Multimodal Identification by Transcriptomics and Multiscale Bioassays of Active Components in Xuanfeibaidu Formula to Suppress Macrophage-Mediated Immune Response

Xuanfeibaidu Formula (XFBD) is a Chinese medicine used in the clinical treatment of coronavirus disease 2019 (COVID-19) patients. Although XFBD has exhibited significant therapeutic efficacy in clinical practice, its underlying pharmacological mechanism remains unclear. Here, we combine a comprehensive research approach that includes network pharmacology, transcriptomics, and bioassays in multiple model systems to investigate the pharmacological mechanism of XFBD and its bioactive substances. High-resolution mass spectrometry was combined with molecular networking to profile the major active substances in XFBD. A total of 104 compounds were identified or tentatively characterized, including flavonoids, terpenes, carboxylic acids, and other types of constituents. Based on the chemical composition of XFBD, a network pharmacology-based analysis identified inflammation-related pathways as primary targets. Thus, we examined the anti-inflammation activity of XFBD in a lipopolysaccharide-induced acute inflammation mice model. XFBD significantly alleviated pulmonary inflammation and decreased the level of serum proinflammatory cytokines. Transcriptomic profiling suggested that genes related to macrophage function were differently expressed after XFBD treatment. Consequently, the effects of XFBD on macrophage activation and mobilization were investigated in a macrophage cell line and a zebrafish wounding model. XFBD exerts strong inhibitory effects on both macrophage activation and migration. Moreover, through multimodal screening, we further identified the major components and compounds from the different herbs of XFBD that mediate its anti-inflammation function. Active components from XFBD, including Polygoni cuspidati Rhizoma, Phragmitis Rhizoma, and Citri grandis Exocarpium rubrum, were then found to strongly downregulate macrophage activation, and polydatin, isoliquiritin, and acteoside were identified as active compounds. Identification of crystal orientation for turbine blades with anisotropy materials

A novel approach to identify the crystal orientation of turbine blades with anisotropy materials is proposed. Based on enhanced mode basis, with the main advantages of its efficiency,accuracy and general applicability, the blade vibration mode of each order is linearly constructed by several specified mode shapes, which are obtained from the considered turbine blade with specified crystal orientations correspondingly. Then, a surrogate model based on Kriging method is introduced for constructing the condensed perturbed matrix of stiffness in order to improve the efficiency even further. The constructed surrogate model allows to perform the modal analysis of turbine blades with arbitrary crystal orientations in higher efficiency, due to the fact that the elements of condensed perturbed matrix of stiffness are considered in construction of the surrogate model rather than concerning the perturbation of all the elements of the initial stiffness matrix for the blade. Genetic algorithm is finally employed to optimize the defined fitness functions in order to identify the crystal orientation angles of turbine blades. Several corresponding examples demonstrated the accuracy, efficiency and general applicability of the proposed method.

近年来,对于可注射水凝胶用于内部组织损伤修复的研究取得了一定进展,但将此类水凝胶用于胃穿孔治疗时,依然面临强酸性胃液及胃壁的剧烈形变带来的严峻问题.鉴于此,我们提出了一种基于纳米离子储库(INR)的双网络水凝胶策略,该水凝胶具有优异的黏附和机械性能,可以简便地涂覆于穿孔部位,在封堵穿孔的同时促进组织修复.结果显示,在蓝光条件下,聚丙烯酰胺(PAM)可以于5 s内在胃组织上交联形成第一重网络,并通过机械互锁增强黏附性能;此外,纳米羟基磷灰石作为纳米离子储库可在酸性环境下释放Ca^(2+),与海藻酸钠(SA)形成第二重网络,抑制水凝胶在胃液中的溶胀.同时,通过在体系中引入碳二亚胺/N-羟基琥珀酰亚胺(EDC/NHS),可以促进水凝胶-组织界面上酯酰胺共价键的形成,从而进一步增强黏附力.因此我们认为,这种通过纳米离子储库策略获得的双网络水凝胶有望用于解决胃穿孔乃至其他类似的生物医学问题.

