In this study, the safety of Levilactobacillus brevis CGMCC1.5954 was evaluated, which has probiotic properties, through both in vitro methods such as antibiotic susceptibility testing, biogenic amine testing, and virulence gene analysis, and in vivo methods including biochemical testing, routine blood index testing, and bacterial displacement capacity analysis. The results of HPLC analysis revealed that during incubation, L. brevis CGMCC1.5954 did not secrete cadaverine, histamine, putrescine, or tyramine. It exhibited sensitivity to tetracyclines, macrolides, cephalosporins, & beta;-lactamase inhibitor complex, and other antibiotics, but not to aminoglycosides and quinolones, which is consistent with the behavior of other lactic acid bacteria. Its genome encodes a total of 2314 genes, including 1743 proteins and 109 signal peptides. Notably, no virulence genes were found in this genome. Additionally, the gene encoding the production of & gamma;-aminobutyric acid was found in it. In vivo experiments indicated that gavage feeding of L. brevis CGMCC1.5954 for 28 d had no significant effect on normal growth, blood hemoglobin level, erythrocytes, white blood cells, and other routine blood parameters in mice. Furthermore, there was no migration of it observed in the liver, kidney, spleen, or blood during the feeding trial, which indicated that it did not cause any infections. Moreover, H & E staining showed no evidence of lesion damage to the liver, kidney, or other organs. To sum up, L. brevis CGMCC1.5954 is considered safe for producing fermented foods, especially those that contain high levels of & gamma;-aminobutyric acid.

Polygonum cuspidatum (P. cuspidatum) is an herbaceous perennial plant known as Asian knotweed that is used in Chinese medicine due to its active components. Recent studies of the uric acid-lowering activity of P. cuspidatum extracts have mainly focused on analysis of its crude extract or specific chemical components, with limited research about its uric acid-lowering activity in vivo. In present study, monomeric compounds with higher uric acid-lowering ability were isolated from P. cuspidatum extract by column chromatography and thin-layer chro-matography (TLC) and tested in a hyperuricemia mouse model and xanthine oxidase (XOD) inhibition assay in vitro. Structural characterization of the isolated components was performed by nuclear magnetic resonance (NMR) spectroscopy and the potential mechanism of decreasing uric acid was analyzed using untargeted metabolomics. Two main novel compounds were isolated and identified, 1-(4-hydroxy-2-methoxyphenyl)-2-(4-hydroxy-3,5-dimethylphenyl)butane-1,2,3-triol (compound 1) and 1-(4-hydroxy-2-methoxyphenyl)-2-(4-hy-droxy-3,5-dimethylphenyl)-3-methylbutane-1,2-diol (compound 2), and compounds 1 was proved more signif-icant inhibitory effect on the activity of XOD (p < 0.01) with inhibition of 49.80%. Metabolomics analysis indicated that the fractions (compound 1 and compound 2) affected hyperuricemic mice mainly through changes in galactose metabolism (p = 5.74E-4) and the effects were closely related to metabolic pathways of organic acid, carbohydrate, and lipid metabolism. These results contribute to providing theoretical support for the utilization of P. cuspidatum as a food and medicinal material, which can increase its use as an alternative treatment for hyperuricemia.

Citral, a natural antimicrobial agent, has been found to induce severe membrane damage resulting in bacterial death. Despite its effectiveness, there is a lack of in-depth research on its interaction with bacterial cells, especially at low concentrations. This work aimed to investigate the impact of citral concentration on its antibacterial effect against Staphylococcus aureus (S. aureus) and to analyze the relevance between membrane damage and cell death. In this study, the minimal inhibitory concentration of citral against S. aureus was measured at 0.25 mg/ mL. At a concentration of 0.5 mg/mL, citral significantly disturbed the cell membrane fluidity and permeability, leading to the leakage of intracellular macromolecules and rapid cell death. However, when the concentration was decreased to 0.25 mg/mL, the membrane damage was greatly reduced, and a complex biological process between citral and membrane components was found. Under continuous drug exposure, the cell membrane underwent gradual rigidification, concomitant with the loss of membrane function, ultimately causing cell death. Despite the fact that no notable disturbance to membrane functions was discovered, the presence of 0.125 mg/ mL citral could still weaken the metabolic vitality and growth of S. aureus. Therefore, it was suggested that there could be multiple mechanisms of citral action on S. aureus cells, with drug concentration being a critical factor affecting the mode of action. Overall, the findings of this study provide valuable insights into the antibacterial mechanisms of natural compounds and broaden the potential applications of citral as a natural antimicrobial.

