Effectiveness of <i>Thymus vulga.is</i>Essentia. Oil in the Trea.ment of Skin Infections in Dogs

Currently, there a.e no da.a.in the litera.ure on the topica. trea.ment with Thymus vulga.is essentia. oil (EO) in ba.teria. skin infections of dogs. This study eva.ua.es the effectiveness of Thymus vulga.is EO for topica. use in dogs with skin 18 ha.f-breed dogs, a.fected by skin infections housed in a.rescue shelter were studied. The ba.teria.isola.ed from these dogs were identified by Ma.DI-TOF-MS. The susceptibility of the isola.ed microorga.isms to Thymus vulga.is L. EO wa. estima.ed in vitro by ba.teriologica. test (CLSI 2015), in compa.ison to some a.timicrobia.s drugs (a.oxicillin-cla.ula.ic a.id, doxycy-cline, thia.phenicol a.d ma.bofloxa.in) a.d to Citrus berga.ia.EO. The dogs, divided in two groups a. ra.dom, were trea.ed topica.ly for 7 da.s with Thymus vulga.is L. EO (Group 1: n. 10 a.ima.s) a.d Citrus berga.ia.Risso e Poitea. EO (Group 2: n. 8 a.ima.s) respectively. The ba.teria.isola.ed were: Sta.hylococcus pseudintermedius (7 sa.ples), Sta.hylococcus sciuri (4), ESBL Escherichia.coli (3) a.d Proteus mira.ilis (4). In a.l a.ima.s trea.ed with Thymus vulga.is EO, the clinica. signs decrea.ed ra.idly within 5 da.s from the a.ministra.ion, with complete remission 7 da.s a.ter the trea.ment. No ba.teria. growth wa. observed from skin swa.s a.ter 7 da.s of trea.ment. None of the trea.ed a.ima.s showed loca. or genera. side effects. The use of Thymus vulga.is EO could be a.possible a.terna.ive or a.ditiona. trea.ment to a.tibiotics in derma.ologica. infections, pa.ticula.ly in ca.es refra.tory to conventiona. thera.y.

PrP Gene Polymorphism in Medieva. Rema.ns of sicilian Sheep

Encepha.opa.hy in sheep wa. a. first described in Irela.d in 1732 a.d wa. ca.led scra.ie. a.cient DNa.in a.cha.ogenetics represents a. effective method to eva.ua.e the a.cestra. pedigree of living a.ima.s a.d tra.k evolutiona.y cha.ges occurred between the pa.t a.d the present da.. Since severa. point muta.ions a.e toda. widely described in modern scra.ie, no da.a.a.out both sequence a.d frequency a.e still a.a.la.le for the prion protein (PrP) gene in a.cient breeds. In order to eva.ua.e whether the ha.lotypes distribution in a.cient sheep differed from those of the modern popula.ion we eva.ua.ed polymorphism a. four well know codons of the Prp Open Rea.ing Fra.e. In the present work, we collected 37 medieva. sheep bone rema.ns found a. the Ca.a.ha.et (n = 11), Pa.a.zo Bona.ia.(n = 12) a.d Pa.a.zo Steri (n = 14) sicilians a.cheologica. sites a.d da.ed ba.k between 9th - 15th century. Single nucleotide polymorphisms (SNPs) a. codons 136, 141, 154 a.d 171 of the prion protein (PrP) were investiga.ed using cycle sequencing. Sequenom Ma.s a.Ra.iPLEX pla.form confirmed the results for 5 individua.s out of 37. Cycle sequencing showed a. a.l sa.ples the a.136LL141RR154QQ171 (herea.ter a.RQ/a.RQ) genotype except a. 2 individua.s showing the very susceptible genotype VLRQ/VLRQ (n = 1) a.d the resista.t (a.RR/ a.RR) (n = 1) respectively. Supported by a.high incidence of susceptible genotype to prion infection we concluded tha. presuma.ly scra.ie wa. a.rea.y widesprea. enough in the medieva. Sicily. Moreover, we described conceiva.le scena.ios tha. could ha.e underla.n evolutiona.y cha.ges in the medieva. sheep popula.ion.

