Although the somatic cell nuclear transfer(SCNT) technique has been used extensively for cloning and generating transgenic pigs,the cloning efficiency is still very *** has been proposed that the low efficiency of this technique is the result of incomplete epigenetic reprogramming and abnormal gene expression during early embryonic *** this study,we investigate the effect of Scriptaid,a low-toxicity histone deacetylase inhibitor,on the developmental competence of porcine SCNT *** found that treating SCNT embryos with 500 nmol/L Scriptaid for 15 h after activation significantly enhanced the blastocyst formation rate(27.7%) compared with the untreated group(control)(12.2%,Pembryos with Scriptaid increased the level of histone acetylation on histone H3 at lysine 14(acH3K14).Furthermore,treating embryos with Scriptaid increased the expression level of three genes that play important roles during embryonic development(Oct4,Klf4 at the blastocyst stage and Nanog at the 4-cell stage).Moreover,the expression level of the apoptosis-related gene Caspase-3 was significantly lower in the Scriptaid-treated SCNT embryos compared with the control SCNT embryos at the 4-cell and blastocyst *** conclusion,these results indicate that Scriptaid treatment improves the development and nuclear reprogramming of porcine SCNT embryos.

CRISPR/Cas9-mediated genome editing can inhibit virus infection by targeting the conserved regions of the viral genomic DNA. Unexpectedly, we found previously that pseudorabies virus(PRV) could escape from CRISPR/Cas9-mediated *** order to elucidate whether the escape of PRV from Cas9-mediated inhibition was due to cell deficiencies, such as genetic instability of sgRNA or Cas9 protein, the positive cells were passaged ten times, and PRV infection in the sgRNA-expressing cells was evaluated in the present study. The results showed that subculturing cells has no effect on Cas9-mediated cleavage of PRV. Different passages of PX459-PRV cells can stably express sgRNA to facilitate Cas9/sgRNA cleavage on the UL30 gene of PRV, resulting in a pronounced inhibition of PRV infection. Studies to elucidate the mechanism of PRV escape are currently in progress.

Relationship between dietary amino acid release kinetics and nitrogen deposition in growing pigs

Although the protein content of swine diets is formulated based on the ileal digestibility of protein and amino acids(AA)under current nutrition requirements,the nitrogen utilization efficiency of swine varies based on protein source,which may be related to AA release *** this experiment,a 2×2 factorial arrangement with casein(CAS)-enriched or corn gluten meal(CGM)-enriched protein sources at different digestible crude protein levels(normal[N],13%;and low[L],11%)were applied to 24 crossbred(DurocLandraceYorkshire)growing pigs(average body weight=43.3±3.5 kg)in 4 treatments(***,***,***,***,respectively)to investigate the effects of AA release kinetics on nitrogen deposition in growing *** ileal digestible AA in all diets were balanced by adding individual AA to meet the nutrient *** AA release kinetics were detected in vitro by measuring the hydrolysis of various protein diets under pepsin and trypsin *** results demonstrated that the time of AA release peak in the CGM diet was 12 h later than that in the CAS *** synchronization indices of dietary AA release in ***,***,***,and *** were 23.73%,29.37%,23.40%,and 26.07%,*** *** had the poorest AA release synchronism while the *** had the greatest among the 4 ***,within the pigs,*** and *** showed the highest(81.08%)and lowest(73.54%)nitrogen biological values,respectively,despite the standard ileal digestible AA levels being equal for all *** results indicate that the release kinetics of dietary AA had great effect on nitrogen *** optimize nitrogen deposition,AA release kinetics and composition should be taken into consideration when formulating diets for growing pigs.

Recently,micro-vibrational perception mechanisms of nocturnal arthropods such as scorpions and spiders are attracting increasingly more attention and *** relevant micro-vibrational receptors are exquisite,in terms of their comprehensive performance such as sensitivity,stability,high anti-interference,and ultralow-power *** this work,we find the Basitarsal Compound Slit Sensilla (BCSS) of scorpion (Heterometrus petersii) are composed of the crack-shaped slits as mechanosensory structure and can efficiently detect substrate-borne vibrational signal in complex natural *** study on microstructures and mechanical properties of tissue phases constituting the BCSS reveals that the strategy of tessellation is used to make crack-shaped slit amplify the tiny mechanical *** addition,the magnitude-frequency characteristics of electrophysiological signals caused by vibration stimulation with different frequencies indicate that the scorpion is sensitive to micro-vibrational signals at a certain frequency ***,the vibrational perception mechanism based on geometrical amplification and resonance is proposed to explain how scorpions detect the tiny biotic vibrational signal efficiently in noise *** finding not only promotes our further understanding of ultra-sensitive mechanism of the vibrational receptors,but also provides biological inspiration for the next generation of mechanosensor for a broad range of applications.

