Genetic identification, pathogenicity and patulin production of Penicillium species from apple blue mold in China

Objectives Blue mold is a common postharvest disease that leads to severe rot and patulin (PAT) contamination in fruits, which is a key factor endangering the storage and quality of apple products. However, the Penicillium species from apple blue mold in China and their pathogenicity and PAT production ability have not been well *** and Methods In this study, 62 strains of pathogenic Penicillium species were isolated from blue mold apples collected from different regions of China. The ITS, BenA, and CaM genes were sequenced for molecular *** Penicillium strains were identified as three species, 57 P. expansum, 3 P. crustosum, and 2 P. polonicum. Twelve variants of the ITS sequence, 61 variants of BenA and 67 variants of CiaM genes were identified between Penicillium species. Fungal morphology was observed on potato dextrose agar (PDA). After 10 d of cultivation, P. expansum cultures were detected with PAT concentrations ranging from 457.98 to 1333.85 mg/kg, and two P. polonicum strains were 6.02 mg/kg and 6.30 mg/kg. The three P. crustosum strains could not produce PAT on PDA. In apple inoculation, P. expansum strains showed significantly faster infection rates than P. crustosum and P. polonicum strains. All P. expansum strains can produce PAT during infection, and the concentrations in the rot apple tissues were 2.31-88.67 mg/kg. Nodulation and molecular characterization of pure culturesisolated from root nodules of Coriaria nepalensis

Four strains with typical morphology of actinomycete genus of Frankia were isolated from root nodules of Coriaria nepalensis. They were shown to nodulate the seedlings of host plant and hybridize with Frankia 16S rRNA targeted oligonucleotide probes, indicating that they did belong to the genus Frankia. Furthermore, by nifHDK probe hybridizations, the homologous fragments of nifHDK genes were detected among the bacteria, and they were located in various sizes of restriction fragments of total DNA, showing diverse patterns of restriction fragment length polymorphisms of nifHDK gene (nifHDK_RFLPs). The PCR_based amplification and cloning of nifH gene throw light on the molecular phylogeny of Coriaria_infective Frankia.

Gene expression profiling related to powdery mildew resistance in wheat with the method of suppression subtractive hybridization

'Bainong 3217 × Mardler' BC5F4 wheat line at the initial stage of inoculation with powdery mildew pathogen (Erysiphe graminis DC) was used to construct a suppression subtractive hybridization (SSH) cDNA library. Totally 760 ESTs were obtained through sequencing. Similarity analysis of ESTs based on BLASTn and BLASTx with the sequences in GenBank, in combination with macroarray differential screening, revealed that 199 ESTs of 65 kinds were known to be functionally disease resistance related. Based on the gene expression profiling in the present study, it is postulated that salicylic acid (SA) and MAP-related signal transduction pathways were involved in powdery mildew resistance in wheat. System acquired resistance genes were predominant in terms of kinds and quantity. With the initiation of cell defense reaction, the genes conferring anti-oxidation substances were largely expressed and thus cell protection mechanism was activated. Germination of embryo in soybean anther culture

A large number of experiments about induction of callus, embryo and germination of regeneration plants were done in soybean anther culture. Screening for genotypes, low_temperature pretreatment for floral buds, constituent of hormone in medium and effects of different temperatures on induction rate of callus and embryo were studied. It was found that the TDZ played an important role in embryo induction and germination. 28 genotypes had reaction competent to the anther culture in all 110 genotypes. Induction rate of callus was 26%-68.1%. Plant differentiation rate was 0.5%-2.0%.

Wild species of domesticated crops provide valuable genetic resources for resistance breeding. Prunus davidiana, a wild relative of peach with high heterozygosity and diverse stress tolerance, exhibits high resistance against aphids. However, the highly heterozygous genome of P. davidiana makes determining the underlying factors influencing resistance traits challenging. Here, we present the 501.7 Mb haplotype-resolved genome assembly of P. davidiana. Genomic comparisons of the two haplotypes revealed 18,152 structural variations, 2,699 Pda_hap1-specific and 2,702 Pda_hap2-specific genes, and 1,118 allele-specific expressed genes. Genome composition indicated 4.1% of the P. davidiana genome was non-peach origin, out of which 94.5% was derived from almond. Based on the haplotype genome, the aphid resistance quantitative trait locus (QTL) was mapped at the end of Pda03. From the aphid resistance QTL, PdaWRKY4 was identified as the major dominant gene, with a 9-bp deletion in its promoter of the resistant phenotype. Specifically, PdaWRKY4 regulates aphid resistance by promoting PdaCYP716A1-mediated anti-aphid metabolite betulin biosynthesis. Moreover, we employed a genome design to develop a breeding workflow for rapidly and precisely producing aphid-resistant peaches. In conclusion, this study identifies a novel aphid resistance gene and provides insights into genome design for the development of resistant fruit cultivars.

