Being sessile organisms, plants evolved an unparalleled plasticity in their post-embryonic development, allowing them to adapt and fine-tune their vital parameters to an ever-changing environment. Cross-talk between plants and their environment requires tight regulation of information exchange at the plasma membrane （PM）. Plasma membrane proteins mediate such communication, by sensing variations in nutrient availability, external cues as well as by controlled solute transport across the membrane border. Localiza-tion and steady-state levels are essential for PM protein function and ongoing research identified cis- and trans-acting determinants, involved in control of plant PM protein localization and turnover. In this overview, we summarize recent progress in our understanding of plant PM protein sorting and degradation via ubiquitylation, a post-translational and reversible modification of proteins. We highlight characterized components of the machinery involved in sorting of ubiquitylated PM proteins and discuss consequences of protein ubiquitylation on fate of selected PM proteins. Specifically, we focus on the role of ubiquitylation and PM protein degradation in the regulation of polar auxin transport （PAT）. We combine this regulatory circuit with further aspects of PM protein sorting control, to address the interplay of events that might control PAT and polarized growth in higher plants.

Treatment of the human esophageal cancer cell line EC8712 with retinoic acid (RA) stopped the cell growth significantly and gave rise to terminal differentiation of the cells characterized by increased expression of involucrin gene. Two cDNA libraries were constructed from the parental and RA-treated cells respectively. Repeated subtractive hybridization of single-stranded plasmid DNA prepared from pooled colonies of cDNA library of the parental cells with cDNA probe generated from the RA-treated cells exhausted sequences common to both libraries of the cell. The unhybridized cDNA probe represented, therefore, the genes activated after RA-treatment. By using these enriched cDNAs as probe to screen the cDNA library constructed from the RA-treated cells thirty-nine positive colonies were obtained, of which two were specifically due to RA-induction. One of these two cDNA clones, designated as pRA538, has undergone further analysis and shown differentiation-inducing effect on parental cancer cells. A novel strategy for cloning genes involved in terminal differentiation of cancer cells is developed.

Objective Different regions of the gut exhibit specific motility patterns regulated by the enteric nervous system(ENS). Currently, little is known about how ENS circuits are differentially organized to generate these regionally distinct motility patterns. The present study aims to explore whether calcium imaging of the ENS can uncover differences in neuronal circuits present in the proximal and distal colon. Methods Live Ca imaging was performed on myenteric plexus preparations from adult Wnt1-Cre;R26-GCa MP3 mice, where all enteric neurons express the genetically-encoded Ca indicator, GCaMP3. Focal electrical stimulation was applied(300μsec, 20 Hz, 2 sec) and responses in individual neurons were analyzed. Immunohistochemistry was performed to compare differences in the neuronal density and phenotype. Furthermore, we transduced the mouse ENS using AAV9-CMV-GFP which made it possible to trace the projections of individual myenteric neurons. Results Using a low magnification(5 X) lens, we were able to simultaneously record the intracellular Ca changes from a large population of neurons spread over many different myenteric ganglia(up to 1.3 mm x 1.7 mm field of view). Analysis of the number of Hu+ myenteric neurons and the intraganglionic fibers showed that both these parameters were significantly higher in the proximal colon, which reflects the higher density of the myenteric neurons and higher packing of intraganglionic fibers at the stimulation site. Live Ca imaging showed that application of the nicotinic receptor antagonist, hexamethonium(200μM), significantly decreased the electricallyevoked Ca transients in the majority of neurons in both regions. Moreover, fraction of neurons in which Ca responses were completely abolished by hexamethonium was significantly larger in the distal colon in comparison with the proximal colon. Relatively large measurement uncertainty severely hindered wide application for laser-induced breakdown spectroscopy(LIBS),therefore it is of great importance to understand the mechanism of signal uncertainty generation,including initiation and *** has been found that the Auctuation of plasma morphology was the main reason for signal ***,it still remains unclear what mechanism leads to laser-induced plasma morphology *** the present work,we employed three fast-imaging cameras to capture three successive plasma images fromn a same laser-induced Titanium alloy plasma,which enables us to understand more clearly of the plasma evolution process especially for the early plasma evolution stage when plasma and surrounding gases interact *** from the images,the plasma experienced an increasing morphological fuctuation as delay time increased,transforming from a“stable plasma”before the delay time of 100 ns to a“fuctuating plasma”after the delay time of 300 ***,the frontier part of plasma showed a significant down-ward motion from the delay time of 150 ns to 200 ns and crashed with the lower part of the plasma,making the plasma fatter and later even splitting the plasma into two parts,which was considered as a critical process for the transformation of“stable plasma”to“unstable plasma”.By calculating the correlation coefficient of plasma image pairs at successive delay times,it was found that the higher the similarity between two plasma at early stage,the more similar at later stage;this implied that the tiny plasma fuctuation earlier than the critical delay time(150-200 ns)was amplifed,causing a large plasma fluctuation at the later stage as well as LIBS measurement *** initation of slight fluctuation was linked with Rayleigh-Taylor Instability(RTI)due to the drastic material interpenetration at the plasma-ambient gas interface at earlier stage(before 50 ns).That is,the uncertainty generation of LIBS was p

Ammonium uptake in plant roots is mediated by AMT/MEP/Rh-type ammonium transporters. Out of five AMTs being expressed in Arabidopsis roots, four AMT1-type transporters contribute to ammonium uptake, whereas no physiological function has so far been assigned to the only homolog belonging to the MEP subfamily, AMT2;1. Based on the observation that under ammonium supply, the transcript levels of AMT2;1 increased and its promoter activity shifted preferentially to the pericycle, we assessed the contribution of AMT2;1 to xylem loading. When exposed to ^15N-labeled ammonium, amt2;1 mutant lines translocated less tracer to the shoots and contained less ammonium in the xylem sap. Moreover, in an amtl;1 amtl;2 amtl ;3 amt2;1 quadruple mutant （qko）, co-expression of AMT2;1 with either AMT1;2 or AMT1;3 significantly enhanced ^15N translocation to shoots, indicating a cooperative action between AMT2;1 and AMT1 transporters. Under N deficiency, proAMT2;1-GFP lines showed enhanced promoter activity predominantly in cortical root cells, which coincided with elevated ammonium influx conferred by AMT2;1 at millimolar sub- strate concentrations. Our results indicate that in addition to contributing moderately to root uptake in the low-affinity range, AMT2;1 functions mainly in root-to-shoot translocation of ammonium, depending on its cell-type-specific expression in response to the plant nutritional status and to local ammonium gradients.

The liver consists predominantly of hepatocytes and biliary epithelial cells(BECs), which serve distinct physiological functions. Although hepatocytes primarily replenish their own population during homeostasis and injury repair, recent findings have suggested that BECs can transdifferentiate into hepatocytes when hepatocyte-mediated liver regeneration is impaired. However, the cellular and molecular mechanisms governing this BEC-to-hepatocyte conversion remain poorly understood largely because of the inefficiency of existing methods for inducing lineage conversion. Therefore, this study introduces a novel mouse model engineered by the Zhou’s lab, where hepatocyte senescence is induced by the deletion of the fumarylacetoacetate(Fah) gene. This model facilitates the efficient conversion of BECs to hepatocytes and allows for the simultaneous lineage tracing of BECs; consequently, a transitional liver progenitor cell population can be identified during lineage conversion. This study also outlines the technical procedures for utilizing this model to determine the underlying cellular and molecular mechanisms of BEC-to-hepatocyte conversion and provides new insights into liver regeneration and its underlying molecular mechanism.
