Systematic identification of wheat spike developmental regulators by integrated multiomics, transcriptional network, GWAS, and genetic analyses

The spike architecture of wheat plays a crucial role in determining grain number,making it a key trait for optimization in wheat breeding programs.In this study,we used a multi-omic approach to analyze the transcriptome and epigenome profiles of the young spike at eight developmental stages,revealing co-ordinated changes in chromatin accessibility and H3K27me3 abundance during the flowering transition.We constructed a core transcriptional regulatory network(TRN)that drives wheat spike formation and experimentally validated a multi-layer regulatorymodule involving TaSPL15,TaAGLG1,and TaFUL2.By integrating the TRN with genome-wide association studies,we identified 227 transcription factors,including 42 with known functions and 185 with unknown functions.Further investigation of 61 novel transcription factors using multiple homozygous mutant lines revealed 36 transcription factors that regulate spike architecture or flowering time,such as TaMYC2-A1,TaMYB30-A1,and TaWRKY37-A1.Of particular interest,TaMYB30-A1,downstream of and repressed by WFzP,was found to regulate fertile spikelet number.Notably,the excellent haplotype of TaMYB30-A1,which contains a C allele at the WFzP binding site,was enriched during wheat breeding improvement in China,leading to improved agronomic traits.Finally,we constructed a free and open access Wheat Spike Multi-Omic Database(http://39.***.48.156:8800/#/).Our study identifies novel and high-confidence regulators and offers an effective strategy for dissecting the genetic basis of wheat spike development,with practical value forwheat breeding.

Background:Physical activity(PA)is important for cancer survivors.Trials of remotely delivered interventions are needed to assist in reaching under-served non-metropolitan cancer survivors.The objective of this study was to ascertain whether wearable technology,coupled with health coaching was effective in increasing PA in breast and colorectal cancer survivors living in regional and remote areas in Australia.Methods:Cancer survivors from 5 states were randomized to intervention and control arms.Intervention participants were given a Fitbit Charge 2TMand received up to 6 telephone health coaching sessions.Control participants received PA print materials.Accelerometer assessments at baseline and 12 weeks measured moderate-to-vigorous PA(MVPA),light PA,and sedentary behavior.Results:Eighty-seven participants were recruited(age=63±11 years;74(85%)female).There was a significant net improvement in MVPA of 49.8 min/week,favoring the intervention group(95%confidence interval(95%CI):13.6-86.1,p=0.007).There was also a net increase in MVPA bouts of 39.5 min/week(95%CI:11.9-67.1,p=0.005),favoring the intervention group.Both groups improved light PA and sedentary behavior,but there were no between-group differences.Conclusion:This’s the first study to demonstrate that,when compared to standard practice(i.e.,PA education),a wearable technology intervention coupled with distance-based health coaching,improves MVPA in non-metropolitan cancer survivors.The results display promise for the use of scalable interventions using smart wearable technology in conjunction with phone-based health coaching to foster increased PA in geographically disadvantaged cancer survivors.

Evolution of Ordovician YJ1X ultra-deep oil reservoir in the Yuecan oilfield, Tarim Basin, NW China

Based on a combined in-situ calcite U-Pb dating, molecular geochemical correlations of reservoir oil and extract from reservoir rocks, and fluid inclusion analysis, the charge and evolution history of the YJ1X ultra-deep oil reservoir of the Ordovician Yijianfang Formation in the southwestern part of the Tabei Uplift has been determined systematically.(1) The reservoir oil, free oil and inclusion oil have similar geochemical characteristics and are presumably derived from marine source rock deposited in similar sedimentary environment.(2) The reservoir oil, free oil and inclusion oil have similar maturities with calculated equivalent vitrinite reflectance values in the range of 0.80%-0.96%.(3) Two types(Group Ⅰ and Ⅱ) of oil inclusion assemblages(OIAs) have been identified in the reservoir, of which Group Ⅰ represents the original gas-saturated oil entering the trap during the initial oil charge, whereas Group Ⅱ represents undersaturated residual oil retained in the reservoir after minor leakage of light hydrocarbon.(4) The reservoir experienced oil charge only once during the Early devonian around 425 Ma and has been well preserved after the minor light hydrocarbon leakage in the Middle devonian. The study shows that there may be old oil and gas accumulations in ultra-deep strata of petroliferous basins with well-developed caprock and stable tectonic background.

Bulk Mg-based hydrogen storage materials have the potential to provide a low-cost solution to diversify energy storage and transportation.Compared to nano powders which require handling and processing under hydrogen or an inert gas atmosphere,bulk Mg-based alloys are safer and are more oxidation re-sistant.Conventional methods and existing infrastructures can be used to process and handle these ma-terials.However,bulk Mg alloys have smaller specific surface areas,resulting in slower hydrogen sorp-tion kinetics,higher equilibrium temperatures,and enthalpies of hydride formation.This work reviews the effects of the additions of a list of alloying elements and the use of innovative processing meth-ods,e.g.,rapid solidification and severe plastic deformation processes,to overcome these drawbacks.The challenges,advantages,and weaknesses of each method and future perspectives for the development of Mg-based hydrogen storage materials are discussed.

