Engineering geological characteristics of slide zone soil and stability for one landslide in Tibet

On the basis of geological investigating work and experimental studies on slide zone soil of one landslide in Tibet,the authors analyzed granulometric composition,clay mineral composition and physical and mechanical properties for the soil in the slide zone.The soil samples are gravel containing fine particle.Particles larger than 2 mm occupy the main proportion with the content 51.5%--68.5%.The relative content of clay minerals is low.The clay minerals are illite smectite mixed layer and kaolinite,and their relative contents are 6%--13% and 4%-11%,respectively.The main mineral ingredient is quartz and the relative content is over 30%.Therefore,the soil’s hydrophily is poor.The cohesion and internal friction angle are high,causing preferable physical-mechanical features of slide zone soil.On the basis of the obtained data,the landslide stability is evaluated by means of limit equilibrium method.The safety factors are 3.191 and 1.92 respectively under both natural and normal water level conditions.The study results show that the landslide is stable.It can provide the appropriate basis and reference for landslide stability evaluation and landslide control in Tibet.

Copper plays an important role in many metabolic activities in the human body.Copper level in the human body is in a state of dynamic equilibrium.Recent research on copper metabolism has revealed that copper dyshomeostasis can cause cell damage and induce or aggravate some diseases by affecting oxidative stress,proteasome,cuprotosis,and angiogenesis.The liver plays a central role in copper metabolism in the human body.Research conducted in recent years has unraveled the relationship between copper homeostasis and liver diseases.In this paper,we review the available evidence of the mechanism by which copper dyshomeostasis promotes cell damage and the development of liver diseases,and identify the future research priorities.

Epidermal growth factor receptor(EGFR)mutations are common oncogenic driver mutations in patients with non-small cell lung cancer(NSCLC).The application of EGFR-tyrosine kinase inhibitors(TKIs)is beneficial for patients with advanced and early-stage NSCLC.With the development of next-generation sequencing technology,numerous patients have been found to have more than one genetic mutation in addition to a single EGFR mutation;however,the efficacy of conventional EGFR-TKIs and the optimal treatments for such patients remain largely unknown.Thus,we review the incidence,prognosis,and current treatment regimens of EGFR compound mutations and EGFR concomitant mutations to provide treatment recommendations and guidance for patients with these mutations.

Background:Radiation(IR)-induced DNA damage triggers cell cycle arrest and has a suppressive effect on the tumor microenvironment(TME).Wee1,a cell cycle regulator,can eliminate G2/M arrest by phosphorylating cyclin-dependent kinase 1(CDK1).Meanwhile,programed death-1/programed death ligand-1(PD-1/PDL-1)blockade is closely related to TME.This study aims to investigate the effects and mechanisms of Wee1 inhibitor AZD1775 and anti-PD-1 antibody(anti-PD-1 Ab)on radiosensitization of hepatoma.Methods:The anti-tumor activity of AZD1775 and IR was determined by 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyltetrazolium bromide(MTT)assay on human and mouse hepatoma cells HepG2,Hepa1-6,and H22.The anti-hepatoma mechanism of AZD1775 and IR revealed by flow cytometry and Western blot in vitro.A hepatoma subcutaneous xenograft mice model was constructed on Balb/c mice,which were divided into control group,IR group,AZD1775 group,IR+AZD1775 group,IR+anti-PD-1 Ab group,and the IR+AZD1775+anti-PD-1 Ab group.Cytotoxic CD8^(+)T cells in TME were analyzed by flow cytometry.Results:Combining IR with AZD1775 synergistically reduced the viability of hepatoma cells in vitro.AZD1775 exhibited antitumor effects by decreasing CDK1 phosphorylation to reverse the IR-induced G2/M arrest and increasing IR-induced DNA damage.AZD1775 treatment also reduced the proportion of PD-1^(+)/CD8^(+)T cells in the spleen of hepatoma subcutaneous xenograft mice.Further studies revealed that AZD1775 and anti-PD-1 Ab could enhance the radiosensitivity of hepatoma by enhancing the levels of interferonγ(IFNγ)^(+)or Ki67^(+)CD8 T cells and decreasing the levels of CD8^(+)Tregs cells in the tumor and spleen of the hepatoma mice model,indicating that the improvement of TME was manifested by increasing the cytotoxic factor IFNγexpression,enhancing CD8^(+)T cells proliferation,and weakening CD8^(+)T cells depletion.Conclusions:This work suggests that AZD1775 and anti-PD-1 Ab synergistically sensitize hepatoma to radiotherapy by enhancing IR-induced DNA damage and improv

