Engineering Photocleavable Protein-decorated Hydrogels to Regulate Cell Adhesion and Migration

Cell adhesion and migration play essential roles in tissue development and maintenance, and abnormal cell migration is involved in life-threatening diseases, including vascular disease, tumor formation, and metastasis. The advances in hydrogel-based 3D cell culture development facilitated the investigation of cell motility behavior, including cell-cell and cell-matrix adhesion and cell migration in a microenvironment more related to in vivo situations. Establishing advanced methods for these in vitro studies is thus necessary. Photo-sensitive proteins show advantages in remote and non-invasive regulation of hydrogels’ properties, and thus are of great potential in regulating 3D cultured cells’ behavior. In the presented study, we engineered photocleavable protein(PhoCl)-decorated hydrogels to regulate cell adhesion and migration of MDA-MB-231. The integrin-binding motif RGD was fused to the PhoCl and was decorated on the hydrogel. After being exposed to light at 405 nm, the PhoCl was cleaved and the RGD motif was released, resulting in detachment of the binding cells. The regulatory effect of the light illumination showed a time-dependent and cell density-dependent manner. Furthermore, the elimination of RGD by patterned light exposure completely suspended the cell migration to the corresponding region, suggesting a controllable regulation of the cell migration direction.

To elucidate the process of adaptation, particularly the traits subject to natural selection and the molecular mechanisms underlying their natural variation, is one of the primary objectives of evolutionary biology. The uplifted landscape offers an excellent framework for understanding how organisms adapt to dramatic climatic gradients. To investigate the genetic basis of plant adaptation to the extremely high altitude, we first compared the genomic and phenotypic variations of two closely related Arabidopsis thaliana accessions from high altitude(Xizang, also known as “Tibet”) and low altitude(Yunnan), respectively. The Xizang population represents a relict group characterized by a small effective population size. Notably, the Xizang genome has more transposable elements(TEs) and more gene loss-offunction(LoF) mutations. Differentially expressed genes were enriched in biological processes of cellular response to oxygen-containing compound, regulation of defense response, and response to light intensity. Intriguingly, the phenotypic selection analysis revealed that silique density was under natural selection. Furthermore, we genetically mapped and validated that the LoF mutation of GA20ox1, the homologous gene of green revolution in rice, resulted in a higher silique density in Xizang Arabidopsis. Given that GA20ox1 is linked to Arabidopsis adaptation to the Alps Mountains, its parallel evolution plays an important role in the adaptation to Alpine habitats. Overall,our results highlight that identifying adaptive traits and elucidating the molecular mechanisms underlying natural variation of these traits is crucial for unraveling the mystery of adaptive evolution and has significant implications for crop breeding.

The sea-to-air flux of dimethylsulphide（DMS） is one of the major sources of marine biogenic aerosol, and can have an important radiative impact on climate, especially in the Arctic Ocean. Satellite-derived aerosol optical depth（AOD） is used as a proxy for aerosol burden which is dominated by biogenic aerosol during summer and autumn. The spring sea ice melt period is a strong source of aerosol precursors in the Arctic. However, high aerosol levels in early spring are likely related to advection of continental pollution from the south（Arctic haze）.Higher AOD was generally registered in the southern part of the study region. Sea ice concentration（SIC） and AOD were positively correlated, while cloud cover（CLD） and AOD were negative correlation. The seasonal peaks of SIC and CLD were both one month ahead of the peak in AOD. There is a strong positive correlation between AOD and SIC. Melting ice is positively correlated with chlorophyll a（CHL） almost through March to September,but negatively correlated with AOD in spring and early summer. Elevated spring and early summer AOD most likely were influenced by combination of melting ice and higher spring wind in the region. The peak of DMS flux occurred in spring due to the elevated spring wind and more melting ice. DMS concentration and AOD were positively correlated with melting ice from March to May. Elevated AOD in early autumn was likely related to the emission of biogenic aerosols associated with phytoplankton synthesis of DMS. The DMS flux would increase more than triple by 2100 in the Greenland Sea. The significant increase of biogenic aerosols could offset the warming in the Greenland Sea.

