Studies onﬁsh fauna should not only focus onﬁsh composition and the comparison ofﬁsh composition among the study region and adjacent regions,but should also explore the origin and uniqueness of different taxa as well as the substitution of genera and species.In this study,the value ofﬂoristic presence method was modiﬁed and renamed the value ofﬁsh fauna presence(VFFP)method.The speciﬁc steps of the VFFP method and the traditionalﬁsh fauna analysis(TFFA)method were reﬁned and standardized.Then,the VFFP and TFFA methods were applied to study theﬁsh fauna of the upper and middle portions of the Mekong River basin.The results indicate that the TFFA method reﬂects the families(subfamilies)and genera that constitute the main body ofﬁsh in the studied river basin.The results of the VFFP method show which families(subfamilies)and genera are representative for the basin.Therefore,combining the TFFA and VFFP methods to analyze the composition ofﬁsh fauna can reﬂect the characteristics ofﬁsh fauna from different perspectives.The case study shows that theﬁsh fauna of the Mekong River is a part of theﬁsh fauna of Southeast Asia.Although it shows some similarities to the composition of South Asianﬁsh fauna,it does not belong to the South Asianﬁsh fauna as a whole,and is essentially different from the East Asianﬁsh fauna.This study provides an objective,quantitative,and veriﬁable method for studyingﬁsh fauna.

Environmental DNA(eDNA) metabarcoding has emerged as a powerful noninvasive tool for biodiversity assessments. Accuracy and limitations of these efforts are highly dependent on the choice of primer pairs. Although various primer sets have been used in DNA metabarcoding studies of amphibians, there is little direct comparison of their reliability and efficiency. Here, we used the Chinese amphibians from all three Orders that includes 13 families, and 83 species to evaluate the versatility and specificity of primer sets for the use of amphibian eDNA metabarcoding based on the publicly available and de novo designed primer sets, including Mod/VertOI, Batra2S, BA6S, Amph1OI and Amph2OI. Three pairs of primers(Batra2S, BA6S, Amph1OI + Amph2OI) were highly recommended as they could successfully amplify all the major clades of Chinese amphibians in our study. Non-amphibian taxa were also amplified by these primers, which implies that further optimization and development of amphibian-specific primers are still needed in future applications. These three primers in our eDNA survey have completely covered the species richness obtained by conventional survey methods and effectively distinguished some of the species(n=20) in Wenshan National Nature Reserve. No one primer set could individually detect all of the species’ richness of the studied area, indicating that multiple primers are required for a comprehensive survey of Chinese amphibians. Besides, seasonal variation in amphibians’ composition were also revealed by eDNA metabarcoding as for the traditional survey methods. These results indicate that eDNA metabarcoding has the potential to be a powerful tool for studying spatial and temporal community changes in amphibian species

Species delimitation is essential to informing conservation policy and understanding ecological and evolutionary processes.Most of our recent gains in knowledge on animal diversity rely on morphological characteristics and mitochondrial(mt)DNA variation.Concordant results based on both have led to an unprecedented acceleration in the identification of new species and enriched the field of taxonomy.However,discordances are also found commonly between morphological and mtDNA evidence.This confounds species delimitation,especially when gene flow or mt genome introgression has occurred.Here,we illustrate how mt genome introgression among species of the Odorrana grahami complex confounds species delimitation using the combined evidence of morphological characters,mt variation,and thousands of nuclear single-nucleotide polymorphisms(SNPs)from genotyping-by-sequencing(GBS).Fifty-eight samples across the distribution of the O.grahami complex were included.The mtDNA matrilineal genealogy indicated 2 clades,with O.grahami and Odorrana junlianensis clustered together.In contrast,all nuclear evidence including gene trees,species trees,and genetic structure analyses based on GBS data support 3 species with distinct genetic clusters.These 3 distinct genetic clusters also correspond to distinct morphological characters.They affirm the distinct taxonomic entities of both O.grahami and O.junlianensis,as well as a third clade distinct from either.Which species the third clade belongs to remains unclear and will require further testing.The nuclear genomic loci contradict the COI evidence,with indications of rampant historical mt genome introgression among the species of the O.grahami complex.These discordant signals previously confused species delimitation efforts in this group.Based on these findings,we recommend the integration of independent data,especially nuclear genomic evidence,in species delimitation so as to be robust against the pitfalls of mt introgression.

We describe a new species of the genus Hebius and provide evidence for the validity of H.septemlineatus comb.nov..Morphological and molecular analyses of Hebius specimens collected in yunnan Province,China,revealed three distinct lineages,namely the newly described Hebius weixiensis sp.nov.,as well as H.octolineatus(Boulenger,1904),and H.septemlineatus comb.nov.(Schmidt 1925),which is removed from synonymy with H.octolineatus.Based on mitochondrial genealogy,Hebius weixiensis sp.***.is sister to H.septemlineatus comb.nov.,while H.octolineatus is sister to H.bitaeniatus.The new species and H.septemlineatus comb.

