Downregulation of Serum PTEN Expression in Mercury-Exposed Population and PI3K/AKT Pathway-Induced Inflammation

Objective This study investigated the impact of occupational mercury(Hg) exposure on human gene transcription and expression, and its potential biological *** Differentially expressed genes related to Hg exposure were identified and validated using gene expression microarray analysis and extended validation. Hg-exposed cell models and PTEN lowexpression models were established in vitro using 293T cells. PTEN gene expression was assessed using qRT-PCR, and Western blotting was used to measure PTEN, AKT, and PI3K protein levels. IL-6 expression was determined by *** Combined findings from gene expression microarray analysis, bioinformatics, and population expansion validation indicated significant downregulation of the PTEN gene in the high-concentration Hg exposure group. In the Hg-exposed cell model(25 and 10 μmol/L), a significant decrease in PTEN expression was observed, accompanied by a significant increase in PI3K, AKT, and IL-6 ***, a low-expression cell model demonstrated that PTEN gene knockdown led to a significant decrease in PTEN protein expression and a substantial increase in PI3K, AKT, and IL-6 *** This is the first study to report that Hg exposure downregulates the PTEN gene, activates the PI3K/AKT regulatory pathway, and increases the expression of inflammatory factors, ultimately resulting in kidney inflammation.

Introduction:In order to enhance the effectiveness of resource allocation,regions must tailor their responses to their specific epidemiological and economic ***:Utilizing Spectrum software,we projected the cost-effectiveness of 10 chosen HIV interventions in six cities in eastern China from 2019 to *** assessed three scenarios—Base,Achievable,and Idealized—for each *** analysis included the projected number of HIV infections and deaths averted,as well as the incremental costeffectiveness ratios for each intervention in the six ***:In Shijiazhuang,Wuxi,Yantai,and Zhenjiang,cities with initially low antiretroviral therapy(ART)coverage,ART showed significant effectiveness,especially for ***,in Foshan and Ningbo,where ART coverage was notably high,oral pre-exposure prophylaxis(PrEP)for men who have sex with men(MSM)proved effective in the Idealized *** outreach,ART for males,and ART for females demonstrated cost-effectiveness across all six cities in both Achievable and Idealized scenarios at the predefined thresholds for each ***:Maintaining an appropriate coverage rate for outreach to MSM can lead to *** cities with low ART coverage,scaling up ART remains a crucial *** regions with high ART coverage,consideration may be given to the utilization of oral PrEP for MSM individuals,requiring budget allocation.

Introduction:Newer skin tests,including the ESAT6-CFP10(EC)skin test,were recommended for diagnosing Mycobacterium tuberculosis(***)***,no data exist assessing the diagnostic performance of the EC skin test among foreign students with different skin ***:A cohort study at Nanjing Medical University screened incoming foreign *** EC skin test was used to assess for *** infection,and results were read at 24,48,72,and 96-hours ***:Among 96 participants,*** infection rates at 24,48,72,and 96-hours post-injection were 3.13%,7.29%,13.54%,and 9.38%,*** infection rates were lower among individuals with darker skin tones,the difference was not statistically significant(P=0.186),and variations were consistent across different measurement *** analysis revealed 5.3%in the continuousincreasing group,86.5%in the low-stable group,and 5.2%in the elevated-decreasing ***,participants in the elevated-decreasing group had lighter skin tones,with trajectory patterns consistent across different skin ***:The EC skin test is safe,and redness diameter is a more reliable indicator than *** should be collected within 48 to 72 hours,with verification at 72 hours crucial if initial results are ***,skin color does not affect EC skin test outcomes.

Stability and variability of molecular subtypes: comparative analysis of primary and metastatic triple-negative breast cancer

Objective: Triple-negative breast cancer(TNBC) is a heterogeneous and aggressive cancer. Although our previous study classified primary TNBC into four subtypes, comprehensive longitudinal investigations are ***: We assembled a large-scale, real-world cohort comprised of 880 TNBC patients [465 early-stage TNBC(e TNBC) and 415 metastatic TNBC(m TNBC) patients] who were treated at Fudan University Shanghai Cancer center. The longitudinal dynamics of TNBC subtypes during disease progression were elucidated in this patient cohort. Comprehensive analysis was performed to compare primary and metastatic lesions within specific TNBC ***: The recurrence and metastasis rates within 3 years after initial diagnosis in the e TNBC cohort were 10.1%(47/465). The median overall survival(OS) in the m TNBC cohort was 27.2 months [95% confidence interval(CI), 24.4±30.2 months], which indicated a poor prognosis. The prognostic significance of the original molecular subtypes in both e TNBC and m TNBC patients was confirmed. Consistent molecular subtypes were maintained in 77.5% of the patients throughout disease progression with the mesenchymal-like(MES) subtype demonstrating a tendency for subtype transition and brain metastasis. Additionally, a precision treatment strategy based on the metastatic MES subtype of target lesions resulted in improved progression-free survival in the FUTURE ***: Our longitudinal study comprehensively revealed the clinical characteristics and survival of patients with the original TNBC subtypes and validated the consistency of most molecular subtypes throughout disease progression. However, we emphasize the major importance of repeat pathologic confirmation of the MES subtype.

Objective This study aimed to evaluate the associations of serum folate and/or vitamin B12 concentrations with obesity among Chinese children and *** A cross-sectional study was conducted including 3,079 Chinese children and adolescents,aged 6 to 17 years,from jiangsu,*** indices,such as,children's body mass index(BMI),BMI z-scores,waist circumference,and waist-to-height ratio were *** linear regression and generalized additive models were used to investigate the associations of serum folate and vitamin B12 levels with anthropometric indices and odds of *** We observed that serum vitamin B12 concentrations were inversely associated with all anthropometric indices and the odds of general obesity[odds ratio(OR)=0.68;95%confidence interval(CI)=0.59,0.78]and abdominal obesity(OR=0.68;95%CI=0.60,0.77).When compared to participants with both serum vitamin levels in the two middle quartiles,those with both serum folate and vitamin B12 levels in the highest quartile were less prone to general(OR=0.31,95%CI=0.19,0.50)or abdominal obesity(OR=0.46,95%CI=0.31,0.67).Conversely,participants with vitamin B12 levels in the lowest quartile alongside folate levels in the highest quartile had higher odds of abdominal obesity(OR=2.06,95%CI=1.09,3.91).Conclusion Higher serum vitamin B12 concentrations,but not serum folate concentrations,were associated with lower odds of childhood *** and adolescents with high levels of vitamin B12 and folate were less likely to be obese.
