A new polyoxygenated dihydropyrano[2,3-c]pyrrole-4,5-dione derivative, pyranonigrin F （1）, together with a related known compound, pyranonigrin A （2）, were isolated and identified from Penicillium brocae MA-231, an endophytic fungus obtained from the fresh tissue of the marine mangrove plant Avicennia marina. The structures of these metabolites were determined based on comprehensive spectral interpretation and the absolute configuration of compound 1 was established by X-ray crystallographic analysis. Compounds 1 and 2 showed potent activity against a broad spectrum of human-, aqua-, and plant-pathogens.

Two new prenylated indole alkaloids,namely,dihydrocarneamide A（1） and iso-notoamide B（2）,were isolated from the marine-derived endophytic fungus Paecilomyces variotii *** structures of these metabolites were determined based on comprehensive spectral analysis,together with chiral HPLC analysis of the acidic *** other prenylated indole alkaloids such as asperparalines,notoamides,and versicolamides,compounds 1 and 2 are the rare examples of C-5 prenylation,forming the fused dimethyldihydropyran ring at C-5 and C-6 of the indole *** cytotoxic activity of the isolated compounds was evaluated.

Gracilaria lemaneiformis Bory is an economically important alga that is primarily used for agar *** tetraspores are ideal seeds for the cultivation of ***,the most popular culture method is currently based on vegetative fragments,which is labor-intensive and *** this study,we optimized the conditions for tetraspore release and evaluated the photosynthetic activities of different colonies formed from the branches of *** using a PAM(pulse-amplitude-modulated) measuring *** results showed that variations in temperature and salinityhad significant effects on tetraspore ***,variations in the photon flux density(from 15 μmol m-2 s-1 to 480 μmol m-2 s-1) had no apparent effect on tetraspore ***,the PAM-parameters Y(I),Y(Ⅱ),ETR(I),ETR(Ⅱ) and Fv/Fm of colonies formed from different branches showed the same trend:parameter values of first generation branches>second generation branches>third generation *** results suggest that the photosynthetic activities of different colonies of branches changed with the same ***,photosynthesis in *** was found to be involved in vegetative reproduction and tetraspore ***,the first generation branches grew slowly,but accumulated organic compounds to form large numbers of *** together,these results showed that the first generation branches are ideal materials for the release of tetraspores.

In crustaceans, the male sexual dif ferentiation and maintenance are specially regulated by androgenic gland(AG). However, little is known about the genes involved in the regulation ***-Seq was performed on AG with ejaculatory duct(AG_ED) and ejaculatory duct(ED) as control in Eriocheir sinensis, one of the most important economic and ?shery crabs with typically sex dimorphism. A total of 925 unigenes were identi?ed as dif ferentially expressed genes(DEGs) and the expression of nine genes randomly selected was con?rmed by qRT-PCR. 667 unigenes were up-regulated in AG_ED, being supposed to be AG preferential genes. Among them, the full length of i nsulin-like androgenic gland factor( IAG) cDNA named as Es-IAG was obtained as a logo gene of AG, which together with the genes i nsulin-like receptor( INR), and s ingle insulin binding domain protein( SIBD), might constitute the sex regulation pathway. Several sex related genes were identi?ed, and their function will have to be investigated. Also,the identi?cation of j uvenile hormone epoxide hydrolase 1( JHEH1), ecdysteroid 22-hydroxylase( DIB) and e cdysone receptor( ECR) preliminarily clari?ed the molecular regulation mechanism of eyestalk-AG-testis axis, which plays important roles in molting and reproduction. The results will enhance our understanding for the molecular basis of the AG involved in male sex regulation in crabs.

Pyropia yezoensis, belongs to the genus of P orphyra before 2011, inhabit on intertidal zone rocks where irradiation changes dramatically, implying that the seaweed has gained certain mechanisms to survive a harsh environment. Based on the photosynthetic parameters with or without the inhibitors determined by a Dual-PAM-100 apparatus, we investigated the photosynthetic performance and the changes in electron fl ow that occurred during the algae were stressed with dif ferent light intensities previously. When the irradiation saturation was approaching, the CEF around PS I became crucial since the addition of inhibitors usually led to an increase in non-photochemical quenching. The inhibitor experiments showed that there were at least three dif ferent CEF pathways in Py. yezoensis and these pathways compensated each other. In addition to maintaining a proper ratio of ATP/NAD(P)H to support effi cient photosynthesis, the potential roles of CEF might also include the regulation of dif ferent photoprotective mechanisms in Py. yezoensis. Under the regulation of CEF, chlororespiration is thought to transport electrons from the reduced plastoquinone(PQ) pool to oxygen in order to mitigate the reduction in the electron transfer chain. When irradiation was up to the high-grade stress conditions, the relative value of CEF began to decrease, which implied that the NADP+ pool or PQ + pool was very small and that the electrons were transferred from reduced PS I to oxygen. The scavenging enzymes might be activated and the water-water cycle probably became an ef fective means of removing the active oxygen produced by the irradiation stressed Py. yezoensis. We believe that the dif ferent mechanisms could make up the photoprotective network to allow Py. yezoensis for survival in a highly variable light stress habitat, which may enlighten scientists in future studies on irradiance stress in other algae species.

