As an important part of nonstructural components,the seismic response of indoor water supply pipes deserves much attention.This paper presents shaking table test research on water supply pipes installed in a full-scale reinforced concrete(RC)frame structure.Different material pipes and different methods for penetrating the reinforced concrete floors are combined to evaluate the difference in seismic performance.Floor response spectra and pipe acceleration amplification factors based on test data are discussed and compared with code provisions.A seismic fragility study of displacement demand is conducted based on numerical simulation.The acceleration response and displacement response of different combinations are compared.The results show that the combination of different pipe materials and different passing-through methods can cause obvious differences in the seismic response of indoor riser pipes.

Gastric cancer is among the most frequently occurring cancers and a leading cause of cancer-related deaths globally. Because gastric cancer is highly heterogenous and comprised of different subtypes with distinct molecular and clinical characteristics, the management of gastric cancer calls for better-defined, biomarker-guided, molecular-based treatment strategies. MET is a receptor tyrosine kinase mediating important physiologic processes, such as embryogenesis, tissue regeneration, and wound healing. However, mounting evidence suggests that aberrant MET pathway activation contributes to tumour proliferation and metastasis in multiple cancer types, including gastric cancer, and is associated with poor patient outcomes. As such, MET-targeting therapies are being actively developed and promising progress has been demonstrated, especially with MET tyrosine kinase inhibitors. This review aims to briefly introduce the role of MET alterations in gastric cancer and summarize in detail the current progress of MET tyrosine kinase inhibitors in this disease area with a focus on savolitinib, tepotinib, capmatinib, and crizotinib. Building on current knowledge, this review further discusses existing challenges in MET alterations testing, possible resistance mechanisms to MET inhibitors, and future directions of MET-targeting therapies.

High fecundity is a common characteristic of insect pests which increases the difficulty of population control.Serine/threonine kinase Akt is an indispensable component of the insulin signaling pathway.Silencing of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect pest.However,the post-transcriptional pathway of LsAkt in L.serricorne remains unknown.This study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of LsAkt.The expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult stages.Luciferase reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of LsAkt.Overexpression of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian development.It also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of Akt.Conversely,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian development.Injection of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt expression.It also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin signaling.These results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of L.serricorne by targeting Akt in response to insulin signaling.The data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

This study presents a probabilistic seismic risk model for the Beijing–Tianjin–Hebei region in China. The model comprises a township-level residential building exposure model, a vulnerability model derived from the Chinese building taxonomy,and a regional probabilistic seismic hazard model. The three components are integrated by a stochastic event-based method of the OpenQuake engine to assess the regional seismic risk in terms of average annual loss and exceedance probability curve at the city, province, and regional levels. The novelty and uniqueness of this study are that a probabilistic seismic risk model for the Beijing–Tianjin–Hebei region in China is developed by considering the impact of site conditions and epistemic uncertainty from the seismic hazard model.

Traditional blockchain key management schemes store private keys in the same location,which can easily lead to security issues such as a single point of failure.Therefore,decentralized threshold key management schemes have become a research focus for blockchain private key protection.The security of private keys for blockchain user wallet is highly related to user identity authentication and digital asset security.The threshold blockchain private key management schemes based on verifiable secret sharing have made some progress,but these schemes do not consider participants’self-interested behavior,and require trusted nodes to keep private key fragments,resulting in a narrow application scope and low deployment efficiency,which cannot meet the needs of personal wallet private key escrow and recovery in public blockchains.We design a private key management scheme based on rational secret sharing that considers the self-interest of participants in secret sharing protocols,and constrains the behavior of rational participants through reasonable mechanism design,making it more suitable in distributed scenarios such as the public blockchain.The proposed scheme achieves the escrow and recovery of personal wallet private keys without the participation of trusted nodes,and simulate its implementation on smart contracts.Compared to other existing threshold wallet solutions and keymanagement schemes based on password-protected secret sharing(PPSS),the proposed scheme has a wide range of applications,verifiable private key recovery,low communication overhead,higher computational efficiency when users perform one-time multi-key escrow,no need for trusted nodes,and personal rational constraints and anti-collusion attack capabilities.

Numerical Simulation of the Seismic Response of a Horizontal Storage Tank Based on a SPH-FEM Coupling Method

A coupled numerical calculation method combining smooth particle hydrodynamics(SPH)and the finite element method(FEM)was implemented to investigate the seismic response of horizontal storage tanks.Anumericalmodel of a horizontal storage tank featuring a free liquid surface under seismic action was constructed using the SPH–FEM coupling method.The stored liquid was discretized using SPH particles,while the tank and supports were discretized using the FEM.The interaction between the stored liquid and the tank was simulated by using the meshless particle contact method.Then,the numerical simulation results were compared and analyzed against seismic simulation shaking table test data to validate the method.Subsequently,a series of numerical models,considering different liquid storage volumes and seismic effects,were constructed to obtain time history data of base shear and top center displacement,which revealed the seismic performance of horizontal storage tanks.Numerical simulation results and experimental data showed good agreement,with an error rate of less than 18.85%.And this conformity signifies the rationality of the SPH-FEM coupling method.The base shear and top center displacement values obtained by the coupled SPH-FEM method were only 53.3% to 69.1% of those calculated by the equivalent mass method employed in the current code.As the stored liquid volume increased,the seismic response of the horizontal storage tank exhibited a gradual upward trend,with the seismic response increasing from 73% to 388% for every 35% increase in stored liquid volume.The maximum von Mises stress of the tank and the supports remained below the steel yield strength during the earthquake.The coupled SPH-FEM method holds certain advantages in studying the seismic problems of tanks with complex structural forms,particularly due to the representation of the flow field distribution during earthquakes by involving reservoir fluid participation.