Macroalgae dominate nutrient dynamics and function as high-value foods for microbial,meio-and macrofaunal communities in coastal *** of this vital role,it is important to clarify the physiological information associated with environmental changes as it reflects their growth *** evaluate the effects of the changes in salinity and nutrients,the photosynthetic efficiency of a green macroalga Ulva fasciata from the Daya Bay was tested at a range of salinity(i.e.,31 to 10 psu)and nitrogen content(i.e.,5 to 60μmol L^(-1)).The results showed that cellular chlorophyll a(Chl a),carbohydrate and protein contents of *** were increased due to reduced salinity,and were decreased by interactive nitrogen *** a short culture period(i.e.,18 h),the reduced salinity decreased the maximum photosynthetic efficiency(rETRmax and Pmax)derived from the rapid light response curve and photosynthetic oxygen evolution rate versus irradiance curve,respectively,as well as the saturation irradiance(E_(K)).This reducing effect diminished with enlonged cultivation time and reversed to a stimulating effect after 24 h of *** nitrogen enrichment stimulated the rETRmax and Pmax,as well as the E_(K),regardless of salinity,especially within short-term cultivation period(i.e.,<24 h).In addition,our results indicate that seawater freshening lowers the photosynthetic efficiency of *** in the short term,which is mitigated by nitrogen enrichment,but stimulates it in the long term,providing insight into how macroalgae thrive in coastal or estuarine waters where salinity and nutrients normally covary strongly.

We investigate the relationship between the variability of broad absorption lines(BALs)or narrow absorption lines(NALs)and that of continuum using a data set of two-epoch SDSS spectra containing 134 C IV NAL-BAL *** analysis reveals an anti-correlation between the fractional equivalent width(EW)variations in NALs(or BALs)and the fractional flux variations of the continuum,with Spearman rank correlation coefficients of r=-0.47(p=1E-08)and r=-0.58(p=1E-13),*** addition,we find a positive correlation between the fractional EW variations in NALs and BALs(r=0.72,p=1E-22),and derive a regression equation ΔEW_(NAL)/=0.803ΔEW_(BAL)/+0.008,with an intrinsic scatter of *** results suggest that the variability in the ionizing continuum may play a significant role in the observed changes in C IV NALs and BALs,supporting the idea of photoionization-driven *** co-variability between C IV NALs and BALs may imply that they originate from outflows with similar physical conditions.

Patterns and drivers of species–genetic diversity correlations(SGDCs)have been broadly examined across taxa and ecosystems and greatly deepen our understanding of how biodiversity is ***,few studies have examined the role of canopy structural heterogeneity,which is a defining feature of forests,in shaping ***,we determine what factors contribute toα-andβ-species–genetic diversity correlations(i.e.,α-andβ-SGDCs)in a Chinese subtropical *** this purpose,we used neutral molecular markers to assess genetic variation in almost all adult individuals of the dominant tree species,Lithocarpus xylocarpus,across plots in the Ailaoshan National Natural *** also quantified microhabitat variation by quantifying canopy structure heterogeneity with airborne laser scanning on 201-ha subtropical forest *** found that speciesα-diversity was negatively correlated with geneticα-*** structural heterogeneity was positively correlated with speciesα-diversity but negatively correlated with geneticα-*** contrasting effects contributed to the formation of a negativeα-***,we found that canopy structural heterogeneity increases speciesα-diversity and decreases geneticα-diversity by reducing the population size of target ***β-diversity,in contrast,was positively correlated with geneticβ-*** in canopy structural heterogeneity between plots had non-linear parallel effects on the two levels ofβ-diversity,while geographic distance had a relatively weak effect onβ-*** study indicates that canopy structural heterogeneity simultaneously affects plot-level community species diversity and population genetic diversity,and species and genetic turnover across plots,thus drivingα-andβ-SGDCs.

Metabolic Effect of Dietary Erythritol Intake on Escherichia coli

Artificial sweeteners are man-made substitutes for diet that needs low sugars or caloric intake. Recent studies have shown that AS consumption is possibly associated with metabolic disturbances and intestinal flora disturbances. Erythritol is a kind of 4-carbon sugar substitute in the form of sugar alcohol, which may contribute to the prevention of gingivitis by inhibit the biofilm formation of oral bacteria. Despite these studies, the data on how erythritol affects commensal flora causing pathogenicity to remain limited. In this study, microbiota (Escherichia coli) models were used to investigate the effects of different concentration of erythritol on the metabolism of bacteria, especially on Escherichia coli’s growth, transformation efficiency and hemolytic activity. The results showed that sweeteners decreased the bacteria’s ability to normally grow in higher concentration, and form biofilms to varying degrees. And the addition of erythritol in low concentration may have an effect of promote the growth of Escherichia coli. Habitual consumption of artificial sweeteners in the diet continues to increase was associated with the maintenance of micro-ecological equilibrium in gut.
