Expression changes of nerve cell adhesion molecules L1 and semaphorin 3A after peripheral nerve injury

The expression of nerve cell adhesion molecule L1 in the neuronal growth cone of the central nervous system is strongly associated with the direction of growth of the axon, but its role in the regeneration of the peripheral nerve is still unknown. This study explored the problem in a femoral nerve section model in rats. L1 and semaphorin 3A m RNA and protein expressions were measured over the 4-week recovery period. Quantitative polymerase chain reaction showed that nerve cell adhesion molecule L1 expression was higher in the sensory nerves than in motor nerves at 2 weeks after injury, but vice versa for the expression of semaphorin 3A. Western blot assay results demonstrated that nerve cell adhesion molecule L1 expression was higher in motor nerves than in the sensory nerves at the proximal end after injury, but its expression was greater in the sensory nerves at 2 weeks. Semaphorin 3A expression was higher in the motor nerves than in the sensory nerves at 3 days and 1 week after injury. Nerve cell adhesion molecule L1 and semaphorin 3A expressions at the distal end were higher in the motor nerves than in the sensory nerves at 3 days, 1 and 2 weeks. Immunohistochemical staining results showed that nerve cell adhesion molecule L1 expression at the proximal end was greater in the sensory nerves than in the motor nerves; semaphorin 3A expression was higher in the motor nerves than in the sensory nerves at 2 weeks after injury. Taken together, these results indicated that nerve cell adhesion molecules L1 and semaphorin 3A exhibited different expression patterns at the proximal and distal ends of sensory and motor nerves, and play a coordinating role in neural chemotaxis regeneration.

Effects of acetylcysteine on micro-inflammation and pulmonary ventilation in chronic obstructive pulmonary disease exacerbation

BACKGROUND Acute exacerbation of chronic obstructive pulmonary disease(AECOPD)is a serious complication of chronic obstructive pulmonary disease,often characterized by increased morbidity and mortality.In traditional Chinese medicine,AECOPD is linked to phlegm-heat and blood-stasis,presenting symptoms like thick sputum,fever,and chest pain.It has been shown that acetylcysteine inhalation in conjunction with conventional therapy significantly reduced inflammatory markers and improved lung function parameters in patients with AECOPD,suggesting that acetylcysteine may be an important adjunctive therapy for patients with phlegm-heat-blood stasis type AECOPD.AIM To investigate the effect of acetylcysteine on microinflammation and lung ventilation in patients with phlegm-heat and blood-stasis-type AECOPD.METHODS One hundred patients with phlegm-heat and blood-stasis-type AECOPD were randomly assigned to two groups.The treatment group received acetylcysteine inhalation(10%solution,5 mL,twice daily)along with conventional therapy,whereas the control group received only conventional therapy.The treatment duration was 14 d.Inflammatory markers(C-reactive protein,interleukin-6,and tumor necrosis factor-alpha)in the serum and sputum as well as lung function parameters(forced expiratory volume in one second,forced vital capacity,and peak expiratory flow)were assessed pre-and post-treatment.Acetylcysteine inhalation led to significant reductions in inflammatory markers and improvements in lung function parameters compared to those in the control group(Pblood-stasis-type AECOPD.RESULTS Acetylcysteine inhalation significantly reduced inflammatory markers in the serum and sputum and improved lung ventilation function parameters in patients with phlegm-heat and blood-stasis type AECOPD compared with the control group.These differences were statistically significant(P<0.05).The study concluded that acetylcysteine i

What is already known about this topic?Allergic diseases have affected an estimated 40%of the population in China.However,our understanding of the full spectrum of these diseases remains insufficient.What is added by this report?Between 2018 and 2021,Yichang City documented 625,929 outpatient visits mainly related to skin and mucous membrane allergies(77.90%)and allergic respiratory conditions(19.64%).In 2021,the occurrence of outpatient visits for conditions such as allergic rhinitis,acute atopic conjunctivitis,and atopic dermatitis increased.The demographic analysis revealed that male patients comprised the majority of the under 18 age bracket(56.05%),while female patients were predominantly represented in the 18 to 65 age bracket(61.79%).What are the implications for public health practice?This constitutes the first analysis of the spectrum of allergic diseases,utilizing regional outpatient data,which has substantial implications for understanding the disease burden.

