Objective:To investigate the effects of Clean-DM1(C-DM1),a polyherbal formulation of Radix Scrophulariae,Radix Astragali,Rhizoma Atractylodis,and Radix Salviae Miltiorrhizae,on high-fat diet(HFD)-induced diabetes ***:The information about active components of C-DM1 extract and molecular mechanism was obtained from network pharmacology *** compounds of C-DM1 extract by high performance liquid chromatography-mass spectrometry(HPLC-MS)analysis were conducted for quality *** in vivo study,mice were induced diabetes by HFD for 12 *** mice in the normal group(Nor)were maintained with a regular diet and treated with saline by *** HFD model mice were randomly divided into 3 groups,including a HFD diabetic model group,a C-DM1 extract-administered group(C-DM1,500 mg/kg),and metformin-administered groups(Met,500 mg/kg),8 mice in each *** intake,body weight(BW),and fasting blood glucose(FBG)levels were recorded weekly for 4 *** 4 weeks of treatment,alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood glucose,low-density lipoprotein cholesterol(LDL-C)were determined using an automated clinical chemistry analyzer,and homeostatic model for assessing insulin resistance(HOMA-IR)levels and oral glucose tolerance test(OGTT)were *** histopathological changes of liver and pancreatic tissues were observed by hematoxylin-eosin *** receptor substrate(IRS)/phosphatidylinositol 3 kinase(PI3K)/protein kinase B(AKT)and adenosine 5'-monophosphate-activated protein kinase(AMPK)expressions in liver and pancreas tissues were detected by Western blot ***:HPLC-MS identified dihydroisotanshinone,dihydroisotanshinone I,cryptotanshinone,harpagoside,and atractyloside A in C-DM1 *** administration of C-DM1 extract significantly decreased body weight,calorie intake,and the levels of blood glucose and insulin in the diabetic mice(P<0.05 or P<0.01).The C-DM1 extract administration improved the im

Dear editor,Severe acute respiratory syndrome coronavirus2(SARS-CoV-2)infection can lead to severe outcomes in patients with cancer[1].It has been reported that patients with lung cancers disproportionately manifest severe COVID-19 with a high rate of hospitalization and death[2].Notably,the SARS-CoV-2 Spike(S)protein can induce hyper-inflammation in both epithelial cells and macrophages through toll-like receptor(TLR)1/TLR2 or TLR2/6-dependent nuclear factor-kappaB(NF-κB)path-way[3].However,molecular and cellular evidence on whether the SARS-CoV-2 virus affects the severity of lung cancer patients through TLR1/2 or TLR2/6 signaling remains unclear.

Spatial-temporal shifting patterns and in situ conservation of spotted seal(Phoca largha)populations in the Yellow Sea ecoregion

Understanding the habitat shifting pattern is a prerequisite for implementing in situ conservation of migratory *** seals(Phoca largha)inhabiting the Yellow Sea ecoregion(YSE)comprise a small population with independent genes and represent a charismatic flagship species in this ***,this population has declined by 80%since the 1940s,and increased support from the countries around the YSE is urgently needed to address the potential local extinction risk.A time-series niche model and life-history weighted systematic conservation planning were designed on the basis of a satellite beacon tracking survey(2010–2020)of the YSE *** results showed clustering and spreading shifting patterns during the breeding and migratory seasons,*** closed-loop migration route formed in the YSE indicated that this population might be geographically isolated from populations in other breeding areas around the *** conservation priority area(CPA),with an area of 19632 km^(2)(3.58%of the total YSE area),was the most effective response to the potential in situ ***,nearly 80%of the CPA was exposed outside the existing marine protected areas(MPAs).Future establishment of MPAs in China should strategically consider the conservation gap identified herein,and it is recommended for Korea’s closed fishing season to be spatially set in the western korean Peninsula from May to *** study also exemplified that the lack of temporal information would lead to the dislocation of niche modeling for migratory species represented by spotted *** should be paid to protecting small and migratory populations in marine biodiversity conservation planning.

