Hepatocellular carcinoma(HCC)is one of the most prevalent cancers globally.In co.trast to the declining death rates observed for all other co.mon cancers such as breast,lung,and prostate cancers,the death rates for HCC co.tinue to increase by^2e3%per year because HCC is frequently diagnosed late and there is no curative therapy for an advanced HCC.The early diagnosis of HCC is truly a big challenge.Over the past years,the early diagnosis of HCC has relied on surveillance with ultrasonography(US)and serological assessments of alpha-fetoprotein(AFP).However,the specificity and sensitivity of US/AFP is not satisfactory enough to detect early onset HCC.Recent technological advancements offer hope for early HCC diagnosis.Herein,we review the progress made in HCC diagnostics,with a focus on emerging imaging techniques and biomarkers for early disease diagnosis.

Background:Epigenetic aberrations have been intensively studied in diverse cancers.However,universal features shared among all cancer types have received minimal attention in the era of precision medicine.For primary cervical cancer screening,the high false-positive rate of the human papillomavirus(HPV) test and cervical cytology and the urgent need in distinguishing between high-grade squamous intraepithelial lesion(HSIL) and low-grade squamous intraepithelial lesion(LSIL) call for potent biomarkers.Methods:We analyzed PCDHGB7 methylation in 17 types of cancer in TCGA datasets(n= 7,069)and verified it in 13 types of clinical cancer samples(n= 727) by bisulfite-PCR pyrosequencing.Next,we co.centrated on cervical cancer and detected PCDHGB7methylation in 86 primary tissues,404 cervical smears,and 273 vaginal secretions by bisulfite-PCR pyrosequencing and methylation-sensitive restriction enzymes-based quantitative PCR(MSRE-qPCR),and evaluated it as a biomarker for early cervical cancer screening.Results:Here,we found PCDHGB7 was universally hypermethylated in 17 cancer types from TCGA datasets,which was proven in 13 types of clinical cancer samples with a high area under the curve(AUC) value(≥ 0.85).The methylation detection of PCDHGB7 can be applied to early cervical cancer screening in cervical smears and even in self-sampled vaginal secretions with a sensitivity of 90.9% and specificity of 90.4%.Furthermore,the methylation status of PCDHGB7 can clearly distinguish HSIL and LSIL,which will co.tribute to early theranostic management of cervical cancer.co.clusions:Hypermethylated PCDHGB7 co.ld serve as a pan-cancer(or "universal-cancer-only methylation",Uco.) marker and its implementation would greatly improve the currently used cervical cancer screening strategy and benefit womens health.

The present study was designed to characterize the blood chemistry, hematology, and lymphocyte subsets in pregnant rhesus monkeys and provide baseline parameters for future studies of reproductive and developmental toxicity and developmental immunotoxicity. Harem-mating was used in 96 female and 16 male rhesus monkeys. Pregnancy was co.firmed on gestation day(GD)18 by ultrasound. The blood samples of rhesus monkeys were co.lected at various times(20 days before pregnancy and GD20, 100 and 150). The analyses of blood chemistry, hematology, and lymphocyte subsets were performed. co.mpared with 20 days before pregnancy, Significant decreases(P biologic change were observed, There were no significant changes in MCH, RDW-CV, MPV, BUN, CD3+, CD4+ and CD8+ during pregnancy. These data provide a database for preclinical study in rhesus monkeys. Physiological anemia, hyperglycemia, and immune suppression may occur in pregnant rhesus monkey which is similar to that found in human, and it is essential to distinguish the physiological changes from the pharmaco.ogical effects in reproductive and developmental toxicity and developmental immunotoxicity studies of pharmaceuticals.

The surface macromolecules of probiotic bacteria play crucial roles in modulating immune responses in the host. Exopolysaccharide (EPS) from lactic acid bacteria (LAB) have been widely reported to exhibit immunomodulatory activity. In this study， the EPS biosynthesis gene cluster of Lacticaseibacillus paracasei S-NB was analyzed and a deletion mutant S-NB△7576 (two genes S-NB＿2175/wze and S-NB＿2176/wzd were responsible for the chain length determination and export of EPS) was successfully co.structed， resulting a 40.02% decrease in the production of EPS. The deletion of wze and wzd had little effect on the monosaccharide co.position and major groups of the two EPS fractions (BEPS1 and BEPS2). Both BEPS1 and BEPS2 co.ld inhibit the transcriptional level of tumor necrosis factor-α (TNF-α)， interleukin-1β (IL-1β)， inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (co.-2) mRNA in RAW 264.7 cells induced by lipopolysaccharide (LPS)， and enhance host immune tolerance via suppressing NF-κB and MAPK signaling. Notably， the S-NB△7576 mutant supplied with the BEPS1/BEPS2 exhibited more significant inhibition of cytokines production and the phosphorylation of p65 and c-Jun N-terminal kinase (JNK) in LPS-stimulated cells co.pared with the S-NB△7576 mutant alone. Our study provided the immunomodulatory effect of BEPS1 and BEPS2 from L. paracasei S-NB， in which the wze and wzd genes associated with EPS biosynthesis may play an important role.

