Mating patterns and pollen dispersal in a Japanese larch(Larix kaempferi) clonal seed orchard： a case study

Pollination dynamics highly determines the genetic quality of seed orchard crops. However, there is less research about the effect of mating patterns on seed productivity of orchard crops. So far, clonal seed orchards have been producing genetically improved seedlings used for most Japanese larch(Larix kaempferi(Lamb.) Carr.) plantations in China. In the present study, a total of 17 highly variable simple sequence repeat(SSR) markers were used for genotyping a progeny trial population consisting of 647 open-pollinated progenies germinated from seeds which were collected from 63 maternal clones with 140 potential paternal clones in a Japanese larch clonal seed orchard in China. Paternity analysis was used in the present case study in order to evaluate the level of paternal gametic contribution, estimate pollen contamination and selfing rates, and investigate pollination patterns,pollen dispersal patterns and the impact of mating patterns on seed productivity of orchard crops. We observed 93.7% of the success rate of the parental assignment, unequal paternal gametic contribution(0–12.4%) with 6.3% of the progenies derived from pollen contamination or unsampled pollen donors, and absence of evidence for selfing. We also found that pollination rate highly depended on the distance between pollen donors and maternal parents, the majority of the identified crossing(65.7%)occurred between clones within a 150-m radius, and large variations in growth performance existed among the paternal halfsiblings. Progeny growth performance(diameter at breast(DBH) and height(HGT)) was measured at Age-20 in order to investigate the impact of mating patterns on timber production of orchard crops. As either the paternal or maternal, two clones(i.e., clones Z38 and Z62) were identified to have produced progenies with higher average stem volume breeding values than that of all of the progenies. Isolation,expression and single nucleotide polymorphisms(SNPs) analysis of LACCASE gene(LkLAC8) from Japanese larch(Larix kaempferi)

Nucleotide diversity (pi) and linkage disequilibrium (LD) analysis based on SNP marker could provide a sound basis for choosing an association analysis method. Japanese larch (Larix kaempferi) is an important timber coniferous tree species for pulping and papermaking, but its high lignin content has significantly restricted it application potential. In this study, the LACCASE gene, that plays an important regulatory role for lignin biosynthesis, was selected as research target. The full-length cDNA and genomic sequences of the encoding LkLAC8 gene were isolated from the LACCASE expressed sequence tags of the Japanese larch transcriptome database using the rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR). The cDNA was determined to be 1940 bp, with an open reading frame (ORF, 1734 bp) that encoded a protein of 577 AA. This protein contains four highly specific Cu2+ binding sites and 11 glycosylation sites, thus belonging to the LACCASE family. The deduced protein sequence shared an 89% identity with the PtaLAC from Pinus taeda. A real-time PCR analysis showed that the LkLAC8 transcript was expressed predominantly in mature xylem, with moderate levels in the immature xylem, cambium and mature leaves, the lowest in the roots. Lastly, the genomic sequences of LkLAC8 in 40 individuals from six naturally distributed populations of Japanese larch were amplified, and a total of 201 SNPs (103 and 98 mutation types of transition and transversion, respectively) were detected; the frequency of the SNPs was 1/19 bp. Nucleotide diversity among the six populations ranged from 0.0034 to 0.0053, which suggested that there were no significant differences among the populations. The LD analysis showed that the LD level decayed rapidly within the increasing length of the LkLAC8 gene. These results implied that LD mapping and association analysis based on candidate gene may be feasible for the marker-assisted breeding of new germplasms with low lignin in Japanese larch.

A facile and efficient route to 4-aryl-2-[2-（trifluoromethylthio） aryl]quinazoline derivatives through a tandem directed Rh-catalyzed C-H iodination and trifluoromethylthiolation is described. The reaction proceeded under mild reaction conditions, exhibited good functional group tolerance with a broad scope of substrate and excellent regi- oselectivity in good to excellent yields.

Wood properties are important traits that determine quality of structural wood. With the aim of performing efficient early selection for wood properties we investigated genetic variation in 20 Larix kaempferi clones aged from 4 to 15 years for four quality traits: wood density, wall thickness to lumen area, microfibrillar angle(MFA) and modulus of elasticity(MOE). We observed that age-related trends in overall means varied for differen traits: MFA decreased with the age, while the others generally increased with the age. Phenotypic variance always showed significant differences from the age of 8 years onward, with CVG ranging from 4% to 25%. Also clonal repeatability increased steadily until 9 years old and then kept medium or higher intensity(0.4–0.8). After the age of 6, genetic correlations were generally higher than phenotypic correlations. Estimates of early selection efficiency suggested that the optimal selection age for wood density was at age 5–6 years, while it was 9–10 years for the other traits. In combination with previous results we proposed a comprehensive early selection strategy for larch clonal breeding that involved selection based on nursery rooting ability, phenology, growth traits, and wood properties.

Patterns of change in the population and spatial distribution of oriental white storks (Ciconia boyciana) wintering in Poyang Lake

Using total counts in simultaneous annual surveys, we monitored the population size and spatial distribution of oriental white storks （Ciconia boyciana） wintering in Poyang Lake between 1998 and 2011. Results showed that Poyang Lake wetland is an important wintering ground for oriental white storks, with an annual average population number of 2 305＋326. The population sizes in 2004, 2005, 2010, and 2011 were higher than the highest-ever estimate of its global population. In 2005, we recorded 3 789 individuals, which was the maximum population number within the period of 1998-2011. The storks inhabited 52 lakes, with the greatest distance between these lakes being 180.3 km. The storks presented a clustered distribution pattern in the Poyang Lake wetland, irrespective of the number of individuals or occurrence frequencies. Shahu, Dahuchi, Banghu, and Hanchihu were most frequently used lakes and had the largest annual average numbers of storks. There was a significant positive correlation between occurrence frequency and annual average number of storks in the lakes. Most of the lakes important for storks were covered by existing nature reserves, though some lakes outside the reserves were also frequently used. About 64.9%＋ 5.5% of the storks were found in nature reserves. In addition, the storks more frequently used and clumped in significantly larger flocks in lakes within nature reserves than lakes outside.

