YWHAZ Binds to TRIM21 but Is Not Involved in TRIM21-stimulated Osteosarcoma Cell Proliferation

Objective Osteosarcoma is the most common type of malignant bone tumor in children and adolescents. The role of E3 ligases in tumorigenesis is currently a focus in tumor research. In the present study, we investigated the role of the E3 ligase tripartite motif 21（TRIM21） in osteosarcoma cell *** 3-（4, 5-dimethylthiazol-2-yl）-2, 5-diphenyltetrazolium bromide（MTT） assays were used to assess osteosarcoma cell viability. U2-OS cells stably carrying a recombinant lentivirus expressing tetracycline-regulated TRIM21 were screened. Co-immunoprecipitation was coupled with LCMS/MS analysis to identify novel interacting partners of TRIM21. Co-immunoprecipitation and bimolecular fluorescence complementation（BIFC） were performed to validate the interactions between TRIM21 and its novel partner YWHAZ. A TRIM21-ΔRING construct was generated to test the effects of TRIM21 ligase activity on *** TRIM21 positively regulated osteosarcoma cell proliferation. Overexpression of TRIM21 enhanced osteosarcoma cell tolerance toward various stresses. YWHAZ protein was identified as a novel interacting partner of TRIM21 and its expression levels were negatively regulated by TRIM21. The RING domain of TRIM21 was required for TRIM21 negative regulation of YWHAZ expression. However,overexpression of YWHAZ did not affect positive regulation of osteosarcoma cell proliferation by *** Our results further clarify the molecular mechanisms underlying the pathogenesis of osteosarcoma.

Individual and joint exposures to PM2.5 constituents and mortality risk among the oldest-old in China

Cohort evidence linking long-term survival of older adults with exposure to fine particulate matter(PM2.5) constituents remains scarce in China. By constructing a dynamic cohort based on the Chinese Longitudinal healthy Longevity Study, we aimed to assess the individual and joint associations of major PM2.5constituents with all-cause death in Chinese oldest-old（≥80 years） adults. Time-dependent Cox proportional hazards models were adopted to estimate death risks of long-term exposure to PM2.5constituents. Among 14,884 participants,totaling 56,342 person-years of follow-up, 12,346 deaths were identified. The highest mortality risk associated with an interquartile range（IQR） increase in exposure was 1.081（95% confidence interval [CI]: 1.055–1.108） for sulfate(IQR=4.1 μg m-3), followed by 1.078（95% CI:1.056–1.101） for black carbon(IQR=1.6 μg m-3), 1.056（95% CI: 1.028–1.084） for ammonium(IQR=3.2 μg m-3), 1.050（95% CI: 1.021–1.080） for nitrate(IQR=5.8 μg m-3), and 1.049（95% CI: 1.024–1.074） for organic matter(IQR=10.3 μg m-3). In joint exposure, each IQRequivalent rise of all five PM2.5constituents was associated with an 8.2%（95% CI: 4.0%–12.6%） increase in mortality risk. The weight analysis indicated the predominant role of sulfate and black carbon in driving PM2.5-related mortality. Octogenarians（aged 80–89 years） and rural dwellers were at significantly greater risk of mortality from individual and joint exposures to PM2.5constituents. This study suggests that later-life exposure to PM2.5constituents, particularly sulfate and black carbon, may curtail long-term survival of the oldest-old in China.

Three-Dimensional Urban Thermal Effect across a Large City Cluster during an Extreme Heat Wave:Observational Analysis

Given extensive and rapid urbanization globally,assessing regional urban thermal effects(UTE)in both canopy and boundary layers under extreme weather/climate conditions is of significant *** population and economic growth in the Yangtze River Delta(YRD)have made it one of the largest city clusters in ***,we explore the three-dimensional(3D)UTE in the YRD using multi-source observations from high-resolution automatic weather stations,radiosondes,and eddy covariance sensors during the record-setting heat wave(HW)of July-August *** is found that the regional canopy layer UTE is up to 0.6-1.2℃,and the nocturnal UTE(0.7-1.6℃)is larger than daytime UTE(0.2-0.5℃)during the *** regional canopy layer UTE is enhanced and expanded northwards,with some rural sites contaminated by the urban influences,especially at *** the boundary layer,the strengthened regional UTE extends vertically to at least 925 hPa(~750 m)during this *** strengthened 3D UTE in the YRD is associated with an enlarged Bowen ratio difference between urban and non-urban *** findings about the 3D UTE are beneficial for better understanding of the thermal environment of large city clusters under HW and for more appropriate adaption and mitigation strategies.

Five-year follow-up study of multi-domain cognitive training for healthy elderly community members

Background:Cognitive training, a safe non-pharmacological intervention, may help mitigate cognitive decline and prevent the development of dementia in elderly individuals. Objective:Evaluate the long-term effects of cognitive training among healthy elderly community members. Methods:healthy individuals 70 years of age or older from one urban community in Shanghai were screened and the 151 individuals who met inclusion criteria were assigned either to an intervention group (n=90) or a control group (n=61). The intervention involved twice-weekly training in reasoning, memory, and strategy that continued for 12 weeks (a total of 24 sessions). Participants were assessed at baseline and at 12 weeks, and 5 years after enrollment using the Chinese versions of the Neuropsychological Test Battery for Elderly persons (NTBE), the Stroop Color-Word Test, and a general health questionnaire. Results:Forty-nine (54%) intervention group subjects and 33 (54%) control group subjects completed the S-year follow-up. There were few differences in the baseline neurocognitive measures of those who did and did not complete the 5-year follow-up, and there were few differences between those who dropped out of the intervention group compared to those who dropped out of the control group. At the 5-year follow-up, individuals in the intervention group performed better than those in the control group on only 5measures (in the Trails Making A Test and the Cancellation Test 3) of the 61measures assessed by NTBE and the Stroop tests, but none of these differences met the pre-determined required level of statistical significance (p=0.0008). Conclusion:We do not confirm the results of previous studies that report long-term benefits of brief cognitive training courses for elderly community residents. Our failure to identify differences in cognitive functioning five years after cognitive training is not likely due to differential dropout between the intervention and control groups but may be related to

Effects of extremely low frequency magnetic field on anxiety level and spatial memory of adult rats

Background As the widespread use of electric devices in modern life, human are exposed to extremely low frequency magnetic fields （ELF MF） much more frequently than ever. Over the past decades, a substantial number of epidemiological and experimental studies have demonstrated that ELF MF （50 Hz） exposure is associated with increased risk of various health effects. The present study examined the effects of chronic exposure to ELF MF on anxiety level and spatial memory of adult rats. Methods The 50-Hz ELF MF was used during the whole experimental procedures and the value of magnetic field （MF） was set to 2 mT. Adult rats were divided randomly to control, MF 1 hour and MF 4 hours group. Anxiety-related behaviors were examined in the open field test and the elevated plus maze; changes in spatial learning and memory were determined in Morris water maze aRer 4 weeks of daily exposure. Results Rats in MF 4 hours group had increased anxiety-like behaviors with unaltered locomotor activity. In the Morris water maze test, rats had reduced latency to find the hidden platform and improved long-term memory of former location of platform without changes in short-term memory and locomotor activity. Conclusion Chronic ELF MF exposure has anxiogenic effect on rats, and the promoting effects on spatial learning and long-term retention of spatial memory.
