BACKGROUND Fracture nonunion represents a challenging complication during fracture repair,often necessitating surgical ***,a recombinant human parathyroid hormone,has demonstrated promise in enhancing fracture healing,although its efficacy in treating established nonunion remains under *** SUMMARY We report a case of a 27-year-old male who presented with a right humerus fracture following a traffic *** undergoing open reduction and internal fixation,the fracture resulted in a delayed union and subsequent *** 4 years of conservative management,teriparatide treatment was initiated due to persistent *** injections were administered daily for 6 months,resulting in complete fracture healing and resolution of *** Our case demonstrates the successful use of teriparatide in treating a prolonged nonunion of a humerus *** may provide a valuable therapeutic option for established bone nonunion,even in cases that have not responded to conservative treatments.

Flexible electrodes have significant potential in fields such as healthcare and neuroscience, as they enable comprehensive recording and analysis of human electrical signals, providing deeper insights into physiological conditions and the state of internal organs and tissues [1, 2]. However, commercial Ag/AgCl wet electrodes face issues such as dehydration,poor comfort, and insufficient mechanical strength, while other types of electrodes, like dry and semi-dry electrodes,also have their own limitations, including the higher contact impedance.

Nanoplastic(NP)contaminants are receiving increasing attention due to the harm they can cause to aquatic organisms and their accumulation in the human body through the food *** goal of this study was to evaluate the oxidative stress caused by NPs on the clams Meretrix meretrix and Sinonovacula *** were exposed to three concentrations of fluorescently labeled polystyrene NPs(PS-NPs;0,0.1,and 1 mg L^(-1))for 7 d followed by a 3 d recovery *** activities of antioxidant enzymes,such as catalase(CAT),superoxide dismutase(SOD),glutathione peroxidase(GPx)and the content of malonaldehyde(MDA)were measured in the digestive gland,gills,and mantle *** showed that the SOD activity in the gills of *** was significantly higher than that of the control at day 1,and PS-NPs exposure inhibited the CAT activity in the gills and SOD and GPx activities in the *** to low or high concentrations of PS-NPs increased the activities of both CAT and GPx in the gills of *** at day *** oxidative damage was more severe in the digestive gland of *** and in the mantle of *** based on MDA level,and it returned to normal after *** biomarker response,index version 2(IBRv2)values showed that the gills were more sensitive to PS-NPs than mantle and digestive ***,the oxidative stress by PS-NPs depended on exposure time,dose,organ,and species.

Previous studies have demonstrated that cis9， trans11， cis15-CLNA (c9， t11， c15-CLNA)， a metabolite of Bifidobacterium breve CCFM683， possesses anti-inflammatory properties in the intestine. However， its safety profile remains unexplored. This study establishes the safe dose range for c9， t11， c15-CLNA and investigate the potential damage pathways. The results indicate that the maximum safe dose for mice is lower than 625 mg/kg， primarily due to hepatocyte damage. In cellular experiments revealed that an overdose of c9， t11， c15-CLNA activates peroxisome proliferator-activated receptor gamma (PPAR-γ)， leading to hepatic lipid accumulation. The addition of the PPAR-γ inhibitor partially mitigated this effect. In summary， this study established that the maximum tolerated dose of c9， t11， c15-CLNA in healthy adult C57BL/6N mice is 625 mg/kg. For human application， the estimated safe dose is below 50.62 mg/kg. Additionally， c9， t11， c15-CLNA was found to activate the PPARγ protein in the liver， leading to lipid accumulation within hepatic tissue.

Premature ovarian insufficiency(POI)caused by chemotherapy is a common complication in female cancer survivors of childbearing *** methods,including mesenchymal stem cell(MSC)transplant and hormone replacement therapy,have limited clinical application because of their drawbacks,and more methods need to be *** the current study,the potential effects and underlying mechanisms of human umbilical cord MSC-derived extracellular vesicles(h UCMSC-EVs)were investigated in a cisplatin(CDDP)-induced POI mouse model and a human granulosa cell(GC)*** results showed that h UCMSC-EVs significantly attenuated body weight loss,ovarian weight loss,ovary atrophy,and follicle loss in moderate-dose(1.5 mg/kg)CDDP-induced POI mice,similar to the effects observed with h *** further found that the h UCMSCEVs inhibited CDDP-induced ovarian GC apoptosis by upregulating anti-apoptotic mi RNA levels in GCs,thereby downregulating the m RNA levels of multiple pro-apoptotic *** general,our findings indicate that the moderate-dose chemotherapy may be a better choice for clinical oncotherapy,considering effective rescue of the oncotherapy-induced ovarian damage with h ***,multiple mi RNAs in h UCMSC-EVs may potentially be used to inhibit the chemotherapy-induced ovarian GC apoptosis,thereby restoring ovarian function and improving the life quality of female cancer patients.