Monochamus alternatus is the primary carrier of pine wood nematodes,which pose a serious threat to Pinus spp.in many countries.Newly emerging M.alternatus adults feed on heathy host pines,while matured adults transfer to stressed host pines for mating and oviposition.Several odorant-binding proteins(OBPs)of M.alternatus have been proved to aid in the complex process of host location.To clarify the corresponding relations between OBPs and pine volatiles,more OBPs need to be studied.In this research,MaltOBP19 showed a specific expression in the antennae and mouthparts of M.alternatus,and it was marked in 4 types of antenna sensilla by immunolocalization.Fluorescence binding assays demonstrated the high binding affinity of MaltOBP19 with camphene and myrcene in vitro.In Y-tube olfactory experiments,M.alternatus adults were attracted by camphene and RNAi of OBP19 via microinjection significantly decreased their attraction index.Myrcene induced phobotaxis,but RNAi had no significant effect on this behavior.Further,we found that ingesting dsOBP19 produced by a bacteria-expressed system with a newly constructed vector could lead to the knockdown of MaltOBP19.These results suggest that MaltOBP19 may play a role in the process of host conversion via the recognition of camphene,which has been identified to be strongly released in stressed host pines.In addition,it is proved that knockdown of OBP can be achieved by oral administration of bacteria-expressed double-stranded RNA in M.alternatus adults,providing a new perspective in the control of M.alternatus.

SubclassⅢsucrose nonfermenting1-related protein kinase 2s(SnRK2s)function in ABA and abiotic stress responses by unknown mechanisms.We found that osmotic stress/ABA-activated protein kinase 10(SAPK10),a member of rice SnRK2s,physically interacted with CBL-interacting protein kinase 1(OsCIPK1).OsCIPK1 expression was up-regulated by ABA and PEG treatment,and overexpression increased the ABA sensitivity of seed germination and root growth and plant osmotic stress tolerance.Osmotic stress or ABA-induced activation of OsCIPK1 is dependent on SAPK10.SAPK10 phosphorylates Thr-24 of OsCIPK1 in vitro,and this phosphorylation increases the activity of OsCIPK1 and positively regulates the function of OsCIPK1 in ABA responses and plant osmotic stress tolerance.This study suggests that OsCIPK1 is a direct phosphorylated substrate of SAPK10,and SAPK10-mediated phosphorylation of OsCIPK1 functions in ABA signaling and increases rice osmotic stress tolerance.

In recent years,biochar has attracted considerable attention for soil quality improvement and carbon sequestration due to its unique physicochemical properties.However,the mechanism by which biochar application negatively affects the growth of crop seedlings has not been fully investigated.In this study,a hydroponic experiment was conducted to evaluate the response of rice,wheat,and corn seedlings to biochar application(CK,0 g/L;BC1,0.5 g/L;and BC2,1.0 g/L).Compared with the CK treatment,the BC1 and BC2 treatments decreased the fresh shoot and root weights of rice and corn seedlings(P0.05).For the contents of nutrient elements in seedlings,both BC1 and BC2 treatments hindered the roots from absorbing Fe and Cu and increased the uptake of Ca and Mn.Compared with the CK treatment,the translocation factor(TF)values of Ca,Mn,and Zn were significantly decreased especially in rice seedlings(35.3%-36.8%,68.7%-76.5%,and 29.8%-22.0%,respectively)under the BC1 and BC2 treatments,while only Mn was significantly decreased in wheat and corn seedlings(P<0.05).Transmission electron microscope(TEM)analysis of root cross-sections showed that nano-sized biochar particles(10~23 nm)were found in the root cells under BC2 treatment conditions.Our findings reveal that a large amount of biochar application can reduce nutrient absorption and translocation,and hinder rice,wheat,and corn seedlings,particularly rice seedling,in hydroponic system.

The infection of host plants by many different viruses causes reactive oxygen species(Ros)accumulation and yellowing symptoms,but the mechanisms through which plant viruses counteract RoS-mediated immunity to facilitate infection and symptom development have not been fully elucidated.Most plant viruses are transmitted by insect vectors in the field,but the molecular mechanisms underlying virus-host-insect interactions are unclear.In this study,we investigated the interactions among wheat,barley yellow dwarf virus(BYDV),and its aphid vector and found that the BYDV movement protein(MP)interacts with both wheat catalases(CATs)and the 26S proteasomeubiquitin receptor non-ATPase regulatorysubunit2homolog(PSMD2)to facilitate the 26S proteasome-mediateddegradation of CATs,promotingviral infection,disease symptom development,and aphid transmission.Overexpression of the BYDV MP gene in wheat enhanced the degradation of CATs,which leading to increased accumulation of ROS and thereby enhanced viral infection.Interestingly,transgenic wheat lines overexpressing BYDV MP showed significantly reduced proliferation of wingless aphids and an increased number of winged aphids.Consistent with this observation,silencing of CAT genes also enhanced viral accumulation and reduced the proliferation of wingless aphids but increased the occurrence of winged aphids.In contrast,transgenic wheat plants overexpressing TaCAT1 exhibited the opposite changes and showed increases in grain size and weight upon infection with BYDV.Biochemical assays demonstrated that BYDV MP interacts with PSMD2 and promotes 26S proteasome-mediated degradation of TaCAT1 likely in a ubiquitination-independent manner.Collectively,our study reveals a molecular mechanism by which a plant virus manipulates the Ros production system of host plants to facilitate viral infection and transmission,shedding new light on the sophisticated interactions among viruses,host plants,and insect vectors.
