Objective:To use fetal heart quantification(fetal HQ)technology to compare the coarctation of the aorta(CoA)and normal fetal heart structure and systolic function and to assess whether there are abnormalities in the fetal heart structure and systolic function associated with ***:This prospective cohort study was conducted from May 2020 to December 2022 and involved 18–40-week-old singleton pregnancies and 30 fetuses diagnosed with CoA using fetal echocardiography at the General Hospital of Ningxia Medical University and Peking University First Hospital Ningxia Women’s andchildren’s Hospital,*** control group contained 60 normal *** following parameters were recorded and analyzed statistically:four-chamber view(4CV)end-diastolic long diameter,4CV epicardial–contralateral epicardial transverse maximum diameter,4CV global sphericity index(GSI),left ventricular(LV)and right ventricular(RV)24-segment end-diastolic diameter(EDD),24-segment sphericity index(SI),LV-fractional area change(LV-FAC),LV-longitudinal strain(LV-LS),RV-fractional area change(RV-FAC),RV-longitudinal strain(RV-LS),and LV and RV 24-segment transverse fractional shortening(FS).Measurement data were compared between the two groups using an independent sample t test,with P0.9,indicating high consistency,and a coefficient of variable ofhearts in the two groups revealed that when compared with the control group,the 4CV end-diastolic long diameter was shortened in

BET bromodomain inhibitor JQ1 regulates spermatid development by changing chromatin conformation in mouse spermatogenesis

As a BET bromodomain inhibitor, JQ1 has been proven have efficacy against a number of different cancers. In terms of male reproduction, JQ1 may be used as a new type of contraceptive, since JQ1 treatment in male mice could lead to germ cell defects and a decrease of sperm motility, moreover, this effect is reversible. However, the mechanism of JQ1 acting on gene regulation in spermatogenesis remains unclear. Here, we performed single-cell RNA sequencing (scRNA-seq) on mouse testes treated with JQ1 or vehicle control to determine the transcriptional regulatory function of JQ1 in spermatogenesis at the single cell resolution. We confirmed that JQ1 treatment could increase the numbers of somatic cells and spermatocytes and decrease the numbers of spermatid cells. Gene Ontology (GO) analysis demonstrated that differentially expressed genes which were down-regulated after JQ1 injection were mainly enriched in “DNA conformation change” biological process in early developmental germ cells and “spermatid development” biological process in spermatid cells. ATAC-seq data further confirmed that JQ1 injection could change the open state of chromatin. In addition, JQ1 could change the numbers of accessible meiotic DNA double-stranded break sites and the types of transcription factor motif that functioned in pachytene spermatocytes and round spermatids. The multi-omics analysis revealed that JQ1 had the ability to regulate gene transcription by changing chromatin conformation in mouse spermatogenesis, which would potentiate the availability of JQ1 in male contraceptive.

Eyeball loss due to severe ocular trauma,intraocular malignancy or infection often requires surgical treatment called orbital implant reconstruction to rehabilitate the orbital volume and restore the aesthetic ***,it remains a challenge to minimize the postoperative exposure and infection complications due to the inert nature of conventional orbital ***,we developed a novel Ca-Zn-silicate bioceramic implant with multi-functions to achieve the expected *** porous hardystonite(Ca2ZnSi2O7)scaffolds with triply periodic minimal surfaces(TPMS)-based pore architecture and graded pore size distribution from center to periphery(from 500 to 800μm or vice versa)were fabricated through the digital light processing(DLP)technique,and the scaffolds with homogeneous pores(500 or 800μm)were fabricated as *** graded porous scaffolds exhibited a controlled bio-dissolving behavior and intermediate mechanical strength in comparison with the homogeneous counterparts,although all of porous implants presented significant antibacterial potential against *** and ***,the pore size-increasing scaffolds indicated more substantial cell adhesion,cell viability and angiogenesis-related gene expression in ***,the gradually increasing pore feature exhibited a stronger blood vessel infiltrating potential in the dorsal muscle embedding model,and the spherical implants with such pore structure could achieve complete vascularization within 4 weeks in the eyeball enucleation rabbit ***,our results suggested that the novel antibacterial hardystonite bioceramic with graded pore design has excellent potential as a next-generation orbital implant,and the pore topological features offer an opportunity for the improvement of biological performances in orbital reconstruction.

