Characterization,expression dynamics,and potential function of OPA1 for regulation of mitochondrial morphology during spermiogenesis in Phascolosoma esculenta

Mitochondria undergo morphological changes during spermatogenesis in some *** mechanism and role of mitochondrial morphology regulation,however,remain somewhat *** this study,we analyzed the molecular characteristics,expression dynamics and subcellular localization of optic atrophy protein 1(OPA1),a mitochondrial fusion and cristae maintenance-related protein,to reveal the possible regulatory mechanisms underlying mitochondrial morphology in Phascolosoma esculenta *** full-length cDNA of the *** opa1 gene(Pe-opa1)is 3743 bp in length and encodes 975 amino *** Pe-OPA1 protein is highly conservative and includes a transmembrane domain,a GTPase domain,two helical bundle domains,and a lipid-interacting *** and protein expression was higher in the coelomic fluid(a site of spermatid development)of male *** and increased first and then decreased from March to ***,their expression during the breeding stage was significantly higher than during the non-breeding stage,suggesting that Pe-OPA1 is involved in *** *** Pe-OPA1 protein was more abundant in components consisting of many spermatids than in components without,indicating that Pe-OPA1 mainly plays a role in the spermatid in coelomic ***,Pe-OPA1 was mainly detected in the spermatid *** experiments showed that the Pe-OPA1 are constitutively expressed and co-localized with mitochondria during spermiogenesis,suggesting its involvement in *** *** results provide evidence for Pe-OPA1's involvement in the regulation of mitochondrial morphology during spermiogenesis.

Combined proteomic and metabolomic studies on the liver of Amur sturgeon Acipenser schrenckii under titanium dioxide nanoparticle exposure

Nanomaterials,particularly titanium dioxide nanoparticles(TiO_(2)-NPs),are extensively utilized across various ***,their environmental release has raised concerns regarding their potential ecological and environmental *** reproductive toxicity of TiO_(2)-NPs in fish species has attracted considerable attention,yet conflicting research outcomes have been *** investigated the effects of TiO_(2)-NPs exposure on the liver of juvenile Amur sturgeon Acipenser schrenckii using label-free proteomic and untargeted metabolomic *** experiment included a control group and three groups exposed to different concentrations of TiO_(2)-NPs(low,TL;medium,TM;high,TH).Compared to the control group,9,19,and 25 proteins and 35,73,and 158 metabolites were differentially expressed in the TH,TM,and TL TiO_(2)-NP-exposed groups,*** differentially expressed genes(DEGs)were enriched in the Kyoto Encyclopedia for Genes and Genomes(KEGG)pathways related to glycolysis and ***,among the 126 correlated proteins,the most enriched pathways were associated with endocytosis and protein processing in the endoplasmic ***,syringic acid was significantly downregulated across all three TiO_(2)-NP-exposed *** obtain a comprehensive overview of the TiO_(2)-NP-induced expression changes,a co-regulated network of proteins and metabolites associated with TiO_(2)-NPs exposure was *** to TiO_(2)-NPs led to enrichment and alteration of pathways related to immune responses,including endocytosis,protein processing in the endoplasmic reticulum,and peroxisome proliferator-activated receptor(PPAR)*** conclusion,our findings indicate that exposure to TiO_(2)-NPs might disrupt glucose metabolism and induce immune responses,thus contributing to our understanding of the environmental impacts of nanomaterials and highlighting the need for further research and development of potential mitigation strategies.

Analysis and Characterization of the GABA Transaminase and Succinate Semialdehyde Dehydrogenase Genes in the Microalga Isochrysis zhanjiangensis in Response to Abiotic Stresses

Gamma-aminobutyric acid(GABA),widely existing in different organisms,is rapidly accumulated in plants in response to environmental *** main biosynthesis and degradation pathways of GABA constitute the GABA shunt,which is tied to the tricarboxylic acid(TCA)*** transaminase(GABA-T)and succinate semialdehyde dehydrogenase(SSADH)are two essential enzymes for the GABA degradation *** there are abundant studies on GABA shunt in higher plants at the physiological and genetic levels,research on its role in microalgae remains *** study aimed at exploring the function of GABA-T and SSADH genes in Isochrysis zhanjiangensis,an important diet microalga,under different *** cloned two GABA-T genes,IzGABA-T1 and IzGABA-T2,and one SSADH gene IzSSADH from Isochrysis zhanjiangensis and conducted heterologous expression *** results showed that the overexpression of IzGABA-T1 or IzGABA-T2 enhanced the survival rates of yeast transformants under heat or NaCl stress,while the overexpression of IzSSADH improved yeast tolerance to NaCl stress but had no obvious effect on heat ***,the results of quantitative real-time polymerase chain reaction(qPCR)showed that IzGABA-T1 transcription increased in the HT(salinity 25,35℃)and LS(salinity 15,25℃)*** 24 h,the IzGABA-T2 transcriptions increased in the HT,LS,and HS(salinity 35,25℃)groups,but their transcription levels decreased in all groups at 48 *** transcription increased in the LS *** results suggest that IzGABA-T1,IzGABA-T2,and IzSSADH are associated with temperature and salinity stresses and possess a certain preference for different stresses.

