Reco.bination and repeat-induced point mutation landscapes reveal trade-offs between the sexual and asexual cycles of Magnaporthe oryzae

The fungal disease caused by Magnaporthe oryzae is one of the most devastating diseases that endanger many crops *** shows that sexual reproduction can be advantageous for fungal diseases as hybridization facilitates ***,the pervasive clonal lineages of *** observed in natural fields co.tradict this expectation.A better understanding of the roles of reco.bination and the fungi-specific repeat-induced point mutation(RIP)in shaping its evolutionary trajectory is essential to bridge this knowledge *** we systematically investigate the RIP and reco.bination landscapes in *** using a whole genome sequencing data from 252 population samples and 92 cross *** data reveal that the RIP can robustly capture the population history of ***,and we provide accurate estimations of the reco.bination and RIP rates across different *** ***,our results highlight a parent-of-origin bias in both reco.bination and RIP rates,tightly associating with their sexual potential and variations of effector *** bias suggests a critical trade-off between generating novel allelic co.binations in the sexual cycle to facilitate host-jumping and stimulating transposon-associated diversification of effectors in the asexual cycle to facilitate host *** findings provide unique insights into understanding the evolution of blast fungus.

Dammarane-type triterpenoids from Gynostemma longipes and their protective activities on hypoxia-induced injury in PC12 cells

Sixteen new dammarane-type triterpenoid saponins(1-16)featuring diverse structural variations in the side chain at C-17,along with twenty-one known analogues(17-37),have been isolated from the rhizomes of Gynostemma longipes ***,a plant renowned for its medicinal and edible *** structural elucidation of these co.pounds was acco.plished through co.prehensive analyses of 1D and 2D NMR and HRMS spectrosco.ic data,supplemented by co.parison with previously reported *** assays on the isolates for their protective effects against hypoxia-induced damage in pheochromocytoma cells(PC12 cells)revealed that nine saponins exhibited significant anti-hypoxic *** investigation into the anti-hypoxia mechanisms of the representative saponins demonstrated that co.pounds 22 and 36 markedly reduced the levels of hypoxia-induced ***,these co.pounds were found to decrease the release of lactate dehydrogenase(LDH)and malondialdehyde(MDA),while increasing the activity of superoxide dismutase(SOD),thereby indicating that the saponins co.ld mitigate hypoxia-induced injuries by ameliorating apoptosis and oxidative *** findings offer substantial evidence for the future utilization and development of ***,identifying dammarane-type triterpenoid saponins as its active anti-hypoxic co.stituents.

Differential metabolite analysis of the pharmaco.ynamic differences between different ratios of Dahuang(Radix Et Rhizoma Rhei Palmati)-Taoren(Semen Persicae) herb pair

OBJECTIVE: To explore the material basis of the difference of efficacy of Dahuang(Radix Et Rhizoma Rhei Palmati)-Taoren(Semen Persicae)(DT) drugs with different proportions. METHODS: Samples of different ratios of Dahuang(Radix et Rhizoma Rhei Palnati, DH) to Taoren(Semen Persicae, TR)(Group A 1∶1, B 2∶3, C 3∶2) were analyzed based on gas chromatography time-of-flight mass spectrometry untargeted metabolomics technique. RESULTS: A total of 240 primary metabolites were detected. Forty-one differential metabolites involved nine differential metabolic pathways, of which four were closely related to the efficacy of DT in the treatment of heat and blood stasis syndrome. These pathways included the biosynthesis of amino acid(phenylalanine tyrosine and tryptophan), flavonoids, unsaturated fatty acids, and the glyco.ysis/glyco.enesis pathway. co.CLUSION: There are significant differences in the efficacy of different ratios of DT drugs, and their optimal ratio for the treatment of heat and blood stasis syndrome should be 1∶1.

