The receptor-like cytoplasmic kinase RIPK regulates broad-spectrum ROS signaling in multiple layers of plant immune system

production of reactive oxygen species(ROS)via the activity of respiratory burst oxidase homologs(RBOHs)plays a vital role in multiple layers of the plant immune system,including pathogen-associated molecular pattern-triggered immunity(PTI),damage-associated molecular pattern-triggered immunity(DTI),effector-triggered immunity(ETI),and systemic acquired resistance(SAR).It is generally established that RBOHD is activated by different receptor-like cytoplasmic kinases(RLCKs)in response to various immune *** this study,we showed that RPM1-INDUCED PROTEIN KINASE(RIPK),an RLCK VII subfamily member,contributes to ROS production in multiple layers of plant immune *** ripk mutants showed reduced ROS production in response to treatment with all examined immune elicitors that trigger PTI,DTI,ETI,and *** found that RIPK can directly phosphorylate the N-terminal region of RBOHD in vitro,and the levels of phosphorylated S343/S347 residues of RBOHD are sigfniciantly lower in ripk mutants compared with the wild type upon treatment with all tested immune *** further demonstrated that phosphorylation of RIPK is required for its function in regulating RBOHD-mediated ROS *** to rbohd,ripk mutants showed reduced stomatal closure and impaired SAR,and were susceptible to the necrotrophic bacterium Pectobacterium ***,our results indicate that RIPK regulates broad-spectrum RBOHD-mediated ROS signaling during PTI,DTI,ETI,and SAR,leading to subsequent RBOHD-dependent immune responses.

The plant hormone jasmonate(JA)regulates plant growth and immunity by orchestrating a genome-wide transcriptional *** the resting stage,JASMONATE-ZIM DOMAIN(JAZ)proteins act as main repressors to regulate the expression of JA-responsive genes in the JA signaling ***,the mechanisms underlying de-repression of JA-responsive genes in response to JA treatment remain ***,we report two nuclear factor Y transcription factors NF-YB2 and NF-YB3(thereafter YB2 and YB3)play key roles in such de-repression in ***2 and YB3 function redundantly and positively regulate plant resistance against the necrotrophic pathogen Botrytis cinerea,which are specially required for transcriptional activation of a set of JA-responsive genes following ***,YB2 and YB3 modulated their expression through direct occupancy and interaction with histone demethylase Ref6 to remove repressive histone ***,YB2 and YB3 physically interacted with JAZ repressors and negatively modulated their abundance,which in turn attenuated the inhibition of JAZ proteins on the transcription of JA-responsive genes,thereby activating JA response and promoting disease ***,our study reveals the positive regulators of YB2 and YB3 in JA signaling by positively regulating transcription of JA-responsive genes and negatively modulating the abundance of JAZ proteins.

Microbes on fresh apples are closely associated with fruit disease,preservation,and quality *** into the microbial commu-nities on apples from different producing regions could reveal the microbial specificity and help disease prevention and quality *** this paper,the apple surface microbes of 44 samples from two main Chinese apple-producing regions,Bohai Bay(BHB)and the Loess Plateau(LP),were investigated by sequencing fungal internal transcribed spacer and bacterial 16s rRNA hypervariable *** and LP apples con-tained significantly different bacterial and fungal *** apples had a higher fungal diversity than LP apples.A total of 102 different fungal and bacterial taxonomies were obtained between apples from the two regions,in which 24 genera were *** apples had higher phytopathogenic fungal genera,such as Tilletiopsis,Acremonium,Candida,and Phoma,indicating the higher phytopathogenic risks of apples from the humid climate of the BHB *** apples contained more bacterial genera identified as gut microbes,indicating the potential risks of contaminating apples with foodborne pathogens in the arid environment of the LP This study highlighted the environment-oriented mi-crobial specificity on apples from two main apple-producing regions,and provided a basis for further investigation.

Oxidation of diclofenac by birnessite: Identification of products and proposed transformation pathway

Diclofenac(DCF), a widely used non-steroidal anti-inflammatory, reacted readily with birnessite under mild conditions, and the pseudo first order kinetic constants achieved 8.84 × 10^-2 hr^-1. Five products of DCF including an iminoquinone product(2,5-iminoquinone-diclofenac) and four dimer products were observed and identified by tandem mass spectrometry during the reaction. Meanwhile, 2,5-iminoquinone-diclofenac was identified to be the major product, accounting for 83.09% of the transformed DCF. According to the results of spectroscopic Mn(III) trapping experiments and X-ray Photoelectron Spectroscopy, Mn(IV) contained in birnessite solid was consumed and mainly converted into Mn(III) during reaction process, which proved that the removal of DCF by birnessite was through oxidation. Based on the identified products of DCF and the changes of Mn valence state in birnessite solid, a tentative transformation pathway of DCF was proposed.