Effect of dietary supplement of inactivated Lactobacillus plantarum Ep-M 17 on growth performance,immune response,disease resistance,and intestinal microbiota in Penaeus vannamei

Our previous study found that feeding with Lactobacillus plantarum Ep-M17 could effectively affect the growth performance,immune response,and gut microbiota of Penaeus ***,high temperature and pressure during feed pelletizing is the main problem that can lead to a decrease in the activity of probiotics or cause their *** investigation needs to investigate whether inactivated Ep-M17 can exert similar effects as live ***,we evaluated the effects of inactivated *** Ep-M17 on growth performance,immune response,disease resistance,and gut microbiota in *** show that adding inactivated Ep-M17 to the feed also promoted body weight gain and increased relative immune protection in ***,histological examination revealed that the administration of inactivated Ep-M17 led to improvements in the density and distribution of microvilli in the intestines and enhancements in the abundance of B and R cells in the ***,the inactivated Ep-M17 supplementation resulted in increased activity levels of nutrient immune-related enzymes in both the shrimp hepatopancreas and ***,it stimulated the expression of Lvlec,PEN-3a,Crustin,LGBP,Lysozyme,and proPo genes in both the hepatopancreas and ***,the inactivated Ep-M17 also increased bacterial diversity in the gut of shrimp and promoted the abundance of specific flora,facilitating the host organism’s metabolism and immunity to improve the disease resistance of ***,supplementation of inactivated *** Ep-M17 in shrimp diets can exert similar effects as live *** Ep-M17 effectively improving growth performance,gut microbiota,immune response,and disease resistance in ***.

Litopenaeus vannamei is the most extensively cultured shrimp species globally,recognized for its scale,production,and economic ***,its aquaculture is plagued by frequent disease outbreaks,resulting in rapid and massive *** research often lags behind the emergence of new diseases,leaving the causal agents of some shrimp diseases unidentified and leading to nomenclature based on symptomatic presentations,especially in cases involving co-and polymicrobial *** data on shrimp disease statuses remain *** this review,we summarize current knowledge on shrimp diseases and their effects on the gut ***,we also propose a workflow integrating primary colonizers,“driver”taxa in gut networks from healthy to diseased states,disease-discriminatory taxa,and virulence genes to identify potential polymicrobial *** examine both abiotic and biotic factors(e.g.,external and internal sources and specific-disease effects)that influence shrimp gut microbiota,with an emphasis on the“holobiome”concept and common features of gut microbiota response to diverse *** excluding the effects of confounding factors,we provide a diagnosis model for quantitatively predicting shrimp disease incidence using disease common-discriminatory taxa,irrespective of the causal *** to the conservation of functional genes used in designing specific primers,we propose a practical strategy applying qPCR-assayed abundances of disease common-discriminatory functional *** review updates the roles of the gut microbiota in exploring shrimp etiology,polymicrobial pathogens,and disease incidence,offering a refined perspective for advancing shrimp aquaculture health management.

Lysine-specific demethylase 4 A(KDM4A,also named JMJD2A,KIA0677,or JHDM3A)is a demethylase that can remove methyl groups from histones H3K9me2/3,H3K36me2/3,and H1.4K26me2/*** evidence suggests that KDM4A is not only involved in body homeostasis(such as cell proliferation,migration and differentiation,and tissue development)but also associated with multiple human diseases,especially ***,an increasing number of studies have shown that pharmacological inhibition of KDM4A significantly attenuates tumor progression in vitro and in vivo in a range of solid tumors and acute myeloid *** there are several reviews on the roles of the KDM4 subfamily in cancer development and therapy,all of them only briefly introduce the roles of KDM4A in cancer without systematically summarizing the specific mechanisms of KDM4A in various physiological and pathological processes,especially in tumorigenesis,which greatly limits advances in the understanding of the roles of KDM4A in a variety of cancers,discovering targeted selective KDM4A inhibitors,and exploring the adaptive profiles of KDM4A ***,we present the structure and functions of KDM4A,simply outline the functions of KDM4A in homeostasis and non-cancer diseases,summarize the role of KDM4A and its distinct target genes in the development of a variety of cancers,systematically classify KDM4A inhibitors,summarize the difficulties encountered in the research of KDM4A and the discovery of related drugs,and provide the corresponding solutions,which would contribute to understanding the recent research trends on KDM4A and advancing the progression of KDM4A as a drug target in cancer therapy.