The causes and pathogenesis of sensitive skin are relatively co.plex.Herein,the causes and pathogenesis of sensitive skin are summarized based on previous research results.The causes of sensitive skin mainly include various external factors,internal factors,and other dermatological factors.The mechanisms are co.plex,including skin barrier function injury,changes in epidermal microbial status,abnormal sensory nerve fiber function,activation of TRPV1 channel,neurovascular-immune-inflammatory pathway,transcriptome,etc.Sensitive skin is often induced by multiple mechanisms.The analysis and discussion of the causes and mechanisms of sensitive skin will be helpful to guide clinical practice.

The effects of temperature and wavelength choice on in-situ dissolution test instrument of Cimetidine were studied. Absorbance （A）〈 1.0 is required when using a fiber-optic dissolution test system. The detection wavelength of 2 （218 nm） was replaced by 244 nm to carry out this test. The absorbance of Cimetidine solution at different temperature showed an obvious change. Calibration of Cimetidine solution should be tested at the same temperature （37° C） with the test solution. A suitable wavelength with smaller tangent slope co.ld be chosen for in-situ dissolution test of Cimetidine tablets.

Study on Skin Care Efficacy of the Active Ingredients in Camellia Japonica Flowers at High and Low Altitudes

The active ingredients of Camellia japonica flowers(CJF)at high and low altitudes,as well as their skin care efficacy were co.pared.The dried red CJF at high and low altitudes were ultrasonically extracted with 70%ethanol,and these extracts were co.centrated and then diluted to a co.stant volume.The co.tent of total flavonoids,total polyphenols and total proteins was tested and analyzed.In addition,DPPH free radical scavenging,inhibition of the formation of advanced glycation end products,and inhibitory activity against elastase was measured to co.pare their skin care efficacy in anti-oxidation,anti-glycation,anti-wrinkling and firming.The results showed that,based on the dry weight of CJF,the co.tent of total flavonoids and total proteins of the CJF at high altitude was approximately 1.5 times of the CJF at low altitude,while the co.tent of total polyphenols was approximately 2.4 times that of the CJF at low altitude.The skin care efficacy of CJF at high altitude was significantly better than that of the CJF at low altitude.This work co.ld provide theoretical basis for the selection and application of Camellia japonica flowers in the field of co.metics.

Triple-negative breast cancer(TNBC)due to lack of clear target and notorious“co.d”tumor microenvironment(TME)is one of the most intractable and lethal malignancies.Tuning“co.d”TME into“hot”beco.es an emerging therapeutic strategy to TNBC.Herewith,we report that integrin-targeting micellar gemcitabine and paclitaxel(ATN-mG/P,ATN sequence:Ac-PhScNK-NH2)co.perating with polymersomal CpG(NanoCpG)effectively“heated up”and treated TNBC.ATN-mG/P exhibited greatly boosted apoptotic activity in 4T1 cells,induced potent immunogenic cell death(ICD),and efficiently stimulated maturation of bone marrow-derived dendritic cells(BMDCs).Remarkably,in a postoperative TNBC model,ATN-mG/P co.bining with NanoCpG promoted strong anti-cancer immune responses,showing a greatly augmented proportion of mature DCs and CD8^(+)T cells while reduced immune-suppressive myeloid-derived suppressor cells(MDSCs)and regulatory T cells(T_(reg)),which led to co.plete inhibition of lung metastasis and 60%mice tumor-free.The co.delivery of gemcitabine and paclitaxel at desired ratio in co.bination with NanoCpG provides a unique platform for potent chemoimmunotherapy of“co.d”tumors like TNBC.

The global market demand for natural astaxanthin is rapidly increasing owing to its safety,the potential health benefits,and the diverse applications in food and pharmaceutical industries.The major native producers of natural astaxanthin on industrial scale are the alga Haematoco.cus pluvialis and the yeast Xanthopyllomyces dendrorhous.However,the natural production via these native producers is facing challenges of limited yield and high co.t of cultivation and extraction.Alternatively,astaxanthin production via metabolically engineered non-native microbial cell factories such as Escherichia co.i,Saccharomyces cerevisiae and Yarrowia lipolytica is another promising strategy to overco.e these limitations.In this review we summarize the recent scientific and biotechnological progresses on astaxanthin biosynthetic pathways,transcriptional regulations,the interrelation with lipid metabolism,engineering strategies as well as fermentation process co.trol in major native and non-native astaxanthin producers.These progresses illuminate the prospects of producing astaxanthin by microbial cell factories on industrial scale.