The Soft X-ray Imager(SXI)is part of the scientific payload of the Solar wind Magnetosphere Ionosphere Link Explorer(SMILE)mission.SMILE is a joint science mission between the European Space Agency(ESA)and the Chinese Academy of Sciences(CAS)and is due for launch in 2025.SXI is a compact X-ray telescope with a wide field-of-view(FOV)capable of encompassing large portions of Earth’s magnetosphere from the vantage point of the SMILE orbit.SXI is sensitive to the soft X-rays produced by the Solar Wind Charge eXchange(SWCX)process produced when heavy ions of solar wind origin interact with neutral particles in Earth’s exosphere.SWCX provides a mechanism for boundary detection within the magnetosphere,such as the position of Earth’s magnetopause,because the solar wind heavy ions have a very low density in regions of closed magnetic field lines.The sensitivity of the SXI is such that it can potentially track movements of the magnetopause on timescales of a few minutes and the orbit of SMILE will enable such movements to be tracked for segments lasting many hours.SXI is led by the University of Leicester in the United Kingdom(UK)with collaborating organisations on hardware,software and science support within the UK,Europe,China and the United States.

Tethyan evolution from early Paleozoic to early Mesozoic in southwest Yunnan

The Tethys orogenic belt in SW Yunnan constitutes a critical part of the expansive Tethys-Himalayan tectonic domain.The abundant,well-preserved geologic records make it an ideal area for studying the tectonic evolution of Proto-and Paleo-Tethys.In this paper,we focus on several major tectonic units in SW Yunnan and reconstruct the Tethyan evolution from the early Paleozoic to the early Mesozoic,based on stratigraphic,sedimentologic,and magmatic evidences.The recently discovered early Paleozoic Yunxian-Menghai ophiolitic belt in the Lincang Terrane situated east of the Changning-Menglian Belt represents the suture zone of the Proto-Tethys.The oceanic basin of Proto-Tehtys opened in the latest Neoproterozoic and subsequently began subducting in the late Miaolingian of the Cambrian(about 505 Ma).From the late Late Ordovician to the ealiest Silurian(about 450–442 Ma),the Proto-Tethys basin gradually closed resulting in the collision of the continental plates on both sides of the Proto-Tethyan ocean.The main collision stage occurred in the early Silurian(about 442–430 Ma)and the postcollision stage lasted from the mid-Silurian to the early Carboniferous(430–355 Ma).The earliest record of Paleo-Tethyan oceanic crust was generated in the late devonian,and the ocean was then subducted in an eastward direction in the middle Late Carboniferous(about 310 Ma).The initial collision stage in the Paleo-Tethys took place at the end of the Permian(about 253Ma),and the main stage of the collision persisted into the early Ladinian(about 253–238 Ma).This was followed by postcollision extension from the late Ladinian to the early Jurassic(ca.238–196 Ma).We suggest that the opening of Paleo-Tethyan Ocean in SW Yunnan was a result of the extensional rift basin of the Proto-Tethys.Additionally,the activity of the Manxin mantle plume was likely a crucial factor in the rapid expansion of the Paleo-Tethyan Ocean.

Optimizing prediction models for pancreatic fistula after pancreatectomy:Current status and future perspectives

Postoperative pancreatic fistula(POPF)is a frequent complication after pancre-atectomy,leading to increased morbidity and mortality.Optimizing prediction models for POPF has emerged as a critical focus in surgical research.Although over sixty models following pancreaticoduodenectomy,predominantly reliant on a variety of clinical,surgical,and radiological parameters,have been documented,their predictive accuracy remains suboptimal in external validation and across diverse populations.As models after distal pancreatectomy continue to be pro-gressively reported,their external validation is eagerly anticipated.Conversely,POPF prediction after central pancreatectomy is in its nascent stage,warranting urgent need for further development and validation.The potential of machine learning and big data analytics offers promising prospects for enhancing the accuracy of prediction models by incorporating an extensive array of variables and optimizing algorithm performance.Moreover,there is potential for the development of personalized prediction models based on patient-or pancreas-specific factors and postoperative serum or drain fluid biomarkers to improve accuracy in identifying individuals at risk of POPF.In the future,prospective multicenter studies and the integration of novel imaging technologies,such as artificial intelligence-based radiomics,may further refine predictive models.Addressing these issues is anticipated to revolutionize risk stratification,clinical decision-making,and postoperative management in patients undergoing pancre-atectomy.

