Unveiling phosphofructokinase as the key target: an innovative strategy combining pull-down assays with optical imaging for snail-killing mechanism exploration

The Pomacea canaliculata, also known as apple snail, is an invasive species in China, posing a serious threat to agricultural production and public health. To prevent its rampant infestation, we developed a highly efficient snail-killing lead compound, 1-(4-chlorophenyl)-3-(pyridin-3-yl)urea (PBQ). However, the killing mechanism of PBQ remains unclear. To elucidate this, two novel compounds, B-PBQ and J-PBQ, were developed through structural modification of PBQ and were used, through both protein pull-down assay and confocal laser scanning microscopy imaging, to identify and confirm that phosphofructokinase(PFK) in the apple snail is the primary target protein of PBQ. Further studies revealed that PBQ successfully inhibits the activity,content, and expression of PFK in apple snail, disrupting the glycolysis pathway and leading to abnormal energy *** disruptions impair the snail’s tolerance to low oxygen, resulting in its efficient eradication. These findings offer a promising direction for developing next-generation PFK-targeted snail-killing pesticides. Furthermore, the strategy of combining pull-down assays with optical imaging used in this work provides a novel approach for the exploration of mechanisms of other pesticide candidates.

Identification and investigation of Calodium hepaticum in rodents and insectivores from Wuhan section of the Yangtze River in China

Objective:To investigate the prevalence of Calodium hepaticum(***)in rodents and insectivores from Wuhan section of the Yangtze River in China,and to provide evidence for the prevention and treatment of hepatic Calodium ***:Rodents and insectivores were captured from three selected Yangtze River beaches using *** three survey sites were divided into six areas according to natural conditions,with 60 mousetraps placed in each *** liver lesions in the captured rodents were observed by the naked eye and the eggs in the liver tissue were observed by microscopic ***:A total of 1080 mousetraps were placed,and 1075 mousetraps were retrieved,with the retrieve rate as 99.5%.A total of 101 Apodemus agrarius,12 Rattus norvegicus,and 9 Crocidura attenuata were *** average density of rodents and insectivores was 10.5%and 0.8%,*** of egg nodules from infected rodents showed 98%similarity with that of *** 18 S rRNA(LC425008.1).One Rattus norvegicus was infected with ***,with an infection rate of 3.23%in the Erqi river beach;the other two beaches did not show the incidence of ***:The monitoring of *** in the Yangtze River beaches should be strengthened to reduce the risk of human *** infection.

Assessment of morbidity due to Schistosoma japonicum infection in China

This paper presents a historical assessment of morbidity due to the Schistosoma japonicum infection in *** to the socio-economic situation,which did not allow for a control program to be implemented until the early 1950s,morbidity was serious and mortality was high before *** on a few investigations and published papers,it can be said that the disease caused millions of deaths,and destroyed numerous families and *** the 1950s,there has been a national control program,intensive control and prevention work has been carried out,and consequently the disease is being *** present,both the prevalence and the morbidity of the disease have been decreasing *** morbidity of the three phases of the disease is outlined in this *** higher morbidity is seen in the acute and advanced phases of the *** four major forms of advanced schistosomiasis i.e.,ascites,megalosplenia,dwarfism,and colonic tumoroid proliferation,are outlined with their characteristic clinical presentations;their proportions are different during various periods of the national control *** schistosomiasis and the relationship between the *** infection and colorectal cancer are also ***-transmission schistosomiasis is briefly discussed(which can happen even if the disease reaches the criteria of elimination,and the infection and transmission have stopped,but yet it still develops).The problem of mammalian reservoir hosts of *** makes the epidemiology and control of schistosomiasis in China even more complicated and arduous,and the control progress in animal reservoirs is briefly presented.

Malaria Transmission Potential in the Three Gorges Reservoir of the Yangtze River, China

Objective To define and evaluate the malaria transmission potential in the Yangtze River, following construction of the Three Gorges Reservoir. Methods Six villages, namely, Kaixian, Fengjie, Wanzhou, Fuling, Yubei, and Zigui were selected for investigating the malaria transmission potential in the reservoir. Transmission potential was estimated by mathematical modeling and evaluation of the local malaria situation. Factors that influenced the transmission potential were analyzed using Poisson regression analysis in combination with Grey Systematic Theory （based on evaluation by the Delphi method）. Results Indirect fluorescent antibody data and the incidence of malaria in the local population were consistent with the malaria transmission potential calculated for the area. Multivariate Poisson regression analysis showed a statistically significant association between the riparian zone and the man‐biting rate. Conclusion The risk of a malaria epidemic can be forecasted using the malaria transmission potential parameters investigated here which was closely correlated with the riparian zone.

schistosomiasis, caused by infections with Schistosoma japonicum, is one of the human parasitic diseases that remains a public health problem throughout the world＇s tropical and subtropical regions. With the sustained efforts for six decades, schistosomiasis has been effectively controlled in China. But in the lake region of the schistosomiasis endemic areas, the disease is still a big burden to some specific population, such as fishmen and boatmen, which hinders the local social-economic development.