Construction of lipopeptide mono-producing Bacillus strains and comparison of their antimicrobial activity

Bacillus amyloliquefaciens generally secretes a mixture of multiple cyclic lipopeptides, including iturin, surfactin, and plipastatin, and it is challenging to discriminate each cyclic lipopeptide's antimicrobial activity directly. In this study, a chassis strain Bacillus subtilis 1A751 WR without cyclic lipopeptides operons was constructed by CRISPR/Cas9. The iturin gene cluster of B. amyloliquefaciens HYM-12 was captured and assembled with sfp and degQ genes using transformation-associated recombination (TAR). It was then integrated into the chassis strain to obtain an iturin mono-producing strain that could synthesize six iturin isoforms. The frameshift sfp gene of strain 1A751 & UDelta;pps was repaired by a scarless gene knock-in method to generate a surfactin mono-producing strain. Antimicrobial activity experiments indicated that the extracts of iturin, surfactin, and our previously constructed plipastatin mono-producing strains exhibited powerful inhibitory effects toward pathogenic bacteria. Interestingly, iturin showed significant antifungal activity, but surfactin and plipastatin exhibited weak antifungal activity. Compared with amphotericin B, iturin has a solid and durable antifungal activity. This study demonstrates that TAR cloning is efficient for cloning large gene clusters, proves the differences in the antimicrobial activity of three cyclic lipopeptides, and lays a theoretical basis for further elucidating their antimicrobial mechanism.

Seasonal dynamics of CO_2 fluxes from subtropical plantation coniferous ecosystem

As one component of ChinaFLUX, the measurement of CO2 flux using eddy covariance over subtropical planted coniferous ecosystem in Qianyanzhou was conducted for a long term. This paper discusses the seasonal dynamics of net ecosystem exchange (NEE), ecosystem respiration (RE) and gross ecosystem exchange (GEE) between the coniferous ecosystem and atmosphere along 2003 and 2004. The variations of NEE, RE and GEE show obvious seasonal variabilities and correlate to each other, i.e. lower in winter and drought season, but higher in summer; light, temperature and soil water content are the main factors determining NEE; air temperature and water vapor pressure deficit (VPD) influence NEE with stronger influence from VPD. Under the proper light condition, drought stress could decrease the temperature range for carbon capture in planted coniferous, air temperature and precipitation controlled RE; The NEE, RE, and GEE for planted coniferous in Qianyanzhou are -387.2 gC·m-2 a-1,1223.3 g C·m-2 a-1, -1610.4 g C·m-2 a-1 in 2003 and -423.8 g C·m-2 a-1, 1442.0 g C·m-2 a-1, -1865.8 g C·m-2 a-1 in 2004, respectively, which suggest the intensive ability of plantation coniferous forest on carbon absorbing in Qianyanzhou.

Effects of ultradry storage on fluidity of plasma membrane of seed

The effects of ultradry seed storage on the fluidity of plasma membrane have been investigated using the DPH fluorescent probe (1,6-diphenyi-1, 3, 5,-hexatriene). The results demonstrated that the micro-viscosity of plasma membrane of ultradried seeds has no significant changes compared with the seeds which were stored under -20℃ condition. However, there is a little adverse effect on the seeds with extreme dehydration. The results were consistent with higher vigor level of ultradried seed. It indicated that ultradry seed storage could maintain the physiological function of seed, protect the integrity of the membrane and improve the storability of seed. Furthermore, the results also revealed that sugar has the effect on protecting membrane structure and preserving the fluidity of the plasma membrane under seed dehydration. In the meantime, some mechanism about ultradry seed storage and the tolerance of dehydration of seed have been discussed.

Tartary buckwheat （Fagopyrum tataricum） is an important pseudocereal crop that is strongly adapted to growth in adverse environments. Its gluten-free grain contains complete proteins with a well-balanced composition of essential amino acids and is a rich source of beneficial phytochemicals that provide significant health benefits. Here, we report a high-quality, chromosome-scale Tartary buckwheat genome sequence of- 489.3 Mb that is assembled by combining whole-genome shotgun sequencing of both Illumina short reads and single-molecule real-time long reads, sequence tags of a large DNA insert fosmid library, Hi-C sequencing data, and BioNano genome maps. We annotated 33 366 high-confidence protein-coding genes based on expression evidence. Comparisons of the intra-genome with the sugar beet genome revealed an independent whole-genome duplication that occurred in the buckwheat lineage after they diverged from the common ancestor, which was not shared with rosids or asterids. The reference genome facilitated the identification of many new genes predicted to be involved in rutin biosynthesis and regulation, aluminum stress resistance, and in drought and cold stress responses. Our data suggest that Tartary buckwheat＇s ability to tolerate high levels of abiotic stress is attributed to the expansion of several gene families involved in signal transduction, gene regulation, and membrane transport. The availability of these genomic resources will facilitate the discovery of agronomically and nutritionally important genes and genetic improvement of Tartary buckwheat.

Microdissection and amplification of the chromosome arm 5S in a rice telo-tetrasomic

The rice aneuploids with telochromosomes are ideal genetic stocks for chromosome arm identification and microdissection. A rice telo-tetrasomic line with two extra short arms of chromosome 5 (5S) was used in the present study. The arms of 5S were microdissected from the prometaphase cells in mitosis and amplified with linker adaptor PCR (LA-PCR). The amplified fragments, ranging from 200-3000 bp, were confirmed to be from the rice chromosome arm 5S by the rice STS and microsatellite markers.
