The miR159a-DUO1 module regulates pollen development by modulating auxin biosynthesis and starch metabolism in citrus

Achieving seedlessness in citrus varieties is one of the important objectives of citrus *** sterility associated with abnormal pollen development is an important factor in ***,our understanding of the regulatory mechanism underlying the seedlessness phenotype in citrus is still ***,we determined that the miR159a-DUO1 module played an important role in regulating pollen development in citrus,which further indirectly modulated seed development and fruit *** the overexpression of csi-miR159a and the knocking out of DUO1 in Hong Kong kumquat(Fortunella hindsii)resulted in small and seedless fruit ***,pollen was severely aborted in both transgenic lines,with arrested pollen mitotic I and abnormal pollen starch *** additional cross-pollination experiments,DUO1 was proven to be the key target gene for miR159a to regulate male sterility in *** on DNA affinity purification sequencing(DAP-seq),RNA-seq,and verified interaction assays,YUC2/YUC6,SS4 and STP8 were identified as downstream target genes of DUO1,those were all positively regulated by *** transgenic *** lines,the miR159a-DUO1 module down-regulated the expression of YUC2/YUC6,which decreased indoleacetic acid(IAA)levels and modulated auxin signaling to repress pollen mitotic *** miR159a-DUO1 module reduced the expression of the starch synthesis gene SS4 and sugar transport gene STP8 to disrupt starch metabolism in ***,this work reveals a new mechanism by which the miR159a-DUO1 module regulates pollen development and elucidates the molecular regulatory network underlying male sterility in citrus.

Flower development and sexual dimorphism in Vernicia montana

The mu oil tree(Vernicia montana Lour.) is a dioecious species, but the genetic mechanisms underlying its phenotypic sexual dimorphism are unclear. In this study, we determined two pivotal phases of sex differentiation of mu oil tree via morphological and histological analyses of unisexual flowers:(Ⅰ) differentiation of male or female primordia to produce staminate flowers(SFs) or transient hermaphrodite flowers(HFs),and(Ⅱ) complete abortion of stamens in transient HFs to generate pistillate flowers(PFs). A total of 1621 sex-biased genes were identified by comparative transcriptome analysis which exhibited elevated rates of protein evolution than unbiased genes. The female-biased genes were enriched in the production of defense compounds while male-biased genes were focused on the production of viable pollens. Transcriptomebased analysis revealed that the differentially expressed genes(DEGs) between PFs and SFs in phase Ⅰ involved in abscisic acid(ABA), auxin(AUX), cytokinin(CK), ethylene(ET), and gibberellin(GA) biosynthesis and signaling showed higher expression levels in males than in females in general, whereas the DEGs involved in jasmonic acid(JA) and salicylic acid(SA) pathways displayed opposite expression patterns. Moreover,differentially expressed endogenous ABA, AUX, GAs, JA, and SA exhibited consistent biased expression patterns with the DEGs by UPLC-MSbased analysis. Exogenous application of an anti-ethylene plant growth regulator could promote the development of stamens in PFs and generated HFs. Comparative transcriptomic and hormonal analyses of PFs and SFs in phase Ⅱ indicated an increase in ET concentration when abortion of stamens in PFs occurred. This study suggested that phytohormones play key roles in sex dimorphism and ET may determine the development of stamens in PFs of mu oil tree, which provides an insight into plant sex differentiation mechanisms.

High temperature treatment induced production of unreduced 2n pollen in Camellia oleifera

Rice Bran-derived Peptide KF-8 Attenuates Dexamethasone-induced Myopathy in Caenorhabditis elegans by Regulating Locomotion-related Genes

Background and aims: Dexamethasone is a common glucocorticoid medication with adverse effects that can cause muscle atrophy， but no drug intervention has been approved or recommended for this condition. KF-8 is a rice bran-derived anti-oxidant peptide that extends the life span of Caenorhabditis elegans. Methods: We established a C. elegans model of dexamethasone-induced myopathy to evaluate the potential therapeutic effects of KF-8 in this model. C. elegans muscle function was assessed in terms of locomotory behaviors including crawling， swimming， burrowing， pharyngeal pumping， and head swing. Muscle actin filament integrity was evaluated using fluorescence imaging. The molecular mechanisms of KF-8 were investigated using transcriptome sequencing， qRT-PCR， RNA interference， and western blot analysis. Results: Dexamethasone disrupted actin filaments in the striated muscles of the body wall and inhibited C. elegans crawling， swimming， burrowing， pharyngeal pumping， and head swing. KF-8 reversed the actin filament disruption and locomotor dysfunction induced by dexamethasone. Transcriptome sequencing， pathway enrichment， and qRT-PCR analyses revealed that KF-8 regulated the locomotion-related genes W04G5.10， vha-12， and ddr-1， as well as age-1 (the catalytic subunit ortholog of PI3K)， and akt1. RNA interference， conducted using a genetically engineered E. coli HT115 strain as a food source， confirmed age-1 as a key regulator of locomotor function of C. elegans. Further mechanistic studies with C2C12 myotubes showed that KF-8 regulated the IRS-PI3K-Akt pathway， the master regulator of protein synthesis and degradation. Conclusion: Together， these findings suggest that KF-8 protects against dexamethasone-induced myopathy in C. elegans by regulating locomotion-related genes and the IRS-PI3K-Akt pathway.

