD4 T cells,CD7 T cells,B cells,ACKR4 cells,serotriflin cells,and ficolin cells)were identified in the peripheral blood cells of the red-eared sliders.In particular,a subtype of erythrocytes(erythrocytes1)that expressed immune signals was identified.Peripheral blood cells were grouped into three lineages:platelet,erythroid/lymphoid,and myeloid cell lineages.Furthermore,based on differentiation trajectory and up-regulated gene expression,ACKR4 cells were newly identified as lymphocytes,and serotriflin and ficolin cells as granulocytes.The single-cell transcriptional atlas of the peripheral blood cells in red-eared sliders provided in the present study will offer a comprehensive transcriptome reference for the exploration of physiological and pathological hematology in this species.

Homogenous coating of Li+ conductive Li5AlO4 on single-crystalline nonstoichiometric Li1.04Ni0.92Al0.04O2 for rechargeable lithium-ion batteries

Cobalt-free,nickel-rich LiNi1-xAlxO2（x ≤ 0.1） is an attractive cathode material because of high energy density and low cost but suffers from severe structural degradation and poor rate performance.In this study,we propose a molten salt-assisted synthesis in combination with a Li-refeeding induced aluminum segregation strategy to prepare Li5AlO4-coated single-crystalline slightly Li-rich Li1.04Ni0.92Al0.04O2.The symbiotic formation of Li5AlO4from reaction between molten lithium hydroxide and doped aluminum in the bulk ensures a high lattice matching between the Ni-rich oxide and the homogenous conductive Li5AlO4that permits high Li+conductivity.Benefiting from mitigated undesirable side reactions and phase evolution,the Li5AlO4-coated single-crystalline Li1.04Ni0.92Al0.04O2delivers a high specific capacity of220.2 mA h g-1at 0.1 C and considerable rate capability(182.5 mA h g-1at 10 C).Besides,superior capacity retention of 90.8% is obtained at 1/3 C after 100 cycles in a 498.1 mA h pouch full cell.Furthermore,the particulate morphology of Li1.04Ni0.92Al0.04O2remains intact after cycling at a cutoff voltage of 4.3 V,whereas slightly Li-deficient Li0.98Ni0.97Al0.05O2features intragranular cracks and irreversible lattice distortion.The results highlight the value of molten salt-assisted synthesis and Li-refeeding induced elemental segregation strategy to upgrade Ni-based layered oxide cathode materials for advanced Li-ion batteries.

Robust, breathable, and chemical-resistant polytetrafluoroethylene (PTFE) films achieved by novel in-situ fibrillation strategy for highperformance triboelectric nanogenerators

In the era of advanced wearable electronic devices,the triboelectric nanogenerators(TENGs)as energy harvesting and self-powered sensing units hold great promise.Selecting appropriate triboelectric material is the crucial factor to optimize the performance of TENG,while polytetrafluoroethylene(PTFE)stands out as a highly versatile option among the various materials.In this study,we present an ultrafine nanofibrous PTFE(NF-PTFE)films prepared by novel in-situ fibrillation strategy as the triboelectric material in TENG devices.The innovative processing methodology facilely addresses the dilemma between high porosity and fine pore size of traditional porous PTFE films,meanwhile achieves exceptional mechanical strength,hydrophobicity,air permeability,and chemical resistance of the films.With the integration of nanofibrous PTFE films into contact-separation mode TENG and droplet-based TENG,these devices realize the peak electrical output of 131 V/10.8μA and 54 V/14μA with great durability,which surpass the performance of TENGs using traditional expanded PTFE films.Furthermore,a smart glove capable of recognizing hand gestures is proposed,which demonstrates the versatility,flexibility,and practicality of this material for potential use in smart devices.This reported NF-PTFE film provides insights for the design of high-performance TENG device for advanced wearable electrical applications.

Energy-based Directional Pilot Protection for Distribution Networks with IBDGs Considering Unmeasured Load Switching

Inverter-based distributed generations(IBDGs)are the main approach to utilizing clean energy in distribution networks(DNs).Compared with synchronous source,fault response of IBDG is very different.As a result,in DNs with high penetration of IBDGs,legacy protection schemes will no longer be applicable.In this paper,an energy-based directional pilot protection scheme suitabie for DNs with IBDGs is proposed.This scheme considers the large range of phase angle caused by IBDG integration and uses improved energy polarity criterion to determine fault direction.In addition,magnitude of energy is used to distinguish between faults and load switching to overcome maloperation of directional pilot protection caused by internal unmeasured load switching.The proposed scheme first uses local measured information to determine fault direction sign and then exchanges the direction sign with the remote terminal.This scheme does not require high-bandwidth communication and strict data synchronization,so it can be implemented at a low cost.Finally,simulation studies verify effectiveness of the proposed scheme.