Bismuth-content-dependent polarized Raman spectrum of InPBi alloy

We systematically investigate the optical properties of the InP1-xBix ternary alloys with 0≤x≤2.46%,by using high resolution polarized Raman scattering measurement.Both InP-like and InBi-like optical vibration modes（LO） are identified in all the samples,suggesting that most of the Bi-atoms are incorporated into the lattice sites to substitute Patoms.And the intensity of the InBi-like Raman mode is positively proportional to the Bi-content.Linear red-shift of the InP-like longitudinal optical vibration mode is observed to be 1.1 cm-1/Bi%,while that of the InP-like optical vibration overtone（2LO） is nearly doubled.In addition,through comparing the（XX） and Z（XY） Raman spectra,longitudinaloptical-plasmon-coupled（LOPC） modes are identified in all the samples,and their intensities are found to be proportional to the electron concentrations.

A standardized dataset of built-up areas of China’s cities with populations over 300,000 for the period 1990-2015

China’s urbanization has attracted a lot of attention due to its unprecedented pace and intensity in terms of land,population,and economic impact.However,due to the lack of consistent and harmonized data,little is known about the patterns and dynamics of the interaction between these different aspects over the past few decades.Along with the implementation of the 2030 Agenda for Sustainable Development,a standardized dataset for assessing the sustainability of urbanization in China is needed.In this paper,we used remote sensing data from multiple sources(time-series of Landsat and Sentinel images)to map the impervious surface area(ISA)at five-year intervals from 1990 to 2015 and then converted the results into a standardized dataset of the built-up area for 433 Chinese cities with 300,000 inhabitants or more.This dataset was produced following the well-established rules adopted by the United Nations(UN).Validation of the ISA maps in urban areas based on the visual interpretation of Google Earth images showed that the average overall accuracy(OA),producer’s accuracy(PA)and user’s accuracy(UA)were 91.24%,92.58%and 89.65%,respec-tively.Comparisons with other existing urban built-up area datasets derived from the National Bureau of Statistics of China,the World Bank and UN-habitat indicated that our dataset,namely the stan-dardized urban built-up area dataset for China(SUBAD-China),provides an improved description of the spatiotemporal character-istics of the urbanization process and is especially applicable to a combined analysis of the spatial and socio-economic domains in urban areas.Potential applications of this dataset include combin-ing the spatial expansion and demographic information provided by UN to calculate sustainable development indicators such as SDG 11.3.1.The dataset could also be used in other multidimensional syntheses related to the study of urbanization in China.

A snake addressing scheme for phase change memory testing

Phase change memory(PCM) is one of the most promising candidates for next generation nonvolatile memory. However, PCM suffers from a variety of faults due to its special device structure and operation mechanism. A snake addressing scheme is introduced into the test algorithms of PCM to reduce the test time and excite proximity disturb faults more effectively. The March test algorithm with the proposed snake addressing scheme is less complex than most traditional test algorithms. In addition to conventional faults, it is capable of covering disturb and parasitic faults. Moreover, when incorporated with the sneak path testing method, it is able to test the read fault, read recovery fault, incomplete program fault 0, and false write fault.

Evaluation and Optimization of Electrode Configuration of Multi-Channel Corona Discharge Plasma for Dye-Containing Wastewater Treatment

A discharge plasma reactor with a point-to-plane structure was widely studied experimentally in wastewater treatment.In order to improve the utilization efficiency of active species and the energy efficiency of this kind of discharge plasma reactor during wastewater treatment,the electrode configuration of the point-to-plane corona discharge reactor was studied by evaluating the effects of discharge spacing and adjacent point distance on discharge power and discharge energy density,and then dye-containing wastewater decoloration experiments were conducted on the basis of the optimum electrode configuration.The experimental results of the discharge characteristics showed that high discharge power and discharge energy density were achieved when the ratio of discharge spacing to adjacent point distance（d/s） was 0.5.Reactive Brilliant Blue（RBB） wastewater treatment experiments presented that the highest RBB decoloration efficiency was observed at d/s of 0.5,which was consistent with the result obtained in the discharge characteristics experiments.In addition,the biodegradability of RBB wastewater was enhanced greatly after discharge plasma treatment under the optimum electrode configuration.RBB degradation processes were analyzed by GC-MS and IC,and the possible mechanism for RBB decoloration was also discussed.