Biological characteristics of dynamic expression of nerve regeneration related growth factors in dorsal root ganglia after peripheral nerve injury

The regenerative capacity of peripheral nerves is limited after nerve injury.A number of growth factors modulate many cellular behaviors,such as proliferation and migration,and may contribute to nerve repair and regeneration.Our previous study observed the dynamic changes of genes in L4–6 dorsal root ganglion after rat sciatic nerve crush using transcriptome sequencing.Our current study focused on upstream growth factors and found that a total of 19 upstream growth factors were dysregulated in dorsal root ganglions at 3,9 hours,1,4,or 7 days after nerve crush,compared with the 0 hour control.Thirty-six rat models of sciatic nerve crush injury were prepared as described previously.Then,they were divided into six groups to measure the expression changes of representative genes at 0,3,9 hours,1,4 or 7 days post crush.Our current study measured the expression levels of representative upstream growth factors,including nerve growth factor,brain-derived neurotrophic factor,fibroblast growth factor 2 and amphiregulin genes,and explored critical signaling pathways and biological process through bioinformatic analysis.Our data revealed that many of these dysregulated upstream growth factors,including nerve growth factor,brain-derived neurotrophic factor,fibroblast growth factor 2 and amphiregulin,participated in tissue remodeling and axon growth-related biological processes Therefore,the experiment described the expression pattern of upstream growth factors in the dorsal root ganglia after peripheral nerve injury.Bioinformatic analysis revealed growth factors that may promote repair and regeneration of damaged peripheral nerves.All animal surgery procedures were performed in accordance with Institutional Animal Care Guidelines of Nantong University and ethically approved by the Administration Committee of Experimental Animals,China(approval No.20170302-017)on March 2,2017.

Blockade of angiopoietin-2/Tie2 signaling pathway specifically promotes inflammation-induced angiogenesis in mouse cornea

AIM： To investigate angiopoietin-2 （Ang-2）/Tie2 signaling pathway involving in inflammatory angiogenesis. METHODS： Three interrupted 11-0 nylon sutures were placed into the corneal stroma of BALB/c mice （6wk old） to induce inflammatory neovascularization. Expression of Ang-2 and Tie2 protein on neovascularization were examined by immunofluorescence. The dynamic expression of Ang-2 mRNA on neovascularization was examined by quantitative real-time reverse transcriptase-polymerase chain reaction （RT-PCR）. Finally, the mouse model of suture- induced corneal neovascularization was used to assess the role of Ang-2/Tie2 signaling pathway in inflammatory angiogenesis by systemic application of L1-10, an Ang-2 specific inhibitor. Mouse corneal hemangiogenesis were evaluated by whole mount immunofluorescence. RESULTS： Both Ang-2 and Tie2 were expressed on newly generated blood vessels in inflammatory cornea. Ang-2 expression was gradually upregulated around 2wk following injury, which was concurrent with an increased number of blood vessels. Blockade of Ang-2/Tie2 signaling pathway obviously promoted angiogenesis in inflammatory cornea. CONCLUSION： Ang-2/Tie2 signaling pathway seems to play an important role during angiogenesis in inflammatory cornea. This may open new therapeutic applications in pathological processes such as corneal graft survival, wound healing and carcinogenesis.

Sequencing analysis of matrix metalloproteinase 7-induced genetic changes in Schwann cells

Previous research revealed the positive activity of matrix metalloproteinase 7(MMP7) on migration and myelin regeneration of Schwa nn cells(SCs). However, understanding of the molecular changes and biological activities induced by increased amounts of MMP7 in SCs remains limited. To better understand the underlying molecular events, primary SCs were isolated from the sciatic nerve stump of newborn rats and cultured with 10 nM human MMP7 for 24 hours. The results of genetic testing were analyzed at a relatively relaxed threshold value(fold change ≥ 1.5 and P-value < 0.05). Upon MMP7 exposure, 149 genes were found to be upregulated in SCs, whereas 133 genes were downregulated. Gene Ontology analysis suggested that many differentially expressed molecules were related to cellular processes, single-organism processes, and metabolic processes. Kyoto Enrichment of Genes and Genomes pathway analysis further indicated the critical involvement of cell signaling and metabolism in MMP7-induced molecular regulation of SCs. Results of Ingenuity Pathway Analysis(IPA) also revealed that MMP7 regulates biological processes, molecular functions, cellular components, diseases and functions, biosynthesis, material metabolism, cell movement, and axon guidance. The outcomes of further analysis will deepen our comprehension of MMP7-induced biological changes in SCs. This study was approved by the Laboratory Animal Ethics Committee of Nantong University, China(approval No. 20190225-004) on February 27, 2019.