Overexpression of PbrGA2ox1 enhances pear drought tolerance through the regulation of GA_(3)-inhibited reactive oxygen species detoxification and abscisic acid signaling

Drought stress is a devastating natural disaster driven by the co.tinuing intensification of global warming,which seriously threatens the productivity and quality of several horticultural crops,including ***(GAs)play crucial roles in plant growth,development,and responses to drought *** studies have shown significant reductions of GA levels in plants under drought stress;however,our understanding of the intrinsic regulation mechanisms of GA-mediated drought stress in pear remains very ***,we show that drought stress can impair the accumulation of bioactive GAs(BGAs),and subsequently identified PbrGA2ox1 as a chloroplast-localized GA deactivation *** gene was significantly induced by drought stress and abscisic acid(ABA)treatment,but was suppressed by GA_(3)***2ox1-overexpressing transgenic tobacco.plants(Nico.iana benthamiana)exhibited enhanced tolerance to dehydration and drought stresses,whereas knock-down of PbrGA2ox1 in pear(Pyrus betulaefolia)by virus-induced gene silencing led to elevated drought *** plants were hypersensitive to ABA,and had a lower BGAs co.tent,enhanced reactive oxygen species(ROS)scavenging ability,and augmented ABA accumulation and signaling under drought stress co.pared to wild-type ***,the opposite effects were observed with PbrGA2ox1 silencing in ***,exogenous GA_(3)treatment aggravated the ROS toxic effect and restrained ABA synthesis and signaling,resulting in the co.promised drought tolerance of *** summary,our results shed light on the mechanism by which BGAs are eliminated in pear leaves under drought stress,providing further insights into the mechanism regulating the effects of GA on the drought tolerance of plants.

Identifying the geographical origin and processing technology of Moyao(Myrrh)on the basis of near-infrared spectrosco.y co.bined with chemometrics

OBJECTIVE:To evaluate the quality of Moyao(Myrrh)in the identification of the geographical origin and processing of the ***:Raw Moyao(Myrrh)and two kinds of Moyao(Myrrh)processed with vinegar from three co.ntries were identified using near-infrared(NIR)spectrosco.y co.bined with chemometric *** co.ponent analysis(PCA)was used to reduce the dimensionality of the data and visualize the clustering of samples from different categories.A classical chemometric algorithm(PLS-DA)and two machine learning algorithms[K-nearest neighbor(KNN)and support vector machine]were used to co.duct a classification analysis of the near-infrared spectra of the Moyao(Myrrh)samples,and their discriminative performance was ***:Based on the accuracy,precision,recall rate,and F1 value in each model,the results showed that the classical chemometric algorithm and the machine learning algorithm obtained positive *** all of the chemometric analyses,the NIR spectrum of Moyao(Myrrh)preprocessed by standard normal variation or Multivariate scattering co.rection co.bined with KNN achieved the highest accuracy in identifying the geographical origins,and the accuracy of identifying the processing technology established by the KNN method after first-order derivative pretreatment was the *** best accuracy of geographical origin discrimination and processing technology discrimination were 0.9853 and 0.9706 ***:NIR spectrosco.y co.bined with chemometric technology can be an important tool for tracking the origin and processing technology of Moyao(Myrrh)and can also provide a reference for evaluations of its quality and the clinical use.

Lung cancer is one of the most co.mon malignant tumors with the fastest increase in the incidence rate and *** after maximum tumor resection assistance with a radiotherapy and chemotherapy co.bination,the recurrence of non-small cell lung cancer is still *** addition,low targeting efficiency and poor permeability of drug delivery systems strongly affect the therapeutic efficiency of anti-cancer drugs on non-small cell lung *** we designed a gemcitabine(GEM)loaded arginineglycine-aspartic acid-cysteine(RGDc)-modified gold mineralization“hybrid nanozyme bomb”(RGTG)to overco.e those *** modification improved the active targeting of liposomes to the tumor tissues with the seco.d near-infrared(NIR-Ⅱ)-triggered gold-shell disruption and GEM *** co.lapsed gold-shell particles with a smaller size co.ld penetrate the tumor solid barrier and act as photothermal therapy(PTT)agents to improve PTT therapy and starvation therapy via generating gluco.ic acid and reactive oxygen species(ROS).Moreover,the resting reversal effect of gold particles on tumor fibroblasts can achieve accelerating tumor penetration of gold particles and *** to monotherapy,RGTG showed significant improvement in tumor inhibition,with a tumor volume reduction of 83%co.pared to the co.trol group,which provides a promising tumor treatment platform for non-small cell lung cancer(NSCLC).

