Live bacteria-mediated antitumor therapies mark a pivotal point in cancer ***,the difficulty in reco.ciling the safety and efficacy of bacterial therapies has limited their *** bacterial tumor-targeted delivery while maintaining biosafety is a critical hurdle for the clinical translation of live microbial therapy for ***,we developed“dead”yet“functional”Salmonella-loaded macrophages using liquid nitrogen co.d shock of an attenuated Salmonella typhimurium VNP20009-co.tained macrophage cell *** obtained“dead”macrophages achieve an average loading of approximately 257 live bacteria per 100 *** engineered cells maintain an intact cellular structure but lose their original pathogenicity,while intracellular bacteria retain their original biological activity and are delay freed,followed by ***“Trojan horse”-like bacterial camouflage strategy avoids bacterial immunogenicity-induced neutrophil recruitment and activation in peripheral blood,reduces the clearance of bacteria by neutrophils and enhances bacterial tumor enrichment efficiently after systemic ***,this strategy also strongly activated the tumor microenvironment,including increasing antitumor effector cells(including M1-like macrophages and CD8+Teffs)and decreasing protumor effector cells(including M2-like macrophages and CD4+Tregs),and ultimately improved antitumor efficacy in a subcutaneous H22 tumor-bearing mouse *** cryo-shocked macrophage-mediated bacterial delivery strategy holds promise for expanding the therapeutic applications of living bacteria for cancer.

For over two decades,the development of B-cell lymphoma-2(Bcl-2)family therapeutics has primarily focused on anti-apoptotic proteins,resulting in the first-in-class drugs called BH3 mimetics,especially for Bcl-2 inhibitor *** pro-apoptotic protein Bcl-2-associated X protein(BAX)plays a crucial role as the executioner protein of the mitochondrial regulated cell death,co.tributing to organismal development,tissue homeostasis,and *** dysregulation of BAX is closely associated with the onset and progression of diseases characterized by pathologic cell survival or death,such as cancer,neurodegeneration,and heart *** addition to co.ducting thorough investigations into the physiological modulation of BAX,research on the regulatory mechanisms of small molecules identified through biochemical screening approaches has prompted the identification of functional and potentially druggable binding sites on BAX,as well as diverse all-molecule BAX *** review presents recent advancements in elucidating the physiological and pharmaco.ogical modulation of BAX and in identifying potentially druggable binding sites on ***,it highlights the structural and mechanistic insights into small-molecule modulators targeting diverse binding surfaces or co.formations of BAX,offering a promising avenue for developing next-generation apoptosis modulators to treat a wide range of diseases associated with dysregulated cell death by directly targeting BAX.

co.nitive dysfunction is a co.e symptom co.mon in psychiatric disorders including depression that is primarily managed by antidepressants lacking efficacy in improving *** this study,we report a novel dual serotonin transporter and voltage-gated potassium Kv7/KCNQ/M-channel inhibitor D01(a 2-methyl-3-aryloxy-3-heteroarylpropylamines derivative)that exhibits both anti-depression effects and improvements in co.nition.D01 inhibits serotonin transporters(K_(i)=30.1±6.9 nmol/L)and M channels(IC_(50)=10.1±2.4μmol/L).D01 also reduces the immobility duration in the mouse FST and TST assays in a dose-dependent manner without a stimulatory effect on *** administrations of D01(20 and 40 mg/kg)can significantly shorten the immobility time in a mouse model of chronic restraint stress(CRS)-induced depression-like ***,D01 dose-dependently improves the co.nitive deficit induced by CRS in Morris water maze test and increases the exploration time with novel objects in normal or sco.olamine-induced co.nitive deficits in mice,but not ***,D01 reverses the long-term potentiation(LTP)inhibition induced by *** together,our findings demonstrate that D01,a dual-target serotonin reuptake and M channel inhibitor,is highly effective in the treatment-resistant depression and co.nitive deficits,thus holding potential for development as therapy of depression with co.nitive deficits.

Raman spectrosco.y has found extensive use in monitoring and co.trolling cell culture *** this co.text,the prediction accuracy of Raman-based models is of paramount ***,models established with data from manually fed-batch cultures often exhibit poor performance in Raman-co.trolled ***,there is a need for effective methods to rectify these *** objective of this paper is to investigate the efficacy of Kalman filter(KF)algorithm in co.recting Raman-based models during cell ***,partial least squares(PLS)models for different co.ponents were co.structed using data from manually fed-batch cultures,and the predictive performance of these models was ***,various co.rection methods including the PLS-KF-KF method proposed in this study were employed to refine the PLS ***,a case study involving the auto-co.trol of gluco.e co.centration demonstrated the application of optimal model co.rection *** results indicated that the original PLS models exhibited differential performance between manually fed-batch cultures and Raman-co.trolled *** gluco.e,the root mean square error of prediction(RMSEP)of manually fed-batch culture and Raman-co.trolled culture was 0.23 and 0.40 g·L^(-1).With the implementation of model co.rection methods,there was a significant improvement in model performance within Raman-co.trolled *** RMSEP for gluco.e from updating-PLS,KF-PLS,and PLS-KF-KF was 0.38,0.36 and 0.17 g·L^(-1),***,the proposed PLS-KF-KF model co.rection method was found to be more effective and stable,playing a vital role in the automated nutrient feeding of cell cultures.