Tumor chemoprevention and treatment are two approaches aimed at improving the survival of patients with cancers.An ideal anti-tumor drug is that which not only kills tumor cells but also alleviates tumor-causing risk factors,such as precancerous lesions,and prevents tumor recurrence.Chinese herbal monomers are considered to be ideal treatment agents due to their multi-target effects.Astragaloside has been shown to possess tumor chemoprevention,direct anti-tumor,and chemotherapeutic drug sensitization effects.In this paper,we review the effects of astragaloside on tumor prevention and treatment and provide directions for further research.

Hepatocellular carcinoma(HCC)is the most common type of primary hepatocellular carcinoma(PHC).Early diagnosis of HCC remains the key to improve the prognosis.In recent years,with the promotion of the concept of precision medicine and more in-depth analysis of the biological mechanism underlying HCC,new diagnostic methods,including emerging serum markers,liquid biopsies,molecular diagnosis,and advances in imaging(novel contrast agents and radiomics),have emerged one after another.Herein,we reviewed and analyzed scientific advances in the early diagnosis of HCC and discussed their application and shortcomings.This review aimed to provide a reference for scientific research and clinical practice of HCC.

Small cell lung cancer(SCLC)is a highly malignant tumor with a very poor prognosis;therefore,more effective treatments are urgently needed for patients afflicted with the disease.In recent years,emerging molecular classifications based on key transcription factors of SCLC have provided more information on the tumor pathophysiology,metastasis,immune microenvironment,and acquired therapeutic resistance and reflected the intertumoral heterogeneity of the various SCLC phenotypes.Additionally,advances in genomics and single-cell sequencing analysis have further revealed the high intratumoral heterogeneity and plasticity of the disease.Herein,we review and summarize these recent lines of evidence and discuss the possible pathogenesis of SCLC.

In this paper,we propose a new stopping criterion for Eckstein and Bertsekas’s generalized alternating direction method of multipliers.The stopping criterion is easy to verify,and the computational cost is much less than the classical stopping criterion in the highly influential paper by Boyd et al.(Found Trends Mach Learn 3(1):1–122,2011).

Platelet-rich plasma promotes peripheral nerve regeneration after sciatic nerve injury

The effect of platelet-rich plasma on nerve regeneration remains controversial.In this study,we established a rabbit model of sciatic nerve small-gap defects with preserved epineurium and then filled the gaps with platelet-rich plasma.Twenty-eight rabbits were divided into the following groups(7 rabbits/group):model,low-concentrati on PRP(2.5-3.5-fold concentration of whole blood platelets),medium-concentration PRP(4.5-6.5-fold concentration of whole blood platelets),and high-concentration PRP(7.5-8.5-fold concentration of whole blood platelets).Electrophysiological and histomorphometrical assessments and proteomics analysis we re used to evaluate regeneration of the sciatic nerve.Our results showed that platelet-rich plasma containing 4.5-6.5-and 7.5-8.5-fold concentrations of whole blood platelets promoted repair of sciatic nerve injury.Proteomics analysis was performed to investigate the possible mechanism by which platelet-rich plasma promoted nerve regeneration.Proteomics analysis showed that after sciatic nerve injury,platelet-rich plasma increased the expression of integrin subunitβ-8(ITGB8),which participates in angiogenesis,and differentially expressed proteins were mainly enriched in focal adhesion pathways.Additionally,two key proteins,ribosomal protein S27 a(RSP27 a)and ubiquilin 1(UBQLN1),which were selected after protein-protein interaction analysis,are involved in the regulation of ubiquitin levels in vivo.These data suggest that platelet-rich plasma promotes peripheral nerve regeneration after sciatic nerve injury by affecting angiogenesis and intracellular ubiquitin levels.