Camellia oil extracted from the seeds of Camellia oleifera *** a popular and high-quality edible oil,but its yield is limited by seed setting,which is mainly caused by self-incompatibility(SI).One of the obvious biological features of SI plants is the inhibition of self-pollen tubes;however,the underlying mechanism of this inhibition in *** is poorly *** this study,we constructed a semi-in vivo pollen tube growth test(SIV-PGT)system that can screen for substances that inhibit self-pollen tubes without interference from the genetic *** with multi-omics analysis,the results revealed the important role of galloylated catechins in self-pollen tube inhibition,and a possible molecular regulatory network mediated by UDP-glycosyltransferase(UGT)and serine carboxypeptidase-like(SCPL)was *** summary,galloylation of catechins and high levels of galloylated catechins are specifically involved in pollen tube inhibition under self-pollination rather than cross-pollination,which provides a new understanding of SI in *** results will contribute to sexual reproduction research on *** and provide theoretical support for improving Camellia oil yield in production.

Effects of IBA on the rooting of branch cuttings of Chinese jujube (Zizyphus jujuba Mill.) and changes to nutrients and endogenous hormones

‘Zhongqiusucui’ jujube secondary shoots were treated with 3-indolebutyric acid(IBA) at three concentrations, 500, 1000 and 1500 mg/L. Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root *** hormones in leaf tips and bases had an impact on rooting. The 218th amino acid change of Ser to Ala in TaAGPS-7A increases enzyme activity and grain weight in bread wheat

ADP-glucose pyrophosphorylase(AGPase)influences cereal *** are few reports on the function of cytosolic AGPase small subunit in bread wheat(TaAGPS).In the present study,TaAGPS was preferentially expressed in developing endosperm during grain-filling stages in bread *** allelic variations were characterized in 143 wheat accessions by PacBio RS II *** haplotypes(TaAGPS-7A-TG and TaAGPS-7A-CT)of TaAGPS-7A were identified and corresponding functional markers were developed,whereas no variants of TaAGPS-7B and TaAGPS-7D were ***-7A was associated with thousand-kernel weight(TKW)by haplotype–trait association analysis in two ***-isogenic lines(NILs)with TaAGPS-7A-TG showed higher TKW and total kernel starch content than those with TaAGPS-7A-CT,owing to the higher AGPase activity of TaAGPS-7A-TG than TaAGPS-7A-CT both in vitro and in *** of TaAGPS-7A-TG in bread wheat doubled the transcription levels of TaAGPS and increased AGPase activity by 55.7%,resulting in a 3.0-g higher TKW than in the wild type(WT).Knockdown of TaAGPS led to reduced expression of TaAGPS,AGPase activity,and TKW than in the ***,owing to the 218th amino acid change of Ser to Ala in TaAGPS-7A,the favorable haplotype TaAGPS-7A-TG showed higher AGPase activity,resulting in higher kernel starch content and grain *** finding could be applied to increasing starch content and grain weight in bread wheat.

Two new Tenthredo species(Hymenoptera: Tenthredinidae) from China

Tenthredo bicuspis Liu Wei sp. nov. and T. biminutidota Liu Wei sp. nov. are described from China. Tenthredo bicuspis is similar to T. amurica Dalla Torre, 1894 from Siberia in Russia but differs in the pterostigma yellow brown, the upper inner orbits, temple, pronotum, tegula, mesoscutellum and mesepisternum with distinct white maculae, the mesoscutellum strongly elevated with two obtuse peaks, the dorsum of head impunctate, the dorsum of thorax and mesopleuron shiny, the supraantennal tubercles strongly elevated with the posterior end abruptly cut off. Tenthredo biminutidota is similar to T. flavobrunneus Malaise,1945 but differs in the postocellar area about 1.5 times as broad as long, the lateral furrows distinctly divergent backwards; the black macula on dorsum of head very large and almost touching the eye or nearly so, the supraantennal tubercles largely black and evenly merging with frontal walls; the mesoscutellum and mesepisternum not strongly elevated with a low peak; each abdominal tergite with a narrow basal black band and a pair of black dots laterally; the female hypopygium with a large middle process and serrulae hardly elevated.
