Evaluation of Fusarium Graminearum Growth and Deoxynivalenol Content in Susceptible and Tolerant Varieties of Triticum Aestivum

The aim of this work was to verify if the wheat susceptibility/tolerance phenotype to Fusarium head blight （FHB） into the field could be related to the ability of the re-activated seeds to hamper deoxynivalenol （DON） synthesis by Fusarium graminearum into non optimal （i.e. high humidity） storage conditions. On this purpose, two Fusariurn graminearum strains, Fg126 and Fg8308, were separately inoculated on active but not germinating kernels of Triticum aestivum L. cv ＂Sagittario＂（FHB-susceptible） and cv ＂Blasco＂（FHB-tolerant）, The growth of the fungal strains on wheat kernels was monitored from 0 to 15 days post inoculation through a quantitative SYBR green real-time PCR, as well as ergosterol content through HPLC for comparison. DON biosynthesis into the contaminated wheat seeds was quantified by HPLC method in the same time intervals as above. Significant differences in the ability to grow and synthesize DON, acetylated and glucosylated forms, emerged between the strains also in relation to the wheat variety on which the pathogen developed. The results obtained indicated that the wheat variety which from the epidemiological studies carried out into the field is FHB-tolerant, i.e. Blasco, resulted as the more efficient in hampering DON synthesis in both F. graminearum strains. Thus, this preliminary study could represent a contribution to find out less time consuming methods for screening the wheat varieties tolerant to DON accumulation in non optimal storage conditions.

Despite the considerable effort,fast and highly sensitive photodetection is not widely available at the low-photon-energy range(~meV)of the electromagnetic spectrum,owing to the challenging light funneling into small active areas with efficient conversion into an electrical ***,we provide an alternative strategy by efficiently integrating and manipulating at the nanoscale the optoelectronic properties of topological Dirac semimetal PtSe_(2)and its van der Waals ***,we realize strong plasmonic antenna coupling to semimetal states near the skin-depth regime(λ/10^(4)),featuring colossal photoresponse by in-plane symmetry *** observed spontaneous and polarization-sensitive photocurrent are correlated to strong coupling with the nonequilibrium states in PtSe_(2)Dirac semimetal,yielding efficient light absorption in the photon range below 1.24 meV with responsivity exceeding∼0.2 A/W and noise-equivalent power(NEP)less than~38 pW/Hz^(0.5),as well as superb ambient *** results pave the way to efficient engineering of a topological semimetal for high-speed and low-energy photon harvesting in areas such as biomedical imaging,remote sensing or security applications.

Semiconductor devices are strong competitors in the race for the development of quantum computational *** this work,we interface two semiconductor building blocks of different dimensionalities with complementary properties:(1)a quantum dot hosting a single exciton and acting as a nearly ideal single-photon emitter and(2)a quantum well in a 2D microcavity sustaining polaritons,which are known for their strong interactions and unique hydrodynamic properties,including ultrafast real-time monitoring of their propagation and phase *** the present experiment,we can thus observe how the injected single particles propagate and evolve inside the microcavity,giving rise to hydrodynamic features typical of macroscopic systems despite their genuine intrinsic quantum *** the presence of a structural defect,we observe the celebrated quantum interference of a single particle that produces fringes reminiscent of wave *** this behavior could be theoretically expected,our imaging of such an interference pattern,together with a measurement of antibunching,constitutes the first demonstration of spatial mapping of the self-interference of a single quantum particle impinging on an obstacle.

药物抗生素(PA)残留物普遍存在于土壤和水环境中;因受去除效率及可产生致癌副产物的制约,目前的生物或光化学降解方法难以有效去除PA,且金属离子与PA的吸附/络合机制尚不清楚。本文用FeCl_(3)从水溶液中去除两种PA—盐酸左氧氟沙星(LH)和盐酸坦索罗辛(TH);发现pH=7.5~8.5时,铁以Fe-PA沉淀物(Fe-PAp)形式将PA从水溶液中分离;连续沉淀5次后,90.3%的LH和87.5%TH被去除;分析LH、TH和Fe-PAp的波峰特征,发现:1)LH和TH主要红外峰:3 433和3 329 cm^(-1)(羧酸OH和NH2)、1 676 cm^(-1)(-COO-)、1 467 cm^(-1)(脂肪族C-H)和1 022 cm^(-1)(-S=O),2)Fe-PAp中前述红外峰强度下降或完全消失。通过化学键分析表明:在Fe-PAp形成过程中,电子从PA内各种官能团转移到Fe3+的空d轨道,形成π-d电子键系统。FeCl_(3)应用广泛:1)作为一种高效且有应用前景的无机吸附剂,能够从水中去除PA,可实现大规模分离生活和工业废水中的有机污染物;2)Fe-PAp的形成表明金属-有机复合物可以在碱性土壤和水中长期稳定,解释了Fe-DOM复合物在环境中的长期稳定/固存机制。

Emerging Role of the Ubiquitin Proteasome System in the Control of Shoot Apical Meristem Function

The shoot apical meristem （SAM） is a population of undifferentiated cells at the tip of the shoot axis that establishes early during plant embryogenesis and gives rise to all shoot organs throughout the plant＇s life. A plethora of different families of transcription factors （TFs） play a key role in establishing the equilibrium between cell differentiation and stem cell maintenance in the SAM. Fine tuning of these regulatory proteins is crucial for a proper and fast SAM response to environmental and hormonal cues, and for development progression. One effective way to rapidly inactivate TFs involves regulated proteolysis by the ubiquitin/26S proteasome system （UPS）. However, a possible role of UPS-dependent protein degradation in the regulation of key SAM TFs has not been thoroughly investigated. Here, we summarize recent evidence supporting a role for the UPS in SAM maintenance and function. We integrate this survey with an in silico analysis of publicly-available microarray databases which identified ubiquitin ligases that are expressed in specific areas within the SAM, suggesting that they may regulate or act downstream of meristem-specific factors.

Sweet cherry(Prunus avium L.)is a stone fruit widely consumed and appreciated for its organoleptic properties,as well as its nutraceutical *** here investigated the characteristics of six non-commercial Tuscan varieties of sweet cherry maintained at the Regional Germplasm Bank of the cnr-IBE in Follonica(Italy)and sampled ca.60 days post-anthesis over three consecutive years(2016-2017-2018).We adopted an approach merging genotyping and targeted gene expression profiling with *** complement the data,a study of the soluble proteomes was also performed on two varieties showing the highest content of *** identified the presence of flavanols and proanthocyanidins in highest abundance in the varieties Morellona and Crognola,while gene expression revealed that some differences were present in genes involved in the phenylpropanoid pathway during the 3 years and among the ***,proteomics on Morellona and Crognola showed variations in proteins involved in stress response,primary metabolism and cell wall *** the best of our knowledge,this is the first multi-pronged study focused on Tuscan sweet cherry varieties providing insights into the differential abundance of genes,proteins and metabolites.