Phytosterol content and the campesterol:sitosterol ratio influence cotton fiber development: role of phytosterols in cell elongation

Phytosterols play an important role in plant growth and development, including cell division, cell elongation, embryogenesis, cellulose biosynthesis, and cell wall formation. Cotton fiber, which undergoes synchronous cell elongation and a large amount of cellulose synthesis, is an ideal model for the study of plant cell elongation and cell wall biogenesis. The role of phytosterols in fiber growth was investigated by treating the fibers with tfidemorph, a sterol biosynthetic inhibitor. The inhibition of phy- tosterol biosynthesis resulted in an apparent suppression of fiber elongation in vitro or in planta. The determination of phy- tosterol quantity indicated that sitosterol and campesterol were the major phytosterols in cotton fibers; moreover, higher con- centrations of these phytosterols were observed during the period of rapid elongation of fibers. Furthermore, the decrease and increase in campesterol：sitosterol ratio was associated with the increase and decease in speed of elongation, respectively, dur- ing the elongation stage. The increase in the ratio was associated with the transition from cell elongation to secondary cell wall synthesis. In addition, a number of phytosterol biosynthetic genes were down-regulated in the short fibers of ligon lintless-1 mutant, compared to its near-isogenic wild-type TM-1. These results demonstrated that phytosterols play a crucial role in cot- ton fiber development, and particularly in fiber elongation.

Imprinting, methylation, and expression characterization of the maize ETHYLENE-INSENSITIVE 2-like gene

The endosperm plays essential roles in embryogenesis and seed germination and provides abundant resources for human food and industrial products. Identification of genes regulating the development of the endosperm and elucidation of their functions is of great importance for maize genetics and breeding. This study showed that the genespecific imprinted gene, ETHYLENE-INSENSITIVE 2-like(EIN2-like), is maternally expressed in both endosperm and embryo. The maternally expressed pattern was maintained throughout later seed developmental stages. Bisulfite sequencing using DNA obtained from hybrid endosperm tissues showed that the upstream regions of the alleles of EIN2-like were highly methylated at symmetrical sites(CG and CHG). A differentially methylated region in the upstream part of the maternal allele of EIN2-like was identified and found to be hypomethylated. Expression analysis showed that EIN2-like was highly expressed in the maize endosperm as well as at different stages of cell differentiation(8–12 days after pollination) in the hybrid endosperm. These results suggest that the maternally expressed gene EIN2-like may play crucial roles in the regulation of seed development.

Anomalous quality factor evolution of induced transparency in a virtualized coupled-oscillator system

The phenomenon of induced transparency, characterized by a high quality(Q) factor, holds significant importance in the fields of high-precision sensing, energy storage and wave manipulation. Conventional transparency systems are often constrained by the limited experimentally accessible parameter space and the lack of independent control over intrinsic parameters, thereby hindering advanced research on high-Q effects. Here we propose a virtualized coupled-oscillator system that enables reconfigurable induced transparency in the reflection spectrum. By virtually incorporating a pair of coupled oscillators into an individual acoustic meta-atom, we achieve the manipulation of induced transparency in a decoupled, wide-ranging, and elaborate manner. Exploiting the diverse evolutions across the intrinsic parameter space, we show experimentally a counter-intuitive increase in the Q-factor of the transparency window, which is attributed to the enhanced dissipation of the bright ***, a constant group delay at the center frequency is observed accompanying this evolution. Our work demonstrates an intriguing platform as well as a novel strategy to engineer high-Q induced transparency phenomenon, and paves the way towards applications in programmable acoustic sensors, filters and slow-wave devices.

Effects of dehydration speed on the metabolism of membrane lipids and its relation to the browning of the Thompson seedless grape

Xinjiang is the main producing area of raisins and the largest green raisins production base in *** browning of Thompson seedless grape raisin is extremely serious during the drying process,and has become the key issue in the development of Xinjiang raisin *** studies have shown that dehydration speed has a great impact on the browning of Thompson seedless grape,but few relevant mechanisms have been ***,we demonstrate the effect of dehydration speed on lipid metabolism and its relation to the browning of the Thompson seedless grape during *** to slow dehydration treatment,rapid dehydration treatment of the Thompson seedless grape exhibited a lower degree of browning and activities of lipoxygenase(LOX),a higher index of unsaturated fatty acids and degree of unsaturated fatty ***,the Thompson seedless grape treated with rapid dehydration resulted in a lower rate of superoxide anion production,hydrogen peroxide content,membrane permeability,and malondialdehyde *** findings demonstrate that rapid dehydration inhibiting the browning of Thompson seedless grapes might be due to the inhibiting activities of LOX and the lower accumulation of reactive oxygen *** activities can inhibit lipid peroxidation and slow the decomposition of unsaturated fatty acid in the membrane in Thompson seedless grapes,protecting the cellular membrane structural integrity,which may result in less contact of polyphenol oxidase with phenolic substrates and less enzymatic browning during *** results provide a theoretical basis for the application of rapid dehydration in drying Thompson seedless grapes.
