The miR159a-DUO1 module regulates pollen development by modulating auxin biosynthesis and starch metabolism in citrus

Flower development and sexual dimorphism in Vernicia montana

The mu oil tree(Vernicia montana Lour.) is a dioecious species, but the genetic mechanisms underlying its phenotypic sexual dimorphism are unclear. In this study, we determined two pivotal phases of sex differentiation of mu oil tree via morphological and histological analyses of unisexual flowers:(Ⅰ) differentiation of male or female primordia to produce staminate flowers(SFs) or transient hermaphrodite flowers(HFs),and(Ⅱ) complete abortion of stamens in transient HFs to generate pistillate flowers(PFs). A total of 1621 sex-biased genes were identified by comparative transcriptome analysis which exhibited elevated rates of protein evolution than unbiased genes. The female-biased genes were enriched in the production of defense compounds while male-biased genes were focused on the production of viable pollens. Transcriptomebased analysis revealed that the differentially expressed genes(DEGs) between PFs and SFs in phase Ⅰ involved in abscisic acid(ABA), auxin(AUX), cytokinin(CK), ethylene(ET), and gibberellin(GA) biosynthesis and signaling showed higher expression levels in males than in females in general, whereas the DEGs involved in jasmonic acid(JA) and salicylic acid(SA) pathways displayed opposite expression patterns. Moreover,differentially expressed endogenous ABA, AUX, GAs, JA, and SA exhibited consistent biased expression patterns with the DEGs by UPLC-MSbased analysis. Exogenous application of an anti-ethylene plant growth regulator could promote the development of stamens in PFs and generated HFs. Comparative transcriptomic and hormonal analyses of PFs and SFs in phase Ⅱ indicated an increase in ET concentration when abortion of stamens in PFs occurred. This study suggested that phytohormones play key roles in sex dimorphism and ET may determine the development of stamens in PFs of mu oil tree, which provides an insight into plant sex differentiation mechanisms.

High temperature treatment induced production of unreduced 2n pollen in Camellia oleifera

Rice Bran-derived Peptide KF-8 Attenuates Dexamethasone-induced Myopathy in Caenorhabditis elegans by Regulating Locomotion-related Genes

Background and aims: Dexamethasone is a common glucocorticoid medication with adverse effects that can cause muscle atrophy， but no drug intervention has been approved or recommended for this condition. KF-8 is a rice bran-derived anti-oxidant peptide that extends the life span of Caenorhabditis elegans. Methods: We established a C. elegans model of dexamethasone-induced myopathy to evaluate the potential therapeutic effects of KF-8 in this model. C. elegans muscle function was assessed in terms of locomotory behaviors including crawling， swimming， burrowing， pharyngeal pumping， and head swing. Muscle actin filament integrity was evaluated using fluorescence imaging. The molecular mechanisms of KF-8 were investigated using transcriptome sequencing， qRT-PCR， RNA interference， and western blot analysis. Results: Dexamethasone disrupted actin filaments in the striated muscles of the body wall and inhibited C. elegans crawling， swimming， burrowing， pharyngeal pumping， and head swing. KF-8 reversed the actin filament disruption and locomotor dysfunction induced by dexamethasone. Transcriptome sequencing， pathway enrichment， and qRT-PCR analyses revealed that KF-8 regulated the locomotion-related genes W04G5.10， vha-12， and ddr-1， as well as age-1 (the catalytic subunit ortholog of PI3K)， and akt1. RNA interference， conducted using a genetically engineered E. coli HT115 strain as a food source， confirmed age-1 as a key regulator of locomotor function of C. elegans. Further mechanistic studies with C2C12 myotubes showed that KF-8 regulated the IRS-PI3K-Akt pathway， the master regulator of protein synthesis and degradation. Conclusion: Together， these findings suggest that KF-8 protects against dexamethasone-induced myopathy in C. elegans by regulating locomotion-related genes and the IRS-PI3K-Akt pathway.

