Coated sodium butyrate ameliorates high‑energy and low‑protein diet induced hepatic dysfunction via modulating mitochondrial dynamics, autophagy and apoptosis in laying hens

Fermented defatted rice bran-derived gut Clostridium butyricum improved the gut barrier function by regulating HIF-1 signaling pathway

Emerging evidence of the beneficial effects of defatted rice bran (DFRB) on gut health has advanced the development of fermented defatted rice bran as a potential functional food. However， less is known about its effects and underlying mechanisms on gut health. In this study， a mouse model together with fecal microbiota transplantation (FMT) was utilized to study the effects and mechanisms of fermented DFRB (FR) on gut barrier function. We found that FR improved the intestinal morphology， gut tight junction proteins， mucin， antimicrobial peptides， and IL-22 and promoted the gut Clostridium butyricum and butyrate. Notably， correlation analysis indicated gut C. butyricum and butyrate were two FR-induced effectors that improved gut health. FMT results suggested that C. butyricum， butyrate， and fecal microbiota from the FR group all reduced PHD2 expression by activating PPAR-γ in the mouse colon. This decrease in gut PHD2 subsequently upregulated the HIF-1α expression， which in turn increased the expression of its targeted downstream tight junction proteins， mucin and antimicrobial peptides， and colonic IL-22 secretion. Overall， FR-derived C. butyricum and butyrate might improve gut barrier function through the HIF-1 signaling pathway， which provides a reference for the application of fermented DFRB as a potential functional food for improving of gut barrier function.

The role of vitamin E in polyunsaturated fatty acid synthesis and alleviating endoplasmic reticulum stress in sub-adult grass carp(Ctenopharyngodon idella)

Vitamin E(VE) is an essential lipid-soluble vitamin that improves the fish flesh quality. However, the underlying molecular mechanisms remain unclear. This study aimed to investigate the effects of VE on growth performance and flesh quality in sub-adult grass carp(Ctenopharyngodon idella). A total of 450fish(713.53 ± 1.50 g) were randomly divided into six treatment groups(three replicates per treatment)and fed for nine weeks with different experimental diets(dietary lipid 47.8 g/kg) that contained different levels of VE(5.44, 52.07, 96.85, 141.71, 185.66, and 230.12 mg/kg diet, supplemented as dl-a-tocopherol acetate). Notably, the treatment groups that were fed with dietary VE ranging from 52.07 to230.12 mg/kg diet showed improvement in the percent weight gain, special growth rate, and feed efficiency of grass carp. Moreover, the treatment groups supplemented with dietary VE level of 141.71,185.66, and 230.12 mg/kg diet showed enhancement in crude protein, lipid, and a-tocopherol contents in the muscle, and the dietary levels of VE ranging from 52.07 to 141.71 mg/kg diet improved muscle pH24h and shear force but reduced muscle cooking loss in grass carp. Furthermore, appropriate levels of VE(52.07 to 96.85 mg/kg diet) increased the muscle polyunsaturated fatty acid content in grass carp. Dietary VE also increased the mRNA levels of fatty acid synthesis-related genes, including fas, scd-1, fad,elovl, srebp1, pparγ, and lxra, and up-regulated the expression of SREBP-1 protein. However, dietary VE decreased the expression of fatty acid decomposition-related genes, including hsl, cpt1, acox1, and ppara,and endoplasmic reticulum stress-related genes, including perk, ire1, atf6, eif2a, atf4, xbp1, chop, and grp78, and down-regulated the expression of p-PERK, p-IRE1, ATF6, and GRP78 proteins. Background Ochratoxin A(OTA),a globally abundant and extremely hazardous pollutant,is a significant source of contamination in aquafeeds and is responsible for severe food *** developmental toxicity of OTA and the potential relieving strategy of natural products remain *** study screened the substance curcumin(Cur),which had the best effect in alleviating OTA inhibition of myoblast proliferation,from 96 natural products and investigated its effect and mechanism in reducing OTA myotoxicity in vivo and in vitro.Methods A total of 720 healthy juvenile grass carp,with an initial average body weight of 11.06±0.05 g,were ran-domly assigned into 4 groups:the control group(without OTA and Cur),1.2 mg/kg OTA group,400 mg/kg Cur group,and 1.2 mg/kg OTA+400 mg/kg Cur *** treatment consisted of 3 replicates(180 fish)for 60 d.Results Firstly,we cultured,purified,and identified myoblasts using the tissue block culture *** pre-liminary screening and re-screening of 96 substances,we examined cell proliferation-related indicators such as cell viability and ultimately found that Cur had the best ***,Cur could alleviate OTA-inhibited myoblast differentiation and myofibrillar development-related proteins(MyoG and MYHC)in vivo and in vitro and improve the growth performance of grass ***,Cur could also promote the expression of OTA-inhibited protein syn-thesis-related proteins(S6K1 and TOR),which was related to the activation of the AKT/TOR signaling ***,Cur could downregulate the expression of OTA-enhanced protein degradation-related genes(murf1,foxo3a,and ub),which was related to the inhibition of the FoxO3a signaling pathway.Conclusions In summary,our data demonstrated the effectiveness of Cur in alleviating OTA myotoxicity in vivo and in *** study confirms the rapidity,feasibility,and effectiveness of establishing a natural product screening method targeting myoblasts to alleviate fungal toxin toxicity.

Background Rumen bacterial groups can affect growth performance,such as average daily gain(ADG),feed intake,and *** study aimed to investigate the inter-relationship of rumen bacterial composition,rumen fermentation indicators,serum indicators,and growth performance of Holstein heifer calves with different *** calves were chosen from a trail with 60 calves and divided into higher ADG(HADG,high pre-and post-weaning ADG,n=6)and lower ADG(LADG,low pre-and post-weaning ADG,n=6)groups to investigate differences in bacterial composition and functions and host *** During the preweaning period,the relative abundances of propionate producers,including g_norank_f_Butyricicoccaceae,g_Pyramidobacter,and g_norank_f_norank_o_Clostridia_vadin BB60_group,were higher in HADG calves(LDA>2,P<0.05).Enrichment of these bacteria resulted in increased levels of propionate,a gluconeogenic precursor,in preweaning HADG calves(adjusted P<0.05),which consequently raised serum glucose concentrations(adjusted P<0.05).In contrast,the relative abundances of rumen bacteria in post-weaning HADG calves did not exert this ***,no significant differences were observed in rumen fermentation parameters and serum indices between the two *** The findings of this study revealed that the preweaning period is the window of opportunity for rumen bacteria to regulate the ADG of calves.