The occurrence of benign prostate hyperplasia(BPH)was related to disrupted sex steroid hormones,and metformin(Met)had a clinical response to sex steroid hormone-related gynaeco.ogical ***,whether Met exerts an antiproliferative effect on BPH via sex steroid hormones remains ***,our clinical study showed that along with prostatic epithelial cell(PEC)proliferation,sex steroid hormones were dysregulated in the serum and prostate of BPH *** the major co.tributor to dysregulated sex steroid hormones,elevated dihydrotestosterone(DHT)had a significant positive relationship with the clinical characteristics of BPH *** of adenosine 5'-monophosphate(AMP)-activated protein kinase(AMPK)by Met restored dysregulated sex steroid hormone homeostasis and exerted antiproliferative effects against DHT-induced proliferation by inhibiting the formation of androgen receptor(AR)-mediated Yes-associated protein(YAP1)-TEA domain transcription factor(TEAD4)***’s anti-proliferative effects were blocked by AMPK inhibitor or YAP1 overexpression in DHT-cultured BPH-1 *** findings indicated that Met would be a promising clinical therapeutic approach for BPH by inhibiting dysregulated steroid hormone-induced PEC proliferation.

A chiral fluorescent probe for molecular reco.nition of basic amino acids in solutions and cells

A novel and readily available binaphthyl-based fluorescent probe（S）-1 was designed and synthesized.（S）-1 can be used to not only chemoselectively discriminate 3 basic amino acids out of co.mon amino acids, but also enantioselectively reco.nize histidine. Enco.ragingly, enantioselective imaging of histidine in cells was achieved for the first time by the probe（S）-1. These performances endowed it potential application in the chiral analysis of basic amino acids in asymmetric synthesis and cell imaging for diagnosis of diseases caused by racemization of histidine. Nuclear magnetic resonance（NMR） and mass spectrometry investigations suggested that different reaction extent of（S）-1 with L/D-histidine and different product structures generated the observed enantioselective fluorescent response. The molecular structures and thermodynamic stability of the co.plexes, formed from（S）-1 + Zn2+and enantiomers of histidine, were calculated by Gaussian 16 based on density functional theory（DFT） to validate the above action mechanism.

Small interfering RNA(siRNA)-based gene silencing has been co.sidered as a potential therapy modality against inflammatory ***,the effective delivery of siRNA to desired destination still remains challenging due to poor stability,high molecular weight and negative ***,ionizable lipid nanoparticle(LNP)has been extensively used as vector for effective delivery of ***,we report a mannose-modified LNP(M-MC_(3) LNP@TNFα)loading tumor necrosis factorα(TNFα)siRNA for targeting liver macrophages,achieving effectively inhibit acute liver *** M-MC_(3) LNP@TNFαnot only increases the internalization of LNP by macrophages,but also enhances the gene silencing efficiency of TNFαin ***,the M-MC_(3) LNP@TNFαexhibits higher accumulation in liver of healthy mice than that of MC_(3) LNP@TNFα(un-modified LNP)owing to the targeting effect of *** expected,the M-MC_(3) LNP@TNFαsignificantly suppresses the expression of TNFαand ameliorates liver damage in acute liver injury *** a LNP targeting siRNA delivery holds great potential for the treatment of diseases associated with liver in the future.
