Resolving the developmental mechanisms of cardiac microthrombosis of SARS-CoV-2 based on single-cell transcriptome analysis

The coronavirus disease 2019（COVID-19） outbreak caused by the severe acute respiratory syndrome coronavirus-2（SARS-CoV-2） developed into a global health emergency. Systemic microthrombus caused by SARS-CoV-2 infection is a common complication in patients with COVID-19. Cardiac microthrombosis as a complication of SARS-CoV-2 infection is the primary cause of cardiac injury and death in patietns with severe COVID-19. In this study, we performed single-cell sequencing analysis of the right ventricular free wall tissue from healthy donors,patients who died during the hypercoagulable period of characteristic coagulation abnormality（CAC）, and patients who died during the fibrinolytic period of CAC. We collected 61,187 cells enriched in 24 immune cell subsets and 13 cardiac-resident cell subsets. We found that in the course of SARS-CoV-2 infected heart microthrombus, MYO1EhighRASGEF1Bhighmonocyte-derived macrophages promoted hyperactivation of the immune system and initiated the extrinsic coagulation pathway by activating chemokines CCL3, CCL5. This series of events is the main cause of cardiac microthrombi following SARS-CoV-2 infection. In a SARS-CoV-2 infected heart microthrombus, excessive immune activation is accompanied by an increase in cellular iron content, which in turn promotes oxidative stress and intensifies intercellular competition. This induces cells to alter their metabolic environment, resulting in increased sugar uptake via the glycosaminoglycan synthesis pathway. In addition, high levels of reactive oxygen species generated by elevated iron levels promote increased endogenous malondialdehyde synthesis in a subpopulation of cardiac endothelial cells. This exacerbates endothelial cell dysfunction and exacerbates the coagulopathy process.

Mammalian hair, as a key feature related to endothermy, physical protection, and camouflage, has a long geohistorical record that dates back even to the Mesozoic. However, the limited information on the taphonomy of hair hinders the understanding of its evolutionary significance in deep time. To address this, we conducted taphonomic experiments to simulate the transformation process of black hair. The changes in the morphological structure, microscopic characteristics, and chemical composition of hair during the simulated fossilization process were documented by scanning electron microscopy, Fourier transform infrared(FTIR) spectroscopy, and Raman spectroscopy. Our results revealed that damage to the morphology of hair began at a mild temperature(150–175°C), and hair transformed into a fluid-like mass at a relatively high temperature(200°C). In addition, the morphological structure of melanosomes had relatively greater preservation potential than keratins did, and the microscopic features of melanosomes were persistently stable. The Raman and FTIR spectra indicated that the characteristic melanin and keratin peaks changed as the temperature increased. Although the chemical signals were not lost completely, the variations in the characteristic amide bands in the FTIR spectra indicate that the keratins severely degraded at high temperatures(>200°C), whereas the characteristic melanin bands in the Raman spectra suggested that melanin signals were still recognizable, even at high temperatures(>200°C). These results were confirmed by principal component analysis(PCA) and principal component analysis linear discriminant analysis(PCA–LDA). In addition, the Raman signals of all the samples in the simulated experiments were distinguished from the signals of common geological carbonaceous materials(humic acid), which suggested that Raman spectroscopy can be used to detect melanin in fossils.

The Guanzhong Basin， located in Shaanxi Province， China， has abundant geothermal resources in its hinterland. Focusing on typical thermal reservoir structural units， namely the Xianli fault-step (XL) and Xi'an depression (XA)， this study reveals the genesis of geothermal water by combining hydrochemistry and environmental isotopes. The geothermal waters of XL uniformly correspond to the Na-Cl type with high contents of major ions， while the geothermal water of XA has complex hydrochemical types. The quartz geothermometer (with maximum steam loss) and Na-K-Ca geothermometer are recommended for XL and the quartz geothermometer (with no and maximum steam loss) and Na-K geothermometer for XA. Their temperature ranges are 99-159℃ and 93-146℃， respectively. The circulation depth of geofluids in XL and XA range from 2579 to 4766 m and from 2541 to 4330 m， with mean apparent ages of 21543 and 20345 yr B.P.， respectively. The geothermal water in XL originates from a mixture of sedimentary water and ancient infiltration water， and its formation mode was the vertical heat conduction with cooling process. In contrast， the geothermal water in XA originates from the dual process of cooling and mixing， with the process being dominated by ancient infiltration water and supplementation by modern infiltration water.

The response of lake environments in arid Central Asia to climate change during the Late Holocene over the centennial to millennial timescales remains contentious. The reason that primarily paleoenvironmental proxies interpreting diverse and the scarcity of accurate quantitative reconstruction records. In this study， we employed diatoms and pollen records from lacustrine sediment in the Aibi Lake of SW Junggar Basin to quantitatively reconstruct salinity and watershed precipitation amounts while exploring the associated forcing mechanism. The results indicate that Aibi Lake salinity varied between 2 g/L and 47 g/L during the Late Holocene period， indicating a generally brackish environment， and corresponding to prevailing Tryblionella granulata diatom in the lake basin. Westerly-dominated annual precipitation varied between 250 mm and 320 mm during the Late Holocene period in the basin， exhibiting a generally semi-arid environment and prevailing desert steppe vegetation. The Aibi Lake has a low salinity of average value of ～15 g/L. It exhibits elevated precipitation (average value of ～280 mm) during the periods of the Roman Warm Period (2900-1990 cal yr BP)， Dark Age Cold Period (1570-1140 cal yr BP)， and Little Ice Age (590-120 cal yr BP). The reconstructed precipitation and salinity exhibit a periodicity of ～200 years， which is consistent with the cycle of phase changes of the North Atlantic Oscillation (NAO) and Total Solar Irradiance (TSI). This correlation suggests that variations in NOA and TSI significantly influence the precipitation and salinity changes in Central Asia over centennial to millennial timescales.