With the completion of the draft assembly of the giant panda genome sequence,RNA sequencing technology has been widely used in genetic research on giant *** used RNA-seq to examine black and white hair follicle samples from adult *** comparison with the giant panda genome,75963 SNP loci were labeled,2426 differentially expressed genes(DEGs)were identiﬁed,and 2029 new genes were discovered,among which 631 were functionally annotated.A cluster analysis of the DEGs showed that they were mainly related to the Wnt signaling pathway,ECM–receptor interaction,the p53 signaling pathway,and ribosome *** enrichment results showed that there were signiﬁcant differences in the regulatory networks of hair follicles with different colors during the transitional stage of hair follicle resting growth,which may play a regulatory role in melanin synthesis during *** conclusion,our results provide new insights and more data support for research on the color formation in giant pandas.

Secreted frizzled-related protein 2 （SFRP2）, a member of the SFRPs family, is associated with cell growth and differentiation in myogenesis. Our previous study suggested that SFRP2 was a potential target of microRNA （miRNA）-I/206, which was considered as myomiRs. To further explore the biological function and regulation mechanisms of the SFRP2 gene in porcine skeletal muscle development, we first analyzed the sequence structure of the porcine SFRP2 gene. Subsequently, we detected its tissue distribution in adult Tongcheng pigs （a Chinese indigenous breed） and investigated its dynamic expression in developmental skeletal muscle （13 prenatal and 7 postnatal time points）in Tongcheng pigs. An interaction analysis between SFRP2 and myomiRs was also performed. The results showed that the expression pattern of the SFRP2 varied greatly across diverse tissues. It exhibited abundant expression in prenatal skeletal muscle and peaked at 55 days post coitus （E55）, and had a lower expression in postnatal skeletal muscle, indicating that the SFRP2 gene might affect porcine embryonic skeletal muscle development. Co-expression analysis revealed that the expression levels of SFRP2 correlated negatively with miRNA-1 （t=-0.570, P-value=0.009） and miRNA-206 （r=-0.546, P-value=0.013）, but positively with SFRP1 （r=0.613, P-value=0.004）. The bioinformatics analysis and dual luciferase assay verified that the SFRP2 was a putative target of miRNA-1/206 in pigs. Therefore, this study is helpful for understanding the biological function and mo- lecular regulation of the SFRP2 gene during porcine skeletal muscle development.

Dear Editor,Mink circovirus (MiCV), which is clustered in the genus Circovirus of the family Circoviridae, was first described in minks from farms in Dalian, China in 2013 (Lian et al.2014). The complete single-stranded circular genome of the virus is 1,753 nucleotides long and contains two major open reading frames (ORFs), designated ORF1 (Rep gene)and ORF2 (Cap gene)(Lian et al. 2014;Ge et al. 2018).

An ultrasensitive and selective sandwich-type fluorescent immunosensor based on fluorescence resonance energy transfer(FRET) between CdSe/ZnS quantum dots(QDs) and Au nanorods(AuNRs) was developed for the rapid detection of porcine reproductive and respiratory syndrome virus(PRRSV).Modified CdSe/ZnS QDs and AuNRs were bioconjugated with primary antibodies M and GP5,*** the target antigen was present,a well-known sandwich-type form CdSe/ZnS QDs-M antibody-antigen-GP5 antibody-AuNRs was produced,and thus reducing the distance between the QDs and *** a result,FRET occurred and the fluorescence intensity of CdSe/ZnS QDs decreased,and this decrease was used to determine the antigen *** detection of PRRSV at low concentrations in swine serum was achieved with a limit of detection of 0.55 TCID50/mL and a linear detection range of 10~1-3.5 × 10~4 TCID50/***,the fluorescent immunosensor is selective and highly sensitive;specifically,it can detect PRRSV at low concentrations in swine serum over a large concentration *** proposed detection method can facilitate the development of high-performance fluorescent immunosensors for the detection of PRRSV and other antigens.
