Development and parentage analysis of SNP markers for Chestnut-vented Nuthatch(Sitta nagaensis)based on ddRAD-seq data

Extra-pair paternity(EPP)is commonly found in socially monogamous birds,especially in small passerine birds,and there are interspecific and intraspecific variations in the extent of EPP.The Chestnut-vented Nuthatch(Sitta nagaensis)is a socially monogamous passerine bird,and verifying whether this species has EPP relies on parentage testing-S.nagaensis is not known to have EPP.In this study,we developed SNP markers of this species that are informative for parentage analysis from double digest restriction site-associated DNA sequencing(ddRAD-seq)data.A panel consisting of 50 SNP markers,with a mean heterozygosity of 0.343,was used to resolve 95% of nestlings to fathers.The combined exclusion probabilities for the first parent and second parent were 0.991 and 0.9999,respectively.This panel of SNP markers is a powerful tool for parentage assignments in S.nagaensis.In addition,we found that three offspring(7.9%)from three nests(23.1%)were the result of extra-pair fertilization out of 38 offspring in 13 nests.Our study provided information on parentage analysis that has not been reported before in S.nagaensis.It also supplemented the understudied EPP behavior of birds in Asia,contributing to a general understanding of the EPP behaviors of birds.

Parkinson’s disease is typically characterized by the progressive loss of dopaminergic neurons in the substantia nigra pars compacta.Many studies have been performed based on the supplementation of lost dopaminergic neurons to treat Parkinson’s disease.The initial strategy for cell replacement therapy used human fetal ventral midbrain and human embryonic stem cells to treat Parkinson’s disease,which could substantially alleviate the symptoms of Parkinson’s disease in clinical practice.However,ethical issues and tumor formation were limitations of its clinical application.Induced pluripotent stem cells can be acquired without sacrificing human embryos,which eliminates the huge ethical barriers of human stem cell therapy.Another widely considered neuronal regeneration strategy is to directly reprogram fibroblasts and astrocytes into neurons,without the need for intermediate proliferation states,thus avoiding issues of immune rejection and tumor formation.Both induced pluripotent stem cells and direct reprogramming of lineage cells have shown promising results in the treatment of Parkinson’s disease.However,there are also ethical concerns and the risk of tumor formation that need to be addressed.This review highlights the current application status of cell reprogramming in the treatment of Parkinson’s disease,focusing on the use of induced pluripotent stem cells in cell replacement therapy,including preclinical animal models and progress in clinical research.The review also discusses the advancements in direct reprogramming of lineage cells in the treatment of Parkinson’s disease,as well as the controversy surrounding in vivo reprogramming.These findings suggest that cell reprogramming may hold great promise as a potential strategy for treating Parkinson’s disease.

In order to improve the stability of Houttuynia cordata flavonoids (HCF), Zein/HCF composite nanoparticles were synthesized from zein (Zein) and HCF by anti-solvent method and characterized for structural analysis. The effects of zein and HCF mass ratio on the composite nanoparticles were further investigated. The results showed that there were intermolecular forces, including hydrogen bonding, hydrophobic and electrostatic interactions, between HCF and zein. When m(Zein)∶m(HCF)=20∶3, Zein/HCF composite nanoparticles exhibited the best performance with the highest melting temperature reaching 85.09 ℃, 12.84 ℃ higher than that of Zein nanoparticles (72.25 ℃), its microstructure uniformly dispersed in a spherical shape with the smallest average particle size and polydispersity index (PDI), the narrowest particle size distribution, and the largest absolute Zeta potential value as well as HCF encapsulation rate. In addition, the composite nanoparticles exhibited higher 1,1-diphenyl-2-picrylhydrazyl and diammonium 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate) radicals scavenging ability, and better heat treatment protection ability by improving the heating stability of HCF. In the simulated gastrointestinal digestion process, the HCF release rate was only 22.31% after 120 min of digestion by simulated gastric fluid (SGF) and reached 75.66% after 120 min of digestion by simulated intestinal fluid (SIF). The HCF release trends in the simulated gastrointestinal fluid were all consistent with the Ritger-Peppas model, but the release mechanisms were different, with a non-Fick diffusion mechanism in SGF while a Fick diffusion mechanism in SIF. © 2024 Fine Chemicals. What is already known about this topic?Kashin-Beck disease(KBD)is a chronic and degenerative osteoarthropathy characterized by cartilage degeneration.It is an endemic disease that is highly prevalent among the Chinese population and poses a significant health risk.What is added by this report?This is the first national report on the economic burden of KBD in China.According to the data from 2021,KBD has caused significant disease and economic burdens.The most substantial reduction in healthy life expectancy was observed among patients with degree II severity and those aged 60 years and older,resulting in a total indirect economic burden of 112.74 million Chinese Yuan(CNY).What are the implications for public health practice?The results of this study will contribute to informing the development of tailored prevention and control strategies by the government.These strategies will include targeted policies and recommendations for appropriate healthcare and financial subsidies,which will be based on the demographic characteristics of the endemic areas.