在资源富集的条件下,物种丰富度、群落地上生产力以及群落稳定性的季节变化通常被忽视。本研究致力于探究在干旱区草原,资源添加如何在生长季的不同月份影响物种丰富度、群落地上生产力及其稳定性。我们在内蒙古草原设置了为期3年的资源添加(氮添加-N、水添加-W以及水、氮共同添加-NW)实验,利用季节性取样的方法,去检验资源添加(水、氮)对物种丰富度、群落地上生产力及其稳定性的影响,并通过构建结构方程模型分析资源添加调控不同月份与整个生长季群落稳定性的内在机制及其相对重要性。研究结果表明,资源添加在整体上未改变5与6月的群落地上生产力,而氮与水氮共同添加显著提高了7与8月的群落地上生产力。资源添加在整体上未改变物种丰富度、物种异步性与群落稳定性。氮添加与水氮共同添加提高了7与8月的群落地上生产力,主要源于其增加了多年生丛生禾草的地上生产力。结构方程模型分析表明:在生长季前期与整个生长季,物种异步性是决定群落稳定性的主要机制;在生长季后期,多年生丛生禾草的稳定性是驱动群落稳定性的主要因子。我们的研究证明:在半干旱草原,季节与资源的有效性可以交互影响群落地上生产力及其稳定性。这些发现对于半干旱草原的季节性可持续管理具有重要意义,以期减轻土地利用与全球变化带来的影响。

Functional trait space and redundancy of plant communities decrease toward cold temperature at high altitudes in Southwest China

Plant communities in mountainous areas shift gradually as climatic conditions change with altitude. How trait structure in multivariate space adapts to these varying climates in natural forest stands is unclear. Studying the multivariate functional trait structure and redundancy of tree communities along altitude gradients is crucial to understanding how temperature change affects natural forest stands. In this study, the leaf area, specific leaf area, leaf carbon, nitrogen, and phosphorous content from 1,590 trees were collected and used to construct the functional trait space of 12 plant communities at altitudes ranging from 800 m to 3,800 m across three mountains. Hypervolume overlap was calculated to quantify species trait redundancy per community. First,hypervolumes of species exclusion and full species set were calculated, respectively. Second, the overlap between these two volumes was calculated to obtain hypervolume overlap. Results showed that the functional trait space significantly increased with mean annual temperature toward lower altitudes within and across three mountains, whereas species trait redundancy had different patterns between mountains. Thus, warming can widen functional trait space and alter the redundancy in plant communities. The inconsistent patterns of redundancy between mountains suggest that warming exerts varying influences on different ecosystems. Identification of climate-vulnerable ecosystems is important in the face of global warming.

Soil carbon budget in different-aged Chinese fir plantations in south China

Understanding the age effect on soil carbon balance in forest ecosystems is important for other material cycles and forest man-agement. In this research we investigated soil organic carbon density, litter production, litter decomposition rate, soil respiration, and soil mi-crobial properties in a chronosequence of four Chinese fir plantations of 7, 16, 23 and 29 years at Dagangshan mountain range, jiangxi Province, south China. There was a significant increasing trend in litter production with increasing plantation age. Litter decomposition rate and soil respira-tion, however, declined from the 7-year to the 16-year plantation, and then increased after 16 years. This was largely dependent on soil micro-organisms. Soil carbon output was higher than carbon input before 16 years, and total soil carbon stock declined from 35.98 t＆#183;ha-1 in the 7-year plantation to 30.12 t＆#183;ha-1 in the 16-year plantation. Greater litter produc-tion could not explain the greater soil carbon stock, suggesting that forest growth impacted this microbial process that controlled rates of soil car-bon balance together with litter and soil respiration. The results highlight＆amp;nbsp;the importance of the development stage in assessing soil carbon budget and its significance to future management of Chinese fir plantations.

Optimization of Germinated Conditions for Somatic Embryos in Liriodendron Hybrids

The development of somatic embryos is a key technology in plant breeding. To gain the best cultural conditions,the factors influencing on germination and transplanting survival rate of the somatic embryos in Liriodendron hybrids were optimized,including the development period of somatic embryos,macro-element,Vitamin C,genotypes,plant hormones and transplant medium in this study. The results showed that the optimal development phase of germination was torpedo-shaped embryo,and the base cultural medium of 3/4 MS with vitamin C can keep normal form from somatic embryos. The germination rate of somatic embryo had obvious genotype difference,among which the hybrid 1 × 5002 was the most sensitive( 82. 46%),and the hybrid 1 × 4088 was the most insensitive( 13. 24%). The medium with 0. 1 mg/L of KT + 0. 1 mg/L of IBA can help promote the germination of somatic embryogenesis. In addition,the yellow soil was more suitable than mixed peat soil for transplanting Liriodendron hybrids seedlings. Therefore,the obtainment of the optimal conditions has important guidance for production practices of Liriodendron hybrids.