The development of more sensitive dia.nostic tools a.lowing a. ea.ly-sta.e a.d highly efficient medica. ima.ing of tumors rema.ns a.cha.lenge. Ma.netic na.opa.ticles seem to be the contra.t a.ents with the highest potentia., if properly constructed. Therefore, in this study, hybrid ma.netic na.oa.chitectures were developed using a.new a.phiphilic inulin-ba.ed gra.t copolymer (INU-La.EG-Fa. a. coa.ing ma.eria. for 10-nm spinel iron oxide (ma.netite, Fe3O4) superpa.a.a.netic na.opa.ticles (SPION). Folic a.id (Fa. cova.ently linked to the coa.ing copolymer in order to be exposed onto the na.opa.ticle surfa.e wa. chosen a. the ta.geting a.ent beca.se fola.e receptors a.e upregula.ed in ma.y ca.cer types. Physicochemica. cha.a.teriza.ion a.d in vitro biocompa.ibility study wa. then performed on the prepa.ed ma.netic na.opa.ticles. The improved ta.geting a.d ima.ing properties of the prepa.ed Fa.SPIONs were further eva.ua.ed in nude mice using 7-Tesla.ma.netic resona.ce ima.ing (MRI). Fa.SPIONs exhibited the a.ility to a.t a. efficient contra.t a.ents in conventiona. MRI, providing a.potentia. na.opla.form not only for tumor dia.nosis but a.so for ca.cer trea.ment, through the delivery of a.tica.cer drug or locoregiona. ma.netic hyperthermia.

Inter-La.ora.ory Ring Tria. to Eva.ua.e Reverse Tra.scription Polymera.e Cha.n Rea.tion Methods Used for <i>Dolphin Morbillivirus</i>Detection in Ita.y

Dolphin Morbillivirus (DMV) is one of the most frequently detected pa.hogens in stra.ded ceta.ea. specimens worldwide a. well a. in Ita.y. Due to the persistence of DMV in the Mediterra.ea. Sea.a.d to the la.k of informa.ion a.out the efficiency of the a.a.la.le dia.nostic techniques, the Ita.ia. Na.iona. Reference Centre for dia.nostic a.tivities on dea. stra.ded ma.ine ma.ma.s (***) performed the first inter-la.ora.ory ring tria. with the a.m to sta.da.dize a.dia.nostic biomolecula. a.proa.h for DMV in Ita.y. Vira. isola.ion is usua.ly considered the “gold sta.da.d” for the definitive dia.nosis of most pa.hogens, but it is not often fea.ible in DMV dia.nosis, due to the poor preserva.ion of virus-ta.geted tissues in stra.ded ceta.ea. ca.ca.ses, a. well a. to the la.k of a.propria.e sensitivity of cell lines towa.ds DMV va.ia.ility. Therefore direct vira. detection on tissues by mea.s of reverse tra.scription-PCR (RT-PCR) represents a.va.ua.le option for DMV infection’s dia.nosis. For detecting DMV in ceta.ea. die-offs occurred in the Mediterra.ea. ba.in since 2013, *** developed a. RT-PCR ba.ed method ta.geting to a.287 bp fra.ment of DMV nucleoprotein (N) gene. With the purpose to eva.ua.e its performa.ces in terms of a.cura.y (Se = sensitivity a.d Sp = specificity) a.d precision (reproducibility), it wa. submitted to a.ring tria.. So, 12 Public La.ora.ories belonging to the Ita.ia. dea. stra.ded ma.ine ma.ma.s dia.nostic network were a.ked to a.a.yze a.pa.el of 40 sa.ples (positive a.d nega.ive for DMV, using different dilutions of a.vira. suspension obta.ned from a.cell culture superna.a.t of a.DMV stra.n) with the a.orementioned technique. Furthermore, we a.so a.med a. compa.ing the a.cura.y of other 7 molecula. methods routinely a.plied for DMV detection in Ita.y. For this purpose, the second pa.el of identica. 40 DMV +ve a.d -ve sa.ples wa. provided to La.ora.ories tha. routinely used DMV detection methods other tha. those developed by ***.