The seemingly useless reeds are prepared as thermal insulation materials,which not only meet the requirements of environmental sustainability but also enhance the added value of reeds,creating new economic *** hydrophobicity of raw biomass surfaces leads to problems such as weak bonding strength and non-dense structure in the formed materials,as well as issues related to the residual insect infestations on the *** this study,reed straw was used as the raw material,and foamed geopolymer was used as the binder to prepare building insulation materials based *** improve the interfacial adhesion performance between reed straw and foamed geopolymer,a thermochemical modification method-thermal carbonization,was *** this study,the mechanical properties and hydraulic properties of the studied materials with different degrees of surface thermal modification were tested,especially the fire resistance performance,and weathering resistance performance rarely found in published *** the surface thermal modification condition of reed straw was 250℃（30 min）,the comprehensive performance of reed-based building insulation materials was the best,when the studied material density was 321.3 kg/m3;the compressive strength was 0.59 MPa;the thermal conductivity was 0.101 W/（m·K）;the pH was 11.27;the moisture absorption rate was 25.1%,and the compressive strength loss rate in wet-dry cycles was 18.5%.In addition,it had excellent fire resistance performance and weathering resistance *** new material can be widely used to improve the thermal insulation of traditional buildings and as sandwich filler in prefabricated buildings,such as preparing insulating walls.

Mangroves are crucial to the ecological security of the Earth and human well-being. Their management, conservation, and restoration are of great importance and necessitate the support of spatio-temporal information and multidisciplinary knowledge such as biology and ecology. Traditional knowledge services such as plant atlas provide illustrated textual knowledge of mangroves. However, this kind of service is oriented to information retrieval and is incapable of effectively mining and utilizing fragmented knowledge from multi-source heterogeneous data, facing the problem of “massive data, rare knowledge”. Knowledge graphs are capable of extracting, organizing, and fusing the knowledge contained in massive data into semantic networks that can be understood and computed by computers. They provide a solution for the realization of intelligent knowledge services. Focusing on the urgent need for mangrove knowledge acquisition, formal representation, and intelligent services, this paper proposes a research prospect on mangrove knowledge graphs and knowledge services. We first analyze the similarities and differences between various domain-specific concepts of Tupu. On this basis, we define the mangrove knowledge graph as a large-scale knowledge base that integrates multi-disciplinary knowledge and spatio-temporal information with mangrove ecosystems as the core. Then, we propose a research framework for mangrove knowledge services that can realize the transformation from multi-modal data to intelligent knowledge services, including multiple research levels such as ubiquitous data sensing and aggregation, knowledge organization and graph construction, and intelligent mangrove knowledge services. Subsequently, the methods and workflow for constructing mangrove knowledge graphs are introduced. Due to the lack of accurate data and complex parameterization,the prediction of groundwater depth is a chal-lenge for numerical *** learning can effectively solve this issue and has been proven useful in the prediction of groundwater depth in many *** this study,two new models are applied to the prediction of groundwater depth in the Ningxia area,*** two models combine the improved dung beetle optimizer(DBO)algorithm with two deep learning models:The Multi-head Attention-Convolution Neural Network-Long Short Term Memory networks(MH-CNN-LSTM)and the Multi-head Attention-Convolution Neural Network-Gated Recurrent Unit(MH-CNN-GRU).The models with DBO show better prediction performance,with larger R(correlation coefficient),RPD(residual prediction deviation),and lower RMSE(root-mean-square error).Com-pared with the models with the original DBO,the R and RPD of models with the improved DBO increase by over 1.5%,and the RMSE decreases by over 1.8%,indicating better prediction *** addition,compared with the multiple linear regression model,a traditional statistical model,deep learning models have better prediction performance.

Nano high-entropy oxide cathode with enhanced stability for direct borohydride fuel cells

High-entropy materials have become high-activity electrocatalysis owing to their high-entropy effect and multiple active ***,we synthesize a series of carbon-supported nano high-entropy oxides（HEOs/C）,specifically （PtFeCoNiCu）O/C,using a carbothermal shock （CTS） method for application as a cathode catalyst in direct borohydride fuel cells （DBFCs）.The microstructure of the prepared catalysts was characterized by X-ray photoelectron spectroscopy,X-ray absorption fine structure,and transmission electron *** prepared （PtFeCoNiCu）O/C,with particle sizes ranging from 2 to 4 nm,demonstrates 3.94 transferred electrons towards the oxygen reduction reaction in an alkaline environment,resulting in a minimal H2O2yield of 2.6%.Additionally,it exhibits a Tafel slope of 61 mV dec-1,surpassing that of commercial Pt/C (82 mV dec-1).Furthermore,after 40,000 cycles of cyclic voltammetry（CV） testing,the half-wave potential of （PtFeCoNiCu）O/C shows a positive shift of 3 mV,with no notable decline in the limiting current *** （PtFeCoNiCu）O/C is used as a cathode catalyst in DBFCs,the DBFC achieves a maximum power density of 441 mW cm-2at 60°C and sustains a cell voltage of approximately 0.73 V after 52 h at 30°*** findings confirm that HEO/C is a promising cathode catalyst for DBFCs.