The giant Kalgoorlie Gold Field revisited

The Neoarchaean Kaigoorlie Gold Field contains the giant Golden Mile and world-class Mt Charlotte deposits,which have been the subject of much research for over 100 years.The Golden Mile deposit is a complex array of ductile to brittle vein and breccia lodes that are predominantly hosted in the highlyfractionated Golden Mile Dolerite sill.The Fimiston lodes comprise an array of narrow lodes that evolved broadly syn- to late-formation of the regional D2 NW-trending foliation.The lodes are characterized by pyrite veinlets and disseminations,quartz veinlets and breccias,and banded quartz-carbonate veins with sericite,carbonate,and pyrite-dominated alteration.Bonanza Green-Leader,or Oroya-style,lodes,with grades in excess of 1000 g/t Au,are similar to the Fimiston-style lodes,but are characterized by abundant visible gold,native tellurium and more abundant telluride minerals within roscoelitebearing alteration zones.The arguably structurally younger Mt Charlotte-style lodes are characterized by a pipe-shaped,coarse-grained quartz,carbonate and scheelite vein-stockwork with distinct verticallyzoned,carbonate-sericite-albite-pyrite ± pyrrhotite dominant alteration assemblages around veins within Unit 8 of the Golden Mile dolerite and porphyry dykes.The network of steep- and gently-dipping extension and shear fracture-fill veins are associated with NE-trending fault se ts that cross cut the regional NW-trend.The deposit area is intruded by swarms of porphyry dykes,including syn-volcanic mafic dykes,early and volumetrically most significant c.2.67 Ga feldspar-phyric porphyry dykes,as well as later c.2.66-2.65 Ga calc-alkaline hornblende-phyric dykes associated with younger c.2.65-2.64 Ga lamprophyre dykes.All post-volcanic dykes have similar orientations to the Fimiston lodes.The feldspar dykes are clearly overprinted by all styles of mineralization,although the relationship between hornblende-phyric and lamprophyre dykes and gold mineralization is more ambiguous.Most agree that gold mineralization was post-peak regional

Source to sink zircon grain shape:Constraints on selective preservation and significance for Western Australian Proterozoic basin provenance

The effect of selective preservation during transportation of zircon grains on the detrital age spectrum is difficult to quantify and could potentially lead to systematic bias in provenance analysis. Here we investigate whether the shape of detrital zircon grains holds provenance information and if the grain shape can assist in understanding preservation. We applied multiple linear regression analysis to identify significant shape properties in detrital zircons from Proterozoic metasediments of the Capricorn and Amadeus basins and their Archean and Proterozoic sources in the Yilgarn Craton and the Musgrave Province in Western Australia. Digital images and isotopic data from 819 SIMS U-Pb dated zircons were examined for correlation between grain shape, age, U and Th content. Out of twelve shape descriptors measured, Minor Axis, the width of zircon grains perpendicular to the crystallographic c-axis, consistently shows the most significant correlation with isotopic age. In the studied population Archean grains are narrower than Proterozoic grains: the probability that grains wider than 75 μm are Archean is less than30%.Calculations of the proportions of source material in sedimentary rocks relative to the proportions of source material in the overall catchment area(erosion parameter 'K' calculated based on age spectra)produced values typical for mature river systems, with K = 6 for the Yilgarn-Capricorn and K = 5.5 for the Musgrave-Amadeus source-sink system. For the Yilgarn-Capricorn system, we also calculated 'K' based on Minor Axis, to determine whether grain width can be linked to age populations. Results of the shapebased K of 5.3 suggest a similarity between age-based and shape-based 'K' values, demonstrating that zircon grain width may be a useful discriminator of provenance. Contrary to commonly applied qualitative shape classifications, we found no consistent correlations between shape descriptors of magmatic zircons and the composition of their host rock. When will it end? Long-lived intracontinental reactivation in central Australia

The post-Mesoproterozoic tectonometamorphic history of the Musgrave Province, central Australia, has previously been solely attributed to intracontinental compressional deformation during the 580 -520 Ma Petermann Orogeny. However, our new structurally controlled multi-mineral geochronology results,from two north-trending transects, indicate protracted reactivation of the Australian continental interior over ca. 715 million years. The earliest events are identified in the hinterland of the orogen along the western transect. The first tectonothermal event, at ca. 715 Ma, is indicated by40 Ar/39 Ar muscovite and U e Pb titanite ages. Another previously unrecognised tectonometamorphic event is dated at ca. 630 Ma by Ue Pb analyses of metamorphic zircon rims. This event was followed by continuous cooling and exhumation of the hinterland and core of the orogen along numerous faults, including the Woodroffe Thrust,from ca. 625 Ma to 565 Ma as indicated by muscovite, biotite, and hornblende40 Ar/39 Ar cooling ages. We therefore propose that the Petermann Orogeny commenced as early as ca. 630 Ma. Along the eastern transect,40 Ar/39 Ar muscovite and zircon(Ue Th)/He data indicate exhumation of the foreland fold and thrust system to shallow crustal levels between ca. 550 Ma and 520 Ma, while the core of the orogen was undergoing exhumation to mid-crustal levels and cooling below 600-660℃. Subsequent cooling to 150 -220℃ of the core of the orogen occurred between ca. 480 Ma and 400 Ma(zircon [Ue Th]/He data)during reactivation of the Woodroffe Thrust, coincident with the 450 -300 Ma Alice Springs Orogeny.Exhumation of the footwall of the Woodroffe Thrust to shallow depths occurred at ca. 200 Ma. More recent tectonic activity is also evident as on the 21 May, 2016(Sydney date), a magnitude 6.1 earthquake occurred, and the resolved focal mechanism indicates that compressive stress and exhumation along the Woodroffe Thrust is continuing to the present day.
