Unveiling an ancient whole-genome duplication event in Stentor,the model unicellular eukaryotes

Whole-genome duplication(WGD) events are widespread across eukaryotes and have played a significant role in moulding the genetic architectures of diverse organisms. In the present study, the newly sequenced genome of a giant ciliated protist, Stentor roeselii, provides an opportunity for the analysis of the collinearity and retention of reciprocal best-hit genes between two Stentor species. As a main result, we have unveiled a previously undetected ancient WGD event shaping the genome of its congener, Stentor coeruleus, a model protist used in cytological and evolutionary studies. Genomes of two congeners, S. coeruleus and S. roeselii, are compared and analyzed, revealing that:(i)the former exhibits a much higher retention rate of colinear-gene pairs(28%) than does S. roeselii, and in S. coeruleus, 75% of genes that have a RBH hit in S. roeselii, have paralogs with high amino-acid identity, consistent with a WGD event in the lineage leading to S. coeruleus;(ii) the S. roeselii genome possesses extremely short intergenic regions, implying that the lengths of intergenic regions are under strong selection;(iii) the unique characteristics of introns may have been shaped in the common ancestor of heterotrichs;(iv) gene families that play a role in activities of multiple protein kinases and voltage-gated ion channels expanded rapidly in the ancestor of both taxa, possibly relating to the remarkable regenerative ability in Stentor. This study offers new insights into the evolutionary dynamics of ciliate genomes,with implications for understanding of the processes underlying the evolution of genomic complexity.

Salsolinol as an RNA m~6A methylation inducer mediates dopaminergic neuronal death by regulating YAP1 and autophagy

Salsolinol(1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline,Sal)is a catechol isoquinoline that causes neurotoxicity and shares structural similarity with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,an environmental toxin that causes Parkinson's ***,the mechanism by which Sal mediates dopaminergic neuronal death remains *** this study,we found that Sal significantly enhanced the global level of N~6-methyladenosine(m~6A)RNA methylation in PC12 cells,mainly by inducing the downregulation of the expression of m~6A demethylases fat mass and obesity-associated protein(FTO)and alk B homolog 5(ALKBH5).RNA sequencing analysis showed that Sal downregulated the Hippo signaling *** m~6A reader YTH domain-containing family protein 2(YTHDF2)promoted the degradation of m~6A-containing Yes-associated protein 1(YAP1)mRNA,which is a downstream key effector in the Hippo signaling ***,downregulation of YAP1 promoted autophagy,indicating that the mutual regulation between YAP1 and autophagy can lead to *** findings reveal the role of Sal on m~6A RNA methylation and suggest that Sal may act as an RNA methylation inducer mediating dopaminergic neuronal death through YAP1 and *** results provide greater insights into the neurotoxic effects of catechol isoquinolines compared with other studies and may be a reference for assessing the involvement of RNA methylation in the pathogenesis of Parkinson's disease.

Evaluating the impact of spatio-temporal scale on CPUE standardization

This study focused on the quantitative evaluation of the impact of the spatio-temporal scale used in data collection and grouping on the standardization of CPUE(catch per unit effort).We used the Chinese squid-jigging fishery in the northwestern Pacific Ocean as an example to evaluate 24 scenarios at different spatio-temporal scales,with a combination of four levels of temporal scale(weekly,biweekly,monthly,and bimonthly)and six levels of spatial scale(longitude×latitude:0.5°×0.5°,0.5°×1°,0.5°×2°,1°×0.5°,1°×1°,and 1°×2°).We applied generalized additive models and generalized linear models to analyze the24 scenarios for CPUE standardization,and then the differences in the standardized CPUE among these scenarios were *** study shows that combinations of different spatial and temporal scales could have different impacts on the standardization of ***,at a fine temporal scale(weekly)different spatial scales yielded similar results for standardized *** choice of spatio-temporal scale used in data collection and analysis may create added uncertainty in fisheries stock assessment and *** identify a cost-effective spatio-temporal scale for data collection,we recommend a similar study be undertaken to facilitate the design of effective monitoring programs.