Spatially explicit modeling techniques recently emerged as an alternative to monitor land use changes. This study adopted the well-known CLUE-S(Conversion of Land Use and its Effects at Small regional extent) model to analyze the spatio-temporal land use changes in a hot-spot in Northeast China(NEC). In total,13 driving factors were selected to statistically analyze the spatial relationships between biophysical and socioeconomic factors and individual land use types. These relationships were then used to simulate land use dynamic changes during 1980–2010 at a 1 km spatial resolution,and to capture the overall land use change patterns. The obtained results indicate that increases in cropland area in NEC were mainly distributed in the Sanjiang Plain and the Songnen Plain during 1980–2000,with a small reduction between 2000 and 2010. An opposite pattern was identified for changes in forest areas. Forest decreases were mainly distributed in the Khingan Mountains and the Changbai Mountains between 1980 and 2000,with a slight increase during 2000–2010. The urban areas have expanded to occupy surrounding croplands and grasslands,particularly after the year 2000. More attention is needed on the newly gained croplands,which have largely replaced wetlands in the Sanjiang Plain over the last decade. Land use change patterns identified here should be considered in future policy making so as to strengthen local eco-environmental security.

Trends of Yield and Soil Fertility in a Long-Term Wheat-Maize System

The sustainability of the wheat-maize rotation is important to China＇s food security. Intensive cropping without recycling crop residues or other organic inputs results in the loss of soil organic matter （SOM） and nutrients, and is assumed to be non- sustainable. We evaluated the effects of nine different treatments on yields, nitrogen use efficiency, P and K balances, and soil fertility in a wheat-maize rotation system （1991-2010） on silt clay loam in Shaanxi, China. The treatments involved the application of recommended dose of nitrogen （N）, nitrogen and phosphorus （NP）, nitrogen and potassium （NK）, phosphorus and potassium （PK）, combined NPK, wheat or maize straw （S） with NPK （SNPK）, or dairy manure （M） with NPK （M1NPK and M2NPK）, along with an un-treated control treatment （CK）. The mean yields of wheat and maize ranged from 992 and 2 235 kg ha-1 under CK to 5 962 and 6 894 kg ha-1 under M2NPK treatment, respectively. Treatments in which either N or P was omitted （N, NK and PK） gave significantly lower crop yields than those in which both were applied. The crop yields obtained under NP, NPK and SNPK treatments were statistically identical, as were those obtained under SNPK and MNPK. However, M2NPK gave a significant higher wheat yield than NP, and MNPK gave significant higher maize yield than both NP and NPK. Wheat yields increased significantly （by 86 to 155 kg ha-1 yr-1） in treatments where NP was applied, but maize yields did not. In general, the nitrogen use efficiency of wheat was the highest under the NP and NPK treatments; for maize, it was the highest under MNPK treatment. The P balance was highly positive under MNPK treatment, increasing by 136 to 213 kg ha-1 annually. While the K balance was negative in most treatments, ranging from 31 to 217 kg ha^-1 yr^-1, levels of soil available K remained unchanged or increased over the 20 yr. Elaborate regulation of gene expression is required for plants to maintain normal growth,development,and reproduction.MicroRNAs(miRNAs)and transcription factors are key players that control gene expression in plant regulatory networks.The TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR(TCP)family comprises plantspecific transcription factors that contain a conserved TCP domain of 59 amino acids.Some members of this family are targeted by miR319,one of the most ancient and evolutionarily conserved miRNAs in plants.Accumulating evidence has revealed that miR319-regulated TCP(MRTCP)genes participate extensively in plant development and responses to environmental stress.In this review,the structural characteristics and classifications of TCP transcription factors and the regulatory relationships between TCP transcription factors and miRNAs are introduced.Current knowledge of the regulatory functions of MRTCP genes in multiple biological pathways including leaf development,vascular formation,flowering,hormone signaling,and response to environmental stresses such as cold,salt,and drought is summarized.This review will be beneficial for understanding the roles of the MRTCP-mediated regulatory network and its molecular mechanisms in plant development and stress response,and provides a theoretical basis for plant genetic improvement.