Inhibiting shuttle effect of lithium polysulfides by double metal selenides for high-performance lithium-sulfur batteries

Lithium-sulfur batteries（LSBs） have attracted the attention of more and more researchers due to the advantages of high energy density,environmental friendliness,and low production cost.However,the low electronic conductivity of active material and shuttling effect of lithium polysulfides（LiPSs） limit the commercial development of LSBs.To solve these problems,we design a core-shell composite with nitrogen-doped carbon（NC） and two types of selenides（FeSe2-NC@ZnSe-NC）.The FeSe2-NC@ZnSe-NC has a strong adsorption capacity,and can effectively adsorb LiPSs.At the same time,it also effectively alleviates the shuttling effect of LiPSs,and improves the utilization of the active substance during the charge/discharge reaction processes.The mechanism involved in FeSe2-NC@ZnSe-NC is demonstrated by both experiments and density-functional theory（DFT） calculations.The electrochemical test results indicate that LSB with S/FeSe2-NC@ZnSe-NC delivers an initial discharge capacity of 1260 mAh·g-1at 0.2C.And after 500 cycles at 1C,the capacity decay rate per cycle is 0.031%,and the capacity retention rate is 85%.The FeSe2-NC@ZnSe-NC core-shell structure verifies a rational strategy to construct an electrode material for high-performance LSBs.

Identification,Characterization,and Probiotic Potentials of Lactobacillus pentosus SF-1

In recent years probiotics have been considered as a potential substitution of antibiotics to control pathogens and treat infectious diseases in aquaculture.In the present study a strain of Lactobacillus pentosus,named as L.pentosus SF-1,was isolated from waters in aquaculture.The species identification of this strain was conducted by 16S rRNA sequence,and the physiological and biochemical characteristics of this strain were assessed.Furthermore,the virulence,antibiotic sensitivity,cell surface characteristics and acid/base-resistance of L.pentosus SF-1 were determined to evaluate the probiotic potentials of this strain.Specifically,L.pentosus SF-1 is sensitive to most common antibiotics,and no hemolysin was generated from it,indicating the safety of this strain to hosts.In addition,L.pentosus SF-1 was able to tolerate the artificial gastric juice at pH 3 for 4 h and the artificial intestinal fluid at pH 6.8 or 8.0 for 6 h.Moreover,the analysis of self-aggregation and the adhesion of L.pentosus SF-1 to organic solvents suggested a high potential of L.pentosus SF-1 to inhabit the hosts,which was confirmed by testing the colonization of L.pentosus SF-1 in germ-free zebrafish.Interestingly,L.pentosus SF-1 displayed a high bactericidal activity against several bacterial pathogens.Consistently,the incubation of L.pentosus SF-1 significantly promoted the expression of antimicrobial components in zebrafish,contributing to the protection of the fish from E.tarda infection in vivo.Taken together,the probiotic strain L.pentosus SF-1 could be applied as anti-infection reagent in aquaculture.

Cryptic divergences and repeated hybridizations within the endangered “living fossil” dove tree(Davidia involucrata) revealed by whole genome resequencing

The identification and understanding of cryptic intraspecific evolutionary units(lineages) are crucial for planning effective conservation strategies aimed at preserving genetic diversity in endangered species.However, the factors driving the evolution and maintenance of these intraspecific lineages in most endangered species remain poorly understood. In this study, we conducted resequencing of 77 individuals from 22 natural populations of Davidia involucrata, a “living fossil” dove tree endemic to central and southwest China. Our analysis revealed the presence of three distinct local lineages within this endangered species, which emerged approximately 3.09 and 0.32 million years ago. These divergence events align well with the geographic and climatic oscillations that occurred across the distributional range.Additionally, we observed frequent hybridization events between the three lineages, resulting in the formation of hybrid populations in their adjacent as well as disjunct regions. These hybridizations likely arose from climate-driven population expansion and/or long-distance gene flow. Furthermore, we identified numerous environment-correlated gene variants across the total and many other genes that exhibited signals of positive evolution during the maintenance of two major local lineages. Our findings shed light on the highly dynamic evolution underlying the remarkably similar phenotype of this endangered species. Importantly, these results not only provide guidance for the development of conservation plans but also enhance our understanding of evolutionary past for this and other endangered species with similar histories.

TaSRO1 interacts with TaVP1 to modulate seed dormancy and pre-harvest sprouting resistance in wheat

Dormancy is an adaptive trait which prevents seeds from germinating under unfavorable environmental conditions.Seeds with weak dormancy undergo pre-harvest sprouting(PHS)which decreases grain yield and quality.Understanding the genetic mechanisms that regulate seed dormancy and resistance to PHS is crucial for ensuring global food security.In this study,we illustrated the function and molecular mechanism of TaSRO1 in the regulation of seed dormancy and PHS resistance by suppressing TaVP1.The tasro1 mutants exhibited strong seed dormancy and enhanced resistance to PHS,whereas the mutants of tavp1 displayed weak dormancy.Genetic evidence has shown that TaVP1 is epistatic to TaSRO1.Biochemical evidence has shown that TaSRO1 interacts with TaVP1 and represses the transcriptional activation of the PHS resistance genes TaPHS1 and TaSdr.Furthermore,TaSRO1 undermines the synergistic activation of TaVP1 and TaABI5 in PHS resistance genes.Finally,we highlight the great potential of tasro1 alleles for breeding elite wheat cultivars that are resistant to PHS.
