Laccase（EC 1.***.3.2）is known to oxidize various aromatic and nonaromatic compounds via a radical-catalyzed reaction,which generally includes two types of laccase,Lac1 and ***1 oxidizes toxic compounds in the diet,and Lac2 is known to play an important role in melanizing the insect *** this study,we cloned and sequenced the cDNA of the diamondback moth,Plutella xylostella Lac2（PxLac2）,from the third instar larvae using polymerase chain reaction（PCR）and rapid amplification of cDNA ends *** results showed that the full-length PxLac2 cDNA was 1 944 bp long and had an open reading frame of 1 794 ***2 encoded a protein with 597 amino acids and had a molecular weight of 66.09 ***,we determined the expression levels of PxLac2 in different stages by quantitative PCR（qPCR）.The results indicated that PxLac2 was expressed differently in different *** observed the highest expression level in pupae and the lowest expression level in fourth instar *** also investigated the enzymatic properties of laccase,which had optimal activity at pH 3.0 and at 35°*** these optimal conditions,laccase had a Michaelis constant（Km）of 0.97 mmol L-1,maximal reaction speed（Vm）of 56.82 U mL-1,and activation energy（Ea）of 17.36 kJ mol-1 to oxidize2,2＇-azino-bis（3-ethylbenzothiazoline-6-sulfonic acid ammonium salt）.Type II copper enhanced laccase activity below 0.8mmol L-1 and reduced enzyme activity above 0.8 mmol L-1 with an IC50 concentration of 1.26 mmol *** study provides insights into the biological function of laccase.

Chlorinated coumarins, which are as cytotoxic as highly toxic halobenzoquinones toward CHO-K1 cells, have recently been identified as disinfection byproducts in drinking water disinfection processes. Therefore, detecting coumarins in water samples collected at various stages from drinking water treatment plants helps assess the formation of chlorinated coumarins in drinking water. Hence, a simple, rapid, accurate, and sensitive method for quantifying coumarins in water samples is required. In this study, a method was developed based on solid-phase extraction coupled with high performance liquid chromatography-triple-quadrupole mass spectrometry for analyzing seven coumarins in water samples from drinking water treatment plants, including 6,7-dihydroxycoumarin, 7-hydroxycoumarin, 6-hydroxy-4-methylcoumarin, 8-chloro-7-hydroxycoumarin, coumarin, 7-chloro-6-hydroxy-4-methylcoumarin, and 3,8-dichloro-7-hydroxycoumarin. Sample pretreatment involved solid-phase extraction using HLB columns, followed by elution with water and methanol, each containing 0.25% formic acid. The extracted solution was separated via gradient elution using a Phenomenex Luna C18 column (100 mm×2.0 mm, 3 mm) with 0.1% formic acid aqueous solution and methanol as the mobile phases, with analytes detected by triple-quadrupole mass spectrometry equipped with electrospray ionization source in multiple reaction monitoring mode. The matrix effect, precision, and accuracy of the developed method were investigated using raw and treated water as matrices. Matrix effects of 0.84-1.12 were recorded for the detection of 6,7-dihydroxycoumarin, 7-hydroxycoumarin, 6-hydroxy-4-methylcoumarin, and coumarin in raw water, while values of 0.67-0.70 were recorded for 8-chloro-7-hydroxycoumarin, 7-chloro-6-hydroxy-4-methylcoumarin, and 3,8-dichloro-7-hydroxycoumarin in finished water. Opioid use disorder(OUD)has become a considerable global public health challenge;however,potential medications for the management of OUD that are effective,safe,and nonaddictive are not *** preclinical evidence indicates that antagonists of the dopamine D3 receptor(D3R)have effects on addiction in different animal *** have previously reported that YQA14,a D3R antagonist,exhibits very high affinity and selectivity for D3Rs over D2Rs,and is able to inhibit cocaine-or methamphetamine-induced reinforcement and reinstatement in self-administration *** the present study,our results illustrated that YQA14 dose-dependently reduced infusions under the fixed-ratio 2 procedure and lowered the breakpoint under the progressive-ratio procedure in heroin self-administered rats,also attenuated heroin-induced reinstatement of drug-seeking *** the other hand,YQA14 not only reduced morphine-induced expression of conditioned place preference but also facilitated the extinguishing process in ***,we elucidated that YQA14 attenuated opioid-induced reward or reinforcement mainly by inhibiting morphine-induced up-regulation of dopaminergic neuron activity in the ventral tegmental area and decreasing dopamine release in the nucleus accumbens with a fiber photometry recording *** findings suggest that D3R might play a very important role in opioid addiction,and YQA14 may have pharmacotherapeutic potential in attenuating opioid-induced addictive behaviors dependent on the dopamine system.

Great efforts have been devoted to improve the photocatalytic activity of TiO2 in the visible light region. Rational design of the external structure and adjustment of intrinsic electronic status by impurity doping are two main effective ways to achieve this purpose. A facile onepot synthetic approach was developed to prepare C-doped hollow TiO2 spheres, which simultaneously realized these advantages. The synthesized TiO2 exhibits a mesoporous hollow spherical structure composed of fine nanocrystals, leading to high specific surface area（~180 m^2/g） and versatile porous texture. Carbonate-doping was achieved by a postthermal treatment at a relatively low temperature（200°C）, which makes the absorption edge red-shifted to the visible region of the solar spectrum. Concomitantly, Ti^（3＋） induced by C-doping also functions in improving the visible-light photocatalytic activity by reducing the band gap. There exists a synergistic effect from multiple stimulatives to enhance the photocatalytic effect of the prepared TiO2 catalyst. It is not out of expectation that the asprepared C-doped hollow TiO2 spheres exhibits an improved photocatalytic activity under visible light irradiation in organic pollutant degradation.