Ulcerative colitis(UC)is a recurrent inflammatory bowel disease that imposes a severe burden on families and *** recent years,exploiting the potential of marine bioactive peptides for the treatment of diseases has become a topic of intense research *** study revealed the mechanism underlying the protective effect of the dominant polypeptide PKKVV(Pro-Lys-Lys-Val-Val)of Rhopilema esculentum cnidoblasts against DSS-induced UC through a combined analysis of the metagenome and serum ***,the polypeptide composition of *** cnidoblasts was determined by matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF/TOF-MS).Molecular docking showed that the dominant peptide PKKVV could bind better with tumor necrosis factor-α(TNF-α)than the original *** animal experiments suggested that PKKVV could modulate disorganized gut microorganisms in mice with UC;affect serum metabolites through the arachidonic acid,glycerophospholipid and linoleic acid metabolism pathways;and further alleviate UC *** study provides a reference for the comprehensive development of marine bioactive substances and nonpharmaceutical treatments for UC.

The demand for high-data-rate underwater acoustic communications(UACs)in marine development is increasing;however,severe multipaths make demodulation a *** decision feedback equalizer(DFE)is one of the most popular equalizers in UAC;however,it is not the optimal *** maximum likelihood sequence estimation(MLSE)is the optimal algorithm,its complexity increases exponentially with the number of channel taps,making it challenging to apply to ***,this paper proposes a complexity-reduced MLSE to improve the bit error rate(BER)performance in multipath *** the proposed algorithm,the original channel is first shortened using a channel-shortening method,and several dominant channel taps are selected for ***,sphere decoding(SD)is performed in the following *** are applied to eliminate inter-symbol interference caused by weak channel *** simulation and sea experiment demonstrate the superiority of the proposed *** simulation results show that channel shortening combined with SD can drastically reduce computational complexity,and iterative SD performs better than DFE based on recursive least squares(RLS-DFE),DFE based on improved proportionate normalized least mean squares(IPNLMS-DFE),and channel estimation-based DFE(CE-DFE).Moreover,the sea experimental results at Zhairuoshan Island in Zhoushan show that the proposed receiver scheme has improved BER performance over RLSDFE,IPNLMS-DFE,and *** with the RLS-DFE,the BER,after five iterations,is reduced from 0.0076 to 0.0037 in the 8–12 k Hz band and from 0.1516 to 0.1145 in the 13–17 k Hz band at a distance of 2000 ***,the proposed algorithm makes it possible to apply MLSE in UAC in practical scenarios.

Development of underwater electric manipulator based on interventional autonomous underwater vehicle (AUV)

In applications such as marine rescue,marine science,archaeology,and offshore industries,autonomous underwater vehicles(AUVs)are frequently used for survey missions and monitoring tasks,with most operations being performed by manned submersibles or remotely operated vehicles(ROVs)equipped with robotic arms,as they can be operated remotely for days without ***,they require expensive marine vessels and specialist pilots to operate *** exploring oceans are no longer satisfied with the use of manned submersibles and *** is a growing desire for seabed exploration to be performed using smarter,more flexible,and automated *** improving the field operation and intervention capability of AUVs,large-scale and long-range seafloor exploration and sampling can be performed without the support of a mother ship,making it a more effective,economical,convenient,and rapid means of seafloor exploration and sampling operations,and playing a critical role in marine resource *** this study,we explored the integration technology of underwater electric robotic arms and AUVs and designed a new set of electric manipulators suitable for water depths greater than 500 *** reliability of the key components was analyzed by finite element analysis and,based on the theory of robot kinematics and dynamics,simulations were performed to verify the reliability of the key *** were conducted on land and underwater,trajectory tracking experiments were completed,and the experimental data in air and water were compared and ***,the objectives for further research on the autonomous control of the manipulator underwater were proposed.