Dear Editor Genetic recombination results in novel allele combinations and is the most important genetic phenomenon for generating variation in breeding populations. However, in most organisms, the number of genetic recombinations is tightly constrained, which hampers the introgression of new genetic traits and limits genetic variation during agricultural breeding （Barakate et al., 2014）. Therefore, the possibility of increasing genetic recombination frequency has attracted much interest from crop breeders because of the obvious practical applications in traditional breeding （Leflon et al., 2010; Crismani et al., 2012）. However, to date, the goal of increasing the genetic recombination frequency has not been achieved in crop plants.

Gene mapping of a new coat mutation in mouse

Gene mapping of a mouse coat mutation has been investigated. First, 100 10-bp random primers were used to amplify DNA, but the mutation could not be located by this method because there were no correlation between the amplified products and coat phenotypes. Second, by using ldh1 , Car2, Mup1 , Pgb1 , Hbb, Es1O, Es1, Mod1, Gdc1 , Ce2, Es3 as genetic markers, linkage test crosses (two-point test) consisting of intercrossing uncovered BALB/c mice (homozygotes) to CBA/N and C57BL/6 mice with normal hair and backcrossing the heterozygotes of the F1 to the uncovered BALB/c mice were made. It was soon evident that the mutation was linked to Es3 on chromosome 11. Furthermore, three-point test was made by using Es3 and D11 Mit8 (a microsatellite DNA) as genetic markers. The result showed that the mutation was linked to Es3 with the percentage recombination of (7.89 ± 2.19) %, and linked to D11Mit8 with the percentage recombination of (26.30 ± 3.57)%. BACKGROUND: Pancreatic cancer is one of the most common malignant tumors. Early diagnosis of pancreatic cancer is difficult because of the latent onset and lack of good biomarkers. This study aimed to look for and identify differentially expressed proteins in tissues of pancreatic cancer and adjacent noncancerous tissues by proteomic approaches so as to provide information about possible pancreatic cancer markers and therapeutic targets. METHODS: Proteins extracted front 3 paired adjacent noncancerous and cancerous pancreatic tissue specimens were separated by two-dimensional gel electrophoresis (2-DE). The protein spots exhibiting statistical alternations between the two groups through computerized image analysis were then identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS). In addition, Western blotting and immunohistochemistry were performed to verify the expression of certain candidate proteins. RESULTS: Twelve proteins were significantly upregulated and 4 were downregulated between cancerous and paired adjacent noncancerous pancreatic tissues. Several proteins (S100A11, Ig gamma-l chain C region, GSTO1 and peroxiredoxin 4) were found for the first time to be associated with pancreatic cancer. Differential expression of some identified proteins was further confirmed by Western blotting analysis and/or immunohistochemical analysis. CONCLUSIONS: Comparative proteomic analysis using 2-DE and MALDI-TOF-MS is an effective method for identifying differentially expressed proteins that may be the potential diagnostic biomarkers and therapeutic targets for pancreatic cancer.

Salt marsh-tidal creek systems as a coastal geomorphological unit represent an important natural resource. The present study on jiangsu salt marshes, eastern China, shows that variations in tidal current velocities in salt marsh creeks are controlled by the local tidal wave characteristics and the bed slope and elevation of the salt marshes and creeks. Likewise, the tidal currents modify the geomorphology of the salt marsh-tidal creek systems by transporting sediments and causing erosion/deposition. Storm events, which appear to have cyclical changes in their intensity relating to sunspot activities, can affect the geomorphic evolution of such systems. Further, in response to accelerated sea-level rise, accretional rates on salt marshes may increase. The tidal creeks have the function of transporting water and sediment onto the salt marsh surface; further, the energy of tidal currents and waves are dissipated within the salt marsh-tidal creek system. This study investigated the effects of highland barley's germination time (24, 48, and 72 h) on the thermomechanical, pasting, fermentation, dynamic rheological, molecular mobility, and protein structural properties of oat-wheat composite flour dough. Germination significantly reduced flour water absorption, freezable water, melting enthalpy, and increased protein weakening compared with native barley (NB) samples. In addition, germination decreased bound and immobilized water and increased the free water compared to the NBcomposite flour dough samples, according to the results of a low-field nuclear magnetic resonance spectrometer (LF-NMR). Germination also increased the protein's extractability and surface hydrophobicity while negatively impacting the starch-protein network;however, it decreased bread-specific volume. Germination for 24 h, on the other hand, had minor effects on dough and bread samples compared to 48 and 72 h. As a result, germination for 24 h under controlled conditions could be used to improve the rheological qualities of highland barley and avoid the adverse rheological effects of the prolonged germination period.
