Establishment of a system for screening and identification of novel bactericide targets in the plant pathogenic bacterium Xanthomonas oryzae pv. oryzae using Tn-seq and SPR

Xanthomonas spp. cause severe bacterial diseases. However, effective strategies for prevention and management of these diseases are scarce. Thus, it is necessary to improve the efficiency of control of diseases caused by Xanthomonas. In this study, Xanthomonas oryzae pv. oryzae(Xoo), which causes rice bacterial leaf blight, has been studied as a representative. A transposon insertion library of Xoo, comprising approximately 200,000 individual insertion mutants, was generated. Transposon sequencing data indicated that the mariner C9 transposase mapped at 35.7–36.4% of all potential insertion sites, revealing 491 essential genes required for the growth of Xoo in rich media. The results show that, compared to the functions of essential genes of other bacteria, the functions of some essential genes of Xoo are unknown, 25 genes might be dangerous for the Xanthomonas group, and 3 are specific to Xanthomonas. High-priority candidates for developing broad-spectrum, Xanthomonas-specific, and environment-friendly bactericides were identified in this study. In addition, this study revealed the possible targets of dioctyldiethylenetriamine using surface plasmon resonance(SPR) in combination with high performance liquid chromatography–mass spectrometry(HPLC–MS). The study also provided references for the research of some certain bactericides with unknown anti-bacterial mode of action. In conclusion, this study urged a better understanding of Xanthomonas,provided meaningful data for the management of bacterial leaf blight, and disclosed selected targets of a novel bactericide.

In this study,a novel residential virtual power plant(RVPP)scheduling method that leverages a gate recurrent unit(GRU)-integrated deep reinforcement learning(DRL)algorithm is *** the proposed scheme,the GRU-integrated DRL algorithm guides the RVPP to participate effectively in both the day-ahead and real-time markets,lowering the electricity purchase costs and consumption risks for *** Lagrangian relaxation technique is introduced to transform the constrained Markov decision process(CMDP)into an unconstrained optimization problem,which guarantees that the constraints are strictly satisfied without determining the penalty ***,to enhance the scalability of the constrained soft actor-critic(CSAC)-based RVPP scheduling approach,a fully distributed scheduling architecture was designed to enable plug-and-play in the residential distributed energy resources(RDER).Case studies performed on the constructed RVPP scenario validated the performance of the proposed methodology in enhancing the responsiveness of the RDER to power tariffs,balancing the supply and demand of the power grid,and ensuring customer comfort.

Insight study of rare earth elements in PM_(2.5)during five years in a Chinese inland city:composition

Two viral proteins translated from one open reading frame target different layers of plant defense

Multilayered defense responses are activated upon pathogen *** utilize a number of strategies to maximize the coding capacity of their small genomes and produce viral proteins for infection,including suppression of host ***,we reveal translation leakage as one of these strategies:two viral effec-tors encoded by tomato golden mosaic virus,chloroplast-localized C4(cC4)and membrane-associated C4(mC4),are translated from two in-frame start codons and function cooperatively to suppress ***4 localizes in chloroplasts,to which it recruits NbPUB4 to induce ubiquitination of the outer membrane;as a result,this organelle is degraded,and chloroplast-mediated defenses are ***,chloroplast-localized cC4 induces the production of singlet oxygen(1O2),which in turn promotes translo-cation of the 1O2 sensor NbMBS1 from the cytosol to the nucleus,where it activates expression of the CERK1 ***,an antiviral effect exerted by CERK1 is countered by mC4,localized at the plasma ***4,like cC4,recruits NbPUB4 and promotes the ubiquitination and subsequent degradation of CERK1,suppressing membrane-based,receptor-like kinase-dependent ***,this translation leakage strategy seems to be conserved in multiple viral species and is related to host *** suggests that stacking of different cellular antiviral responses could be an effective way to abrogate viral infection and engineer sustainable resistance to major crop viral diseases in thefield.

Oil and gas pipelines are affected by many factors,such as pipe wall thinning and pipeline *** prediction of failure pressure of oil and gas pipelines can provide technical support for pipeline safety *** at the shortcomings of the BP Neural Network(BPNN)model,such as low learning efficiency,sensitivity to initial weights,and easy falling into a local optimal state,an Improved Sparrow Search Algorithm(ISSA)is adopted to optimize the initial weights and thresholds of BPNN,and an ISSA-BPNN failure pressure prediction model for corroded pipelines is *** 61 sets of pipelines blasting test data as an example,the prediction model was built and predicted by MATLAB software,and compared with the BPNN model,GA-BPNN model,and SSA-BPNN *** results show that the MAPE of the ISSA-BPNN model is 3.4177%,and the R2 is 0.9880,both of which are superior to its comparison *** the ISSA-BPNN model has high prediction accuracy and stability,and can provide support for pipeline inspection and maintenance.

REGENERATION FACTOR 1, a peptide boost for wound healing and plant biotechnology

plants respond to wounding such as caused by insect herbivores by activating their defenses and thus increasing their resistance against attacking pests and opportunistic pathogens(Howe and Schaller,2008).The phenomenon was first described more than 50 years ago by Green and Ryan,who reported that wounding triggers tomato and potato plants to produce proteinase inhibitors that inhibit digestive proteases in the gut of leaf-eating insects to starve them of nutrients(Green and Ryan,1972).They also observed that the accumulation of proteinase inhibitors is not restricted to wounded leaves but occurs throughout the aerial parts of the *** search for a chemical signal that is released locally in response to wounding and that mediates systemic activation of defense gene expression led to the identification of systemin,the first ever peptide with hormonelike activity in plants(Pearce et al.,1991).These seminal findings sparked much research into systemic defense responses and peptide signaling in general,and we know now that systemin is one of many phytocytokines that regulate a multitude of immune responses in plants(Schilmiller and Howe,2005;Gust et al.,2017).