When faced with danger,pufferfish react with both a fast-start escape response and inflation *** neural basis of these stress responses in the pufferfish has not been *** present study reveals that during inflation behavior,the buccal cavity expands and compresses as a pump to control the direction of water flow and draws water in and *** inflation involves a series of major anatomical modifications of the *** greatly enlarged first branchiostegal ray and its associated hyohyoideus abductor muscle are the key mechanisms responsible for this *** nerve branch innervating the hyohyoideus abductor muscle is highly developed,and its central origin at the motor nucleus of the VIIth cranial nerve was revealed by tract-tracing using the carbocyanine dye *** central connections from its origin were found to be several motor nuclei in the medulla and the spinal cord,the nuclei of cranial nerve III and IV in the mesencephalon,and the pretectalis superficialis and periventricular preoptic nuclei in the *** the sympathetic cells and the parvocellular part of the periventricular preoptic nucleus might play a neuro-endocrine role in the rapid movement of the buccal cavity during puffing behavior in this species of *** central circuit revealed by this study is hypothesized to mediate the C-start escape behavior and puffing behavior.

Food safety regulatory systems in Europe and China:A study of how co-regulation can improve regulatory effectiveness

Food safety has received a great deal of attention in both developed and developing countries in recent years. In China, the numerous food scandals and scares that have struck over the past decade have spurred significant food safety regulatory reform, which has been increasingly oriented towards the public-private partnership model adopted by the Europe Union＇s （EU） food safety regulatory system. This paper analyzes the development of both the EU＇s and China＇s food safety regu- latory systems, identifies the current challenges for China and additionally considers the role of public-private partnership. The success of co-regulation in the food regulatory system would bring significant benefits and opportunities for China. Finally, this paper recommends additional measures like training and grants to improve the private＇s sector effectiveness in co-regulating China＇s food safety issues.

Optimizing the key parameter to accelerate the recovery of AMOC under a rapid increase of greenhouse gas forcing

Atlantic Meridional Overturning Circulation(AMOC)plays a central role in long-term climate variations through its heat and freshwater transports,which can collapse under a rapid increase of greenhouse gas forcing in climate *** studies have suggested that the deviation of model parameters is one of the major factors in inducing inaccurate AMOC *** this work,with a low-resolution earth system model,the authors try to explore whether a reasonable adjustment of the key model parameter can help to re-establish the AMOC after its *** a new optimization strategy,the extra freshwater flux(FWF)parameter is determined to be the dominant one affecting the AMOC’s *** traditional ensemble optimal interpolation(EnOI)data assimilation and new machine learning methods are adopted to optimize the FWF parameter in an abrupt 4×CO_(2) forcing experiment to improve the adaptability of model parameters and accelerate the recovery of *** results show that,under an abrupt 4×CO_(2) forcing in millennial simulations,the AMOC will first collapse and then re-establish by the default FWF parameter ***,during the parameter adjustment process,the saltier and colder sea water over the North Atlantic region are the dominant factors in usefully improving the adaptability of the FWF parameter and accelerating the recovery of AMOC,according to their physical relationship with FWF on the interdecadal timescale.

Effective mechanical state prediction systems are critical to modern manufacturing systems and *** a method of deep learning algorithm,Recurrent neural network,（RNN） has been playing an increasingly important role in the field of time series *** order to solve the problem of hard training and gradient extinction of RNN model,a long short-term memory network（LSTM） algorithm is proposed and applied to the prediction of mechanical state（PMS）.On the basis of the motor bearing data,the simulation is carried *** at the non-stationary of bearing data,the empirical mode decomposition（EMD） is used to decompose the bearing data into stationary singles,and the intrinsic mode function（IMF） energy entropy is calculated as the feature of mechanical *** comparison with support vector regression machine（SVRM）,LSTM achieves a better result in the single-step prediction of mechanical *** shows that LSTM is superior in machine state prediction and monitoring.