Effects of IBA on the rooting of branch cuttings of Chinese jujube (Zizyphus jujuba Mill.) and changes to nutrients and endogenous hormones

‘Zhongqiusucui’ jujube secondary shoots were treated with 3-indolebutyric acid(IBA) at three concentrations, 500, 1000 and 1500 mg/L. Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root *** hormones in leaf tips and bases had an impact on rooting. Unfavorable environmental cues severely affect crop productivity resulting in significant economic losses to farmers. In plants, multiple regulatory genes, such as the WRKY transcription factor (TF) family, modulate the expression of defense genes. However, the role of the pineapple WRKY genes is poorly understood. Here, we studied the pineapple WRKY gene, AcWRKY28, by generating AcWRKY28 over-expressing transgenic pineapple plants. Overexpression of AcWRKY28 enhanced the salt stress resistance in transgenic pineapple lines. Comparative transcriptome analysis of transgenic and wild-type pineapple plants showed that “plant-pathogen interaction” pathway genes, including 9calcium-dependent protein kinases (CPKs), were up-regulated in AcWRKY28 over-expressing plants. Furthermore, chromatin immunoprecipitation and yeast one-hybrid assays revealed AcCPK12, AcCPK3, AcCPK8, AcCPK1, and AcCPK15 as direct targets of AcWRKY28. Consistently, the study of AcCPK12 over-expressing Arabidopsis lines showed that AcCPK12 enhances salt, drought, and disease resistance. This study shows that AcWRKY28 plays a crucial role in promoting salt stress resistance by activating the expression of AcCPK genes.

Natural product chlorogenic acid achieves pharmacological activity and health protection via regulating gut microbiota

Chlorogenic acid (CA) is a natural plant-derived polyphenol compound that is widely present in beverages (coffee and tea)， fruits (blueberry， apple， and mulberry)， and medicinal plants (Lonicera japonica， Eucommia ulmoides， and Cichorium intybus). In recent years， CA has attracted extensive attention due to its various health benefits， such as anti-inflammatory， immunomodulatory， anti-obesity， anti-diabetes mellitus， and neuroprotection activities. Interestingly， the low bioavailability of CA corresponds to high bioactivity， which raises a key scientific question of how low bioavailability CA can achieve high biological activity. In this review， we highlight the gut microbiota as a key target for connecting the two by summarizing a large amount of evidence. Specifically， the composition and abundance of gut microbiota， its metabolites， intestinal immunity， barrier function and so on were all changed under different pathological conditions such as inflammatory bowel disease， liver disease， kidney disease， cardiovascular disease， diabetes mellitus， obesity， etc. Conversely， these changes could be reversed by CA. In a word， CA could achieve pharmacological activity as well as health-protective effects by modulating intestinal immune and barrier function， gut microbiota composition， gut microbiota metabolites， and signaling pathways. This review provides new targets for the prevention and treatment of diseases by CA. Meanwhile， regulating the composition of the gut microbiota through natural products may be a potential strategy to achieve health protection.

Residue behaviors and dietary risk assessment of dinotefuran and its metabolites in Lycium barbarum from farm to fork

Lycium barbarum， known as wolfberry or goji berry， is consumed by humans as a medicine and a food homology product. Conventionally grown wolfberry is often treated extensively with pesticides， which could pose a hazard to humans. Here， the degradation dynamics of dinotefuran and its two metabolites (UF and DN)， during wolfberry cultivation and processing was investigated. The half-life (T1/2) of dinotefuran was 11.36 d and 9.12 d， respectively， under the recommended dosage and double the recommended dosage. During the oven and sun drying processes， processing factors (PFs) of dinotefuran were 1.07 - 1.34， implying the enrichment of pesticide residues. Decoction process made the removal rate of dinotefuran reach 87.48%， which is higher than that of the brewing process (14.7%)， while dinotefuran remained in the wine with high ethanol content in the alcohol soaking process. The hazard quotient (HQ) of dinotefuran， as determined via dietary risk assessment combined with PFs， was gastric (5.45% - 86.72%) > oral (23.18%) via in vitro simulated digestive system. These findings provide scientific evidence for reasonable application and risk assessment of dinotefuran residues in wolfberry.