High mobility group box 1(HMGB1) interacts with pattern-recognition receptors of immune cells to activate the inflammatory response. Astrocytes play a positive role in the inflammatory response of the central nervous system by expressing a broad range of patternrecognition receptors. However, the underlying relationship between HMGB1 and the inflammatory reaction of astrocytes remains unclear. In this study, we established rat models of spinal cord injury via laminectomy at the T8–10 level, and the injured spinal cord was subjected to transcriptome sequencing. Our results showed that the HMGB1/Toll-like receptor 4(TLR4) axis was involved in the activation of astrocyte inflammatory response through regulation of cyclooxygenase 2(COX2)/prostaglandin E2(PGE2) signaling. Both TLR4 and COX2 were distributed in astrocytes and showed elevated protein levels following spinal cord injury. Stimulation of primary astrocytes with recombinant HMGB1 showed that COX2 and microsomal PGE synthase(mPGES)-1, rather than COX1, mPGES-2, or cytosolic PGE synthase, were significantly upregulated. Accordingly, PGE2 production in astrocytes was remarkably increased in response to recombinant HMGB1 challenges. Pharmacologic blockade of TLR2/4 attenuated HMGB1-mediated activation of the COX2/PGE2 pathway. Interestingly, HMGB1 did not impact the production of tumor necrosis factor-α or interleukin-1β in astrocytes. Our results suggest that HMGB1 mediates the astrocyte inflammatory response through regulating the COX2/PGE2 signaling pathway. 目的调查行维持性血液透析的终末期肾病患者姑息照护需求和症状负担的现况,并比较老年与非老年患者姑息照护需求和症状负担的差异。方法采用横断面调查法,选取2021年11月至2022年6月南京市2所三级医院血透中心的患者作为研究对象,采用一般资料调查表、姑息照护结局量表(Palliative Care Outcome Scale,POS)、透析症状指数(Dialysis Symptom Index,DSI)、欧洲五维健康量表(The European Quality of Life 5-Dimensions 3-Levels questionnaire,EQ-5D-3L)进行资料收集。采用描述性统计方法、组间比较和相关分析等进行资料分析。结果本研究共纳入236例患者,其中非老年和老年患者各118例。结果显示维持性血液透析患者POS中位数得分为16.0(12.0,23.0),老年患者得分更高(P<0.01)。患者平均症状数目为15.04±5.06,症状严重程度的中位数得分为59.0(52.0,71.0),老年患者得分更高(P<0.01)。患者最常见的症状是口干(91.5%),其次是瘙痒(83.1%)和皮肤干燥(82.2%)。此外,姑息照护需求与症状负担和生活质量之间存在显著相关性(P<0.01)。结论行维持性血液透析的终末期肾病患者症状负担严重并有中等程度的姑息照护需求,且在老年患者中尤为明显。未来需组建跨学科的姑息照护团队,除了积极管理患者的症状外,还需满足与姑息照护相关的生理、心理、社会和精神需求,以改善患者的生活质量。

The interaction effect of grip strength and lung function (especially FVC) on cardiovascular diseases: a prospective cohort study in jiangsu province, China

OBJECTIVE Lung function and grip strength(GS)are associated with cardiovascular disease(CVD),but whether these risk factors interact to affect CVD is unknown.This study aimed to explore the interactions between lung function and GS with major CVD(defined as fatal/non-fatal myocardial infarction,stroke,and heart failure)incidence.METHODS We conducted a prospective cohort study on the Chinese population in jiangsu province.Cox proportional haz-ards models were used to explore the associations between GS,lung function,and major CVD incidence.RESULTS A total of 5967 participants were included in our study;among them,182 participants developed major CVD.Parti-cipants with low forced vital capacity(FVC)had a higher risk of major CVD(hazard ratio(HR)=1.45;95%confidence interval(CI):1.05-2.01;P<0.05)compared with normal FVC.The risk of major CVD incidence(HR=0.54;95%CI:0.35-0.83;P<0.01)was significantly lower in participants with high GS than in individuals with low GS.The interaction between FVC and GS for major CVD incidence(P=0.006)was statistically significant.Compared with normal FVC participants with high GS,low FVC parti-cipants with low GS had the highest risk of major CVD incidence(HR=2.50;95%CI:1.43-4.36;P<0.01).CONCLUSION Among people with low FVC,the risk of major CVD is lower with high GS.Participants with low FVC and low GS have the highest risk of major CVD.Therefore,more attention should be paid to the incidence of major CVD in individu-als with low FVC,especially those who have lower GS.