Previous studies have shown that deep learning is very effective in detecting known ***,when facing unknown attacks,models such as Deep Neural Networks(DNN)combined with Long Short-Term Memory(LSTM),Convolutional Neural Networks(CNN)combined with LSTM,and so on are built by simple stacking,which has the problems of feature loss,low efficiency,and low ***,this paper proposes an autonomous detectionmodel for Distributed Denial of Service attacks,Multi-Scale Convolutional Neural Network-Bidirectional Gated Recurrent Units-Single Headed Attention(MSCNN-BiGRU-SHA),which is based on a Multistrategy Integrated Zebra Optimization Algorithm(MI-ZOA).The model undergoes training and testing with the CICDDoS2019 dataset,and its performance is evaluated on a new GINKS2023 *** hyperparameters for Conv_filter and GRU_unit are optimized using the Multi-strategy Integrated Zebra Optimization Algorithm(MIZOA).The experimental results show that the test accuracy of the MSCNN-BiGRU-SHA model based on the MIZOA proposed in this paper is as high as 0.9971 in the CICDDoS 2019 *** evaluation accuracy of the new dataset GINKS2023 created in this paper is *** to the MSCNN-BiGRU-SHA model based on the Zebra Optimization Algorithm(ZOA),the detection accuracy on the GINKS2023 dataset has improved by 5.81%,precisionhas increasedby 1.35%,the recallhas improvedby 9%,and theF1scorehas increasedby 5.55%.Compared to the MSCNN-BiGRU-SHA models developed using Grid Search,Random Search,and Bayesian Optimization,the MSCNN-BiGRU-SHA model optimized with the MI-ZOA exhibits better performance in terms of accuracy,precision,recall,and F1 score.

Seven compounds (1–7) were identified from the cultivation of the endophytic fungus Exophiala oligosperma (EN-21) that was isolated from the inner tissue of the marine red alga Laurencia similis. Their structures were identified with spectroscopic and chemical methods as 2-phenoxynaphthalene (1), (2S, 3R, 4E, 8E)-1-O-β-D-glucopyranosyl-3-hydroxy-2-[(R)-2′-hydroxyoctadecanoyl] amino-9-methyl-4, 8-octadeca-diene (2), (22E,24R)-ergosta-7,22-dien-3β,5α,6β-triol (3), (22E, 24R)-3β, 5α, 9α-trihydroxy- ergosta-7, 22-dien-6-one (4), (22E, 24R)-5α, 6α-epoxy-ergosta-8, 22-dien-3β, 7α-diol (5), (22E, 24R)- ergosta-4, 6, 8(14), 22-tetraen-3-one (6), and euphorbol (7). This paper reports for the first time the chemical constituents of fungus Exophiala oligosperma and the discovery of compound 1 as a natural product from the fungus.

Platymonas (Tetraselmis) subcordiformis is a unicellular marine green alga. It was found to be very sensitive to the herbicide Basta through a sensitivity test indicating it could be employed as a selective agent. The bar gene is a practicable and selectable marker gene. The vector containing the expression cassette of the bar gene was transferred to P. subcordiformis by both particle bombardment and glass-bead agitation and transformants were then selected using Basta. Finally, Southern blotting analysis indicated that the bar gene had been successfully integrated into the nuclear genome of P. subcordiformis using both of the transgenic techniques, with the transformation efficiency of the glass-bead method being slightly higher than that of particle bombardment. This is the first report on stable transformation of P. subcordiformis, and will improve fundamental research and enlarge application of this alga.

Exploring valid internal-control genes in Porphyra yezoensis(Bangiaceae) during stress response conditions

To screen the stable expression genes related to the stress （strong light, dehydration and temperature shock） we applied Absolute real-time PCR technology to determine the transcription numbers of the selected test genes in Porphyra yezoensis, which has been regarded as a potential model species responding the stress conditions in the intertidal. Absolute real-time PCR technology was applied to determine the transcription numbers of the selected test genes in Porphyra yezoensis, which has been regarded as a potential model species in stress responding. According to the results of photosynthesis parameters, we observed that Y（II） and Fv/Fm were significantly affected when stress was imposed on the thalli of Porphyra yezoensis, but underwent almost completely recovered under normal conditions, which were collected for the following experiments. Then three samples, which were treated with different grade stresses combined with salinity, irradiation and temperature, were collected. The transcription numbers of seven constitutive expression genes in above samples were determined after RNA extraction and cDNA synthesis. Finally, a general insight into the selection of internal control genes during stress response was obtained. We found that there were no obvious effects in terms of salinity stress （at salinity 90） on transcription of most genes used in the study. The 18S ribosomal RNA gene had the highest expression level, varying remarkably among different tested groups. RPS8 expression showed a high irregular variance between samples. GAPDH presented comparatively stable expression and could thus be selected as the internal control. EF-la showed stable expression during the series of multiple-stress tests. Our research provided available references for the selection of internal control genes for transcripts determination of P. yezoensis.