The research and development of new traditional Chinese medicine(TCM) drugs have progressively established a novel system founded on the integration of TCM theory, human experience, and clinical trials(termed the “Three Combinations”). However, considering TCM's distinctive features of “syndrome differentiation and treatment” and “multicomponent formulations and complex mechanisms”, current TCM drug development faces challenges such as insufficient understanding of the material basis and the overall mechanism of action and an incomplete evidence chain system. Moreover, significant obstacles persist in gathering human experience data, evaluating clinical efficacy, and controlling the quality of active ingredients, which impede the innovation process in TCM drug development. Network pharmacology, centered on the “network targets” theory, transcends the limitations of the conventional“single target” reductionist research model. It emphasizes the comprehensive effects of disease or syndrome biological networks as targets to elucidate the overall regulatory mechanism of TCM prescriptions. This approach aligns with the holistic perspective of TCM, offering a novel method consistent with TCM's holistic view for investigating the complex mechanisms of TCM and developing new TCM drugs. It is internationally recognized as a “next-generation drug research model”. To advance the research of new tools, methods, and standards for TCM evaluation and to overcome fundamental, critical, and cutting-edge technical challenges in TCM regulation, this consensus aims to explore the characteristics, progress, challenges, applicable pathways, and specific applications of network pharmacology as a new theory, method, and tool in TCM drug development. The goal is to enhance the quality of TCM drug research and development and accelerate the efficiency of developing new TCM products.

Delayed tooth extraction socket(TES) healing can cause failure of subsequent oral implantation and increase socioeconomic burden on patients. Excessive amounts of M1 macrophages, apoptotic neutrophils(ANs), and neutrophil extracellular traps(NETs) impair alveolar bone regeneration during TES healing. In the present study, we first discovered that conditioned medium(CM) collected from berberine-treated human bone marrow mesenchymal stem cells(BBR-HB-CM) accelerated TES healing. BBR-HB-CM contained bioactive materials that promoted the polarization of macrophages from M1 to M2, impeded the formation of ANs and NETs, and modulated M2 macrophage efferocytosis in vivo and in vitro. Mechanistically, BBR-HB-CM promoted bone formation by inhibiting macrophage-myofibroblast transition and reprogrammed macrophage polarization through p85/AKT/mTOR pathway-dependent autophagy. The 3-methyladenine abolished the therapeutic effects of BBR-HB-CM. Further studies revealed that BBR-HB-CM accelerated TES healing in rats with type 2 diabetes mellitus. Overall, our results demonstrated that BBR-HB-CM had high potential to promote rapid TES healing.

Current research indicates that beta cell loss in type 2 diabetes may be attributed to beta cell dedifferentiation rather than apoptosis;however,the mechanisms by which this occurs remain poorly *** previous study demonstrated that elevation of microRNA-24(miR-24)in a diabetic setting caused beta cell dysfunction and replicative *** this study,we focused on the role of miR-24 in beta cell apoptosis and dedifferentiation under endoplasmic reticulum(ER)stress *** found that miR-24 overabundance protected beta cells from thapsigargin-induced apoptosis at the cost of accelerating the impairment of glucosestimulated insulin secretion(GSIS)and enhancing the presence of dedifferentiation ***?Pathway Analysis(IPA)revealed that elevation of miR-24 had an inhibitory effect on XBP1 and ATF4,which are downstream effectors of two key branches of ER stress,by inhibiting its direct target,***,elevated miR-24 initiated another pathway that targeted Mafa and decreased GSIS function in surviving beta cells,thus guiding their dedifferentiation under ER stress *** results demonstrated that the elevated miR-24,to the utmost extent,preserves beta cell mass by inhibiting apoptosis and inducing *** study not only provides a novel mechanism by which miR-24 dominates beta cell turnover under persistent metabolic stress but also offers a therapeutic consideration for treating diabetes by inducing dedifferentiated beta cells to re-differentiation.