Effect of Asiasari radix on osteoblastic differentiation of stem cells derived from gingiva

OBJECTIVE: To examine the dose-dependent impact of Asiasari Radix(A. radix) on the cell viability,differentiation and mineralization of stem cells derived from ***: Stem cells that were derived from gingiva were grown in the presence of A. radix at final concentrations that ranged from 0.001 to 10 μg/m L. The morphology of the cells was viewed under an inverted microscope and the analysis of cell proliferation was performed by using Cell Counting Kit-8(CCK-8) on day 1. The alkaline phosphatase activity test was used to assess differentiation and Alizarin red S staining was used to assess mineralization of treated ***: The control group showed spindleshaped, fibroblast-like morphology and the shapesof the cells in 0.001, 0.01, 0.1, 1 and 10 μg/mL of *** were similar to that of the control group at day 1. The cultures growing in the presence of0.001 μg/m L of A. radix at day 1 showed an increase in the CCK-8 value(P 0.05). Mineralized extracellular deposits were observed after Alizarin Red S staining and the cultures grown in the presence of 0.001 μg/m L of A. radix showed the highest value for quantitative results for bound dye(P < 0.05).CONCLUSION: Within the limits of this study, A. radix influenced the proliferation of stem cells derived from the gingiva and low concentrations of *** might enhance osteogenic differentiation of the stem cells.

Genetic information is transcribed from genomic DNA to mRNA,which is then translated into threedimensional *** can undergo various post-transcriptional modifications,including RNA editing that alters mRNA sequences,ultimately affecting protein *** this study,RNA editing was identified at the 499th base(c.499)of human vaccinia-related kinase 2(VRK2).This RNA editing changes the amino acid in the catalytic domain of VRK2 from isoleucine(with adenine base)to valine(with guanine base).Isoleucine-containing VRK2 has higher kinase activity than the valine-containing VRK2,which leads to an increase in tumor cell *** we reported that VRK2 directly interacts with dystrobrevin-binding protein(dysbindin)and results in reducing its ***,we demonstrate that isoleucine-containing VRK2 decreases the level of dysbindin than valinecontaining *** interacts with cyclin D and thereby regulates its expression and *** reduction in the level of dysbindin by isoleucine-containing VRK2 further enhances the cyclin D expression,resulting in increased tumor growth and reduction in survival *** has also been observed that in patient samples,VRK2 level was elevated in breast cancer tissue compared to normal breast ***,the isoleucine form of VRK2 exhibited a greater increase in breast cancer ***,it is concluded that VRK2,especially dependent on the 167th variant amino acid,can be one of the indexes of tumor progression and proliferation.

Current therapeutic approaches for volumetric muscle loss(VML)face challenges due to limited graft availability and insufficient *** overcome these limitations,tissue-engineered scaffolds have emerged as a promising *** this study,we developed aligned ternary nanofibrous matrices comprised of poly(lactide-co-ε-caprolactone)integrated with collagen and Ti_(3)C_(2)T_(x)MXene nanoparticles(NPs)(PCM matrices),and explored their myogenic potential for skeletal muscle tissue *** PCM matrices demonstrated favorable physicochemical properties,including structural uniformity,alignment,microporosity,and *** vitro assays revealed that the PCM matrices promoted cellular behaviors and myogenic differentiation of C2C12 ***,in vivo experiments demonstrated enhanced muscle remodeling and recovery in mice treated with PCM matrices following VML *** insights from next-generation sequencing revealed that MXene NPs facilitated protein and ion availability within PCM matrices,leading to elevated intracellular Ca^(2+)levels in myoblasts through the activation of inducible nitric oxide synthase(i NOS)and serum/glucocorticoid regulated kinase 1(SGK1),ultimately promoting myogenic differentiation via the m TOR-AKT ***,upregulated i NOS and increased NO–contributed to myoblast proliferation and fiber fusion,thereby facilitating overall myoblast *** findings underscore the potential of MXene NPs loaded within highly aligned matrices as therapeutic agents to promote skeletal muscle tissue recovery.

Therapeutic Effects of traditional Herbal medicine on Cerebral Ischemia:A Perspective of Vascular Protection

Although many agents for acute ischemic stroke treatment have been developed from extensive preclinical studies, most have failed in clinical trials. As a result, researchers are seeking other methods or agents based on previous studies. Among the various prospective approaches, vascular protection might be the key for development of therapeutic agents for stroke and for improvements in the efficacy and safety of conventional therapies. traditional medicines in Asian countries are based on clinical experiences and literature accumulated over thousands of years. To date, many studies have used traditional herbal medicines to prove or develop new agents based on stroke treatments mentioned in traditional medicinal theory or other clinical data. In the current review, we describe the vascular factors related to ischemic brain damage and the herbal medicines that impact these factors, including Salviae Miltiorrhizae Radix, Notoginseng Radix, and Curcumae Rhizoma, based on scientific reports and traditional medical theory. Further, we point out the problems associated with herbal medicines in stroke research and propose better methodologies to address these problems.