Dear Editor,e family Ranidae is one of the largest and most families of amphibians(Che et al.,2007),with 64 recognized genera and429 species(Frost,2022).However,the systematics and natural history of some ranid groups have not been well-studied.One such example is the taxonomic relationship between Sylvirana,H ylarana and other closely-related taxa(Che et al.,2007;Pyron and Wiens,2011;Oliver et al.,2015).China has a large diversity of ranid frogs,with 146 speciesbelonging to 7 genera,and48 species belonging to 7 genera in the southern province of yunnan(AmphibiaChina,2022).

DEAR EDITOR,A new loach species(Cypriniformes: Nemacheilidae:yunnanilini), yunnanilus chuanheensis sp. nov., was caught from Chuanhe in the upper reaches of the Lixianjiang River, a tributary of the Red River in Nanjian County, yunnan Province, China. This species is a member of the traditional Y.pleurotaenia species group based on the presence of a lateral line and cephalic lateral-line canals.

A new species of the genus Brachytarsophrys,named Brachytarsophrys wenshanensis sp.nov.,has been identified in southeastern yunnan,China.This new species can be readily distinguished from other known congeners by both morphological criterion and molecular analysis of three mitochondrial gene segments:16S,COI,and Cytb.This classification is based on the following morphological characters:(1)medium body size(SVL 83.8–85.1 mm in two adult males);(2)enormous head,with head width nearly 1.2 times the length;(3)tongue pyriform,feebly notched;(4)non-meeting heels;(5)male lacking nuptial pad;(6)tibiotarsal articulation reaching angle of mouth when hindlimbs are extended forward alongside the body;(7)absence of outer metatarsal tubercle,inner metatarsal tubercle elliptic and approximately equal to first toe;(8)rudimentary toe webbing,webbing formula:Ⅰ(2–)–(2^(++))Ⅱ(2^(–))–(3^(++))Ⅲ(2^(½))–(4)Ⅳ(4^(+))–(2^(⅔))V;(9)lateral fringes narrow;(10)dermal ridge or glandular fold on dorsum absent;(11)pectoral glands distinct and irregular,femoral gland small.Our work increases the number of species within the genus Brachytarsophrys to 9.

Cyrtodactylus geckos are one of the most speciose and diverse groups of extant lizards known,distributed throughout the Asian and Pacific realms.Using molecular phylogenetic methods and supporting morphological data,we describe a new species of Cyrtodactylus in Daweishan National Nature Reserve,yunnan Province,China.Cyrtodactylus hekouensis sp.***.can be morphologically distinguished from its nearby congeners by the following characters:maximum SVL 92.3 mm and TL 98.5 mm;11-12 supralabials;11-12 infralabials;36-57 scale rows between the fifth supralabials;10-13 dorsal tubercles rows;3 postnasals on blunt and smooth front snout;precloacalfemoral pores in a continuous series of 33-39(females with pitted scales)located under vent/cloaca and thighs in both sexes;precloacal groove absent;3/3 postcloacal tubercles;subdigital lamellae under the fourth finger 21 or 22,under the fourth toe 20-23;smooth midbody with smooth venter and tuberculate dorsal scale rows,tubercles from head to tail base;dorsal transverse patterns are generally large,bilaterally symmetrical.The results of the phylogenetic analysis recover specimens of this new species as sister to a clade containing C.wayakonei and C.martini.Uncorrected pairwise intraspecific distances were<1%,and distances between our new species and other Cyrtodactylus species from nearby countries ranged from 14.2%to 26.8%.

The Dianchi golden-line barbel,Sinocyclocheilus grahami(Regan,1904),is one of the“Four Famous Fishes”of yunnan Province,China.Given its economic value,this species has been artificially bred successfully since 2007,with a nationally selected breed(“S.grahami,Bayou No.1”)certified in 2018.For the future utilization of this species,its growth rate,disease resistance,and wild adaptability need to be improved,which could be achieved with the help of molecular markerassisted selection(MAS).In the current study,we constructed the first chromosome-level genome of S.grahami,assembled 48 pseudo-chromosomes,and obtained a genome size of 1.49 Gb.We also performed QTL-seq analysis of S.grahami using the highest and lowest bulks(i.e.,largest and smallest size)in both a sibling and random population.We screened two quantitative trait loci(QTLs)(Chr3,14.9-39.1 Mb and Chr17,4.1-27.4 Mb)as major growth-related locations.Several candidate genes(e.g.,map2k5,stat1,phf21a,sox6,and smad6)were also identified,with functions related to growth,such as cell differentiation,neuronal development,skeletal muscle development,chondrogenesis,and immunity.These results built a solid foundation for in-depth MAS studies on the growth traits of S.grahami.