Recently, a new radio millisecond pulsar(MSP) J1740-5340B, hosted in the globular cluster(GC) NGC 6397,was reported with a 5.78 ms spin period in an eclipsing binary system with a 1.97 days orbital period. Based on a modified radio ephemeris updated by tool tempo2, we analyze the ~15 yr γ-ray data obtained from the Large Area Telescope on board the Fermi Gamma-ray Space Telescope and detect PSR J1740-5340B's γ-ray pulsation at a confidence level of ~4σ with a weighted H-test value of ~26. By performing a phase-resolved analysis, the γ-ray luminosity in on-pulse interval of PSR J1740-5340B is L_(γ)~ 3.8 × 10^(33) erg s^(-1) using NGC 6397's distance of 2.48 kpc. And γ-rays from the on-pulse part of PSR J1740-5340B contribute ~90% of the total observed γ-ray emissions from NGC 6397. No significant γ-ray pulsation of another MSP J1740-5340A in the GC is detected.Considering that the previous four cases of MSPs in GCs, more data in γ-ray, X-ray, and radio are encouraged to finally confirm the γ-ray emissions from MSP J1740-5340B, especially starving for a precise ephemeris.

BACKGROUND Extraction of impacted third molars often leads to severe complications caused by damage to the inferior alveolar nerve(IAN).AIM To proposes a method for the partial grinding of an impacted mandibular third molar(IMM3)near the IAN to prevent IAN injury during IMM3 extraction.METHODS Between January 1996 and March 2022,25 patients with IMM3 roots near the IAN were enrolled.The first stage of the operation consisted of grinding a major part of the IMM3 crown with a high-speed turbine dental drill to achieve sufficient space between the mandibular second molar and IMM3.After 6 months,when the root tips were observed to be away from the IAN on X-ray examination,the remaining part of the IMM3 was completely removed.RESULTS All IMM3s were extracted easily without symptoms of IAN injury after extraction.CONCLUSION Partial IMM3 grinding may be a good alternative treatment option to avoid IAN injury in high-risk cases.

BACKGROUND In this study,we retrospectively analysed macrophage infiltration and podocyte injury in three patients with diffuse proliferative lupus nephritis(LN)who un-derwent repeated renal biopsy.CASE SUMMARY Clinical data of three diffuse proliferative LN patients with different pathological characteristics(case 1 was LN IV-G(A),case 2 was LN IV-G(A)+V,and case 3 was LN IV-G(A)+thrombotic microangiopathy)were reviewed.All patients underwent repeated renal biopsies 6 mo later,and renal biopsy specimens were studied.Macrophage infiltration was assessed by CD68 expression detected by immunohistochemical staining,and an immunofluorescence assay was used to detect podocin expression to assess podocyte damage.After treatment,Case 1 changed to LN III-(A),Case 2 remained as type V LN lesions,and Case 3,which changed to LN IV-S(A),had the worst prognosis.We observed reduced macro-phage infiltration after therapy.However,two of the patients with active lesions after treatment still showed macrophage infiltration in the renal interstitium.Before treatment,the three patients showed discontinuous expression of podocin.Notably,the integrity of podocin was restored after treatment in Case 1.CONCLUSION It may be possible to reverse podocyte damage and decrease the infiltrating ma-crophages in LN patients through effective treatment.

The sediment particles play a huge role in shaping the bed load transport.In this research,240 water-tunnel experiments are carried out to investigate the incipient velocity of the observation particles in two particle arrangements.To accurately predict the incipient velocity of the observation particles,the equation is conceived by the rolling instability mechanism.The incipient velocity equations and experimental data are used to analyze the trend of dispersive pressure and the effect of arrangement position on velocity.We find that it is appropriate to choose the coefficient of drag as 0.261 and the coefficient of lift as 0.198 for the incipient velocity equation of spherical particles on the hemispherical bed surface.Furthermore,the dispersive pressure is closely related to the flow state,particle size,and particle arrangement,which leads to the incipient velocity of the observation particle being at a minimum when the interference particle angle is 45°.Finally,the particle spacing and the projected area changed with the arrangements,directly affecting the incipient velocity of the observed particle.The analysis of four aspects for the coefficients,dispersive pressure,different particle spacing,and projected area will facilitate the prediction of particle incipient velocity,especially on hemispherical beds.
