The 218th amino acid change of Ser to Ala in TaAGPS-7A increases enzyme activity and grain weight in bread wheat

ADP-glucose pyrophosphorylase(AGPase)influences cereal *** are few reports on the function of cytosolic AGPase small subunit in bread wheat(TaAGPS).In the present study,TaAGPS was preferentially expressed in developing endosperm during grain-filling stages in bread *** allelic variations were characterized in 143 wheat accessions by PacBio RS II *** haplotypes(TaAGPS-7A-TG and TaAGPS-7A-CT)of TaAGPS-7A were identified and corresponding functional markers were developed,whereas no variants of TaAGPS-7B and TaAGPS-7D were ***-7A was associated with thousand-kernel weight(TKW)by haplotype–trait association analysis in two ***-isogenic lines(NILs)with TaAGPS-7A-TG showed higher TKW and total kernel starch content than those with TaAGPS-7A-CT,owing to the higher AGPase activity of TaAGPS-7A-TG than TaAGPS-7A-CT both in vitro and in *** of TaAGPS-7A-TG in bread wheat doubled the transcription levels of TaAGPS and increased AGPase activity by 55.7%,resulting in a 3.0-g higher TKW than in the wild type(WT).Knockdown of TaAGPS led to reduced expression of TaAGPS,AGPase activity,and TKW than in the ***,owing to the 218th amino acid change of Ser to Ala in TaAGPS-7A,the favorable haplotype TaAGPS-7A-TG showed higher AGPase activity,resulting in higher kernel starch content and grain *** finding could be applied to increasing starch content and grain weight in bread wheat.

Two new Tenthredo species(Hymenoptera: Tenthredinidae) from China

Tenthredo bicuspis Liu Wei sp. nov. and T. biminutidota Liu Wei sp. nov. are described from China. Tenthredo bicuspis is similar to T. amurica Dalla Torre, 1894 from Siberia in Russia but differs in the pterostigma yellow brown, the upper inner orbits, temple, pronotum, tegula, mesoscutellum and mesepisternum with distinct white maculae, the mesoscutellum strongly elevated with two obtuse peaks, the dorsum of head impunctate, the dorsum of thorax and mesopleuron shiny, the supraantennal tubercles strongly elevated with the posterior end abruptly cut off. Tenthredo biminutidota is similar to T. flavobrunneus Malaise,1945 but differs in the postocellar area about 1.5 times as broad as long, the lateral furrows distinctly divergent backwards; the black macula on dorsum of head very large and almost touching the eye or nearly so, the supraantennal tubercles largely black and evenly merging with frontal walls; the mesoscutellum and mesepisternum not strongly elevated with a low peak; each abdominal tergite with a narrow basal black band and a pair of black dots laterally; the female hypopygium with a large middle process and serrulae hardly elevated.

Review of Propodea Malaise(Hymenoptera: Tenthredinidae) with descriptions of two new species, three new records from China and a new record from Nepal

Propodea Malaise, 1945 is reviewed and redescribed. Seven species of Propodea are *** sinica Wu Wei sp. nov. and P. xanthocera Wu Wei sp. nov. are described as new to *** rotundiventris（Cameron, 1899）, P. fentoni（Kirby, 1882） and Propodea ussuriensis（Malaise, 1931）are recorded for the first time from China. A key to species of Propodea Malaise is also provided. Tenthredo rufonotalis Mallach, 1936 is removed from being a junior synonym of Dipteromorpha ussuriensis Malaise,1931. Propodea rufonotalis（Mallach, 1936） is a new combination transferred from Tenthredo. The species previously recorded as Propodea rotundiventris（Cameron, 1899） from China before this paper is a misidentification of P. sinica sp. nov. The real Propodea rotundiventris（Cameron, 1899） is redescribed based on the holotype and new material. Propodea spinosa（Cameron, 1899） is recorded for the first time from Nepal.

Factors involved in the success of Castanea henryi stem cuttings in different cutting mediums and cutting selection periods

Chinquapin(Castanea henryi) is a dual-purpose tree species in China valued for as a source of timber and *** investigated the effect of four cutting mediums(pure vermiculite;peat:river sand at 3:1 v/v;peat:krasnozem at 1:1 v/v;and pure krasnozem) and three stem cutting periods(March,May,and July) on rooting performance of *** *** cutting periods and cutting mediums greatly influenced the rooting rate of ***,ranging from 3.35 to 77.31%.Principal component analysis indicated that the best combination of cutting period and cutting medium was semi-hardwood cuttings(May cuttings)+*** evidence indicated that adventitious root initials were present by week 5-6,and that the site of root primordia initiation was observed in the vascular *** anatomical structures observed at different periods indicated that a xylem/radius ratio of29.90-37.42% and a fractured phloem fiber ring are indicative of rooting *** relational model between rooting index and medium properties indicated that nutrient content and porosity significantly influenced callus ***,pH strongly affected *** root formation,with the Pearson correlation coefficients for May and July cuttings of-0.856 and-0.947,*** protocol is helpful to achieve mass clone propagation of improved *** genotypes,thus overcoming a common hurdle in chinquapin breeding programs.

Two new species of Janus Stephens(Hymenoptera:Cephidae) from China with a key to Chinese species

The diagnosis of Janus Stephens 1829 and a key to the Chinese species of the genus is provided. Janus rnegarnaculatus Liu ＆ Wei sp. nov. and J. rufus Liu ＆ Wei sp. nov. are described from China. Janus megamaculatus sp. nov. differs from its known congeners by the body very large, head and thorax yellow brown with few black macula, the abdomen yellow brown with transverse black bands, the mesoscutal middle lobe with a deep middle furrow and the apical sheath as long as the basal sheath. Janus rufus sp. nov. is similar to Janus xanthus Naito ＆ Smith, 1998 in structure but differs from the latter by the head and thorax almost entirely yellow brown, the antenna brown and about 2 times as long as head and thorax together, the hind femur yellow brown, the cell Rs in hindwing closed and cereus 0.14 times as long as the apical sheath.

Establishment of Self-incompatibility Gene cDNA Microarray to Identify S-genotypes of Pyrus pyrifolia

Based on the c DNA sequences from hyper variable(HV) regions of identified 52 S-alleles in Oriental pear cultivars, S-RNase c DNA probes were designed, and a c DNA microarray for S-RNase detections was established. Each microarray contained 240 sites from 55 c DNA probes, including all specific c DNA sequences from the HV regions of the S-alleles. Using the c DNA of pistils of tested pear cultivars as template and Cy3 fluorescently labeling primers by PCR amplification, microarray hybridization detected the S-genotype of each pear cultivar. The genotypes inferred from the c DNA microarray hybridization signals of pear cultivars such as ‘Lijiang Huangsuanli', ‘Xiuyu', ‘Midu Yuli', ‘Baimianli', and ‘Deshengxiang' were similar to the known genotypes of all tested cultivars. The S-RNase c DNA microarrays and the oligonucleotide gene chips were then used to conduct parallel testing of 24 P. pyrifolia cultivars with unknown S-genotypes. In conclusion, the construction of c DNA microarrays has further improved the pear S-RNase detection platform.

Loquat(Eriobotrya japonica Lindl.)is an evergreen fruit tree species of the Rosaceae,and its unique flowering time greatly hinders its *** explore the artificial regulation of loquat flowering time,we removed the main inflorescence(by cutting it)to induce *** different loquat tree cultivars with different stages,the inflorescence was removed by cutting the main floral axis at two alternative positions:the upper or the lower position beneath the inflorescence,and it was found that the proportion of reflowering resulting from removing the upper position of the main floral axis of yellow-flesh loquat cultivars during the full-bloom stage was the *** addition,compared with those of the normal-growing panicles,the number of flower buds and branch axes of the reflowering panicles decreased significantly after ***,these newly produced inflorescences flowered 2–4 months later than normal-growing inflorescences did,effectively prolonging both the flowering and fruiting *** addition,qRT-PCR results showed that EjFT1,EjFT2,EjAP1–1 and EjAP1–2 were highly expressed in the floral *** findings highlighted a new method for extending the production cycle of loquat and provided a reference for the flowering regulation of loquat and other economically important fruit tree species.