Preoperative P-wave duration as a predictor of atrial fibrillation after coronary artery bypass grafting: A prospective cohort study with meta-analysis

Objectives:Reported prediction rules for postoperative atrial fibrillation(AF)have suffered from inconsistent results and controversy surrounding the predictive value of a preoperative P-wave duration(PreOPWD).This study examined PreOPWD as a predictor for AF after coronary artery bypass grafting(CABG).Methods:Two-hundred-and-ninety-nine patients with sinus rhythm before off-pump CABG were recruited into the ***'demographic and clinical data were evaluated *** were continuously monitored for the first seven postoperative *** logistic regressionwas used to determine significant predictors of *** were then combined with similar studies and a metaanalysis was ***:Postoperative AF was observed in 33.1%of 299 *** with AF were older,had a prolonged PreOPWD,higher incidences of hypertension,aortic regurgitation,and mitral regurgitation.A cut-off point of PreOPWD105ms achieved a specificity of 74%,and a sensitivity of 65%for predictive of *** analysis showed that PreOPWD105ms(odds ratio[OR]4.63,95%confidence intervals[CI]2.66 to 8.03,P<0.001),age60 years(OR 2.72,95%CI 1.51 to 4.90,P<0.01)and hypertension(OR 2.10,95%CI 1.08 to 4.07,P<0.05)independently predicted postoperative AF.A meta-analysis of this data combined with those of ten other studies showed that PreOPWD was greater in patients with POAF,with a weighted mean difference of 3.95 ms(95%CI 1.97 to 5.92,P<0.001).Conclusion:This study confirmed,among other predictive characteristics,that PreOPWD is a powerful independent predictor of POAF.

THE COMBINED TESTS OF OVERNIGHT 12-HOUR URINE E3/C RATIO AND FETAL heart RATE IN PERINATAL MONITORING

Using perinatal monitoring, we evaluated retrospectively 793 late pregnancies with regard to the overnight 12-hour urine estriol/creatinine ratio (E3/C) and fetal heart rate (FHR). The results revealed that l) FHR corrected the false positive rate of E3/C by 13.7% (93/677); 2) E3/C gave reliable results in perinatal prediction in 81 of the 116 suspicious FHR cases (70%, 81/116); 3) When both tests were abnormal the incidence of poor perinatal results increased by 83.7% (36/43), and the perinatal mortality (7%, 3/43) in patients with abnormal values in both methods was 35 times higher than that in patients with normal results in both tests (0.2%, 1/462); 4) Both negative FHR test and normal E3/C could be reassured, as the risk of death in uterus within a week of both normal tests was only 0.13% (1/793). It is suggested that the E3/C may be used as a first line test for perinatal monitoring, where E3/C value is lower than normal, FHR test is added to increase the reliability of perinatal diagnosis, especially in the moderate and severe hypertensive disorders of pregnancy.

Long interpregnancy interval and adverse perinatal outcomes: a retrospective cohort study

We conducted a retrospective cohort study of 9,552 women experiencing their second delivery between 2014 and 2016 at the international Peace Maternity and child Health Hospital to investigate the association between the interpregnancy interval(IPI)and adverse perinatal outcomes. With the 12–23-mon IPI as the reference category, logistic regression analyzes were used to examine associations between different IPIs(<12, 12–23, 24–59, 60–119, and ≥120 mon) and perinatal outcomes(gestational diabetes mellitus, premature membrane rupture, gestational hypertension, preterm birth, low birth weight, and macrosomia).Compared with the 12–23-mon IPI category, women with longer IPIs had a higher risk of adverse perinatal outcomes, and those with an IPI ≥120 mon had the highest risk of gestational diabetes mellitus and premature membrane rupture(adjusted odds ratio(OR) 1.76, 95% confidence interval(CI) 1.32–2.35 and adjusted OR 2.03, 95% CI 1.53–2.67, respectively). These results indicate that a longer IPI is associated with a higher risk of adverse perinatal outcomes and an IPI of ≥120 mon appears to be independently associated with a higher risk of gestational diabetes mellitus and premature membrane rupture.

Cancer cell genomes originate from single-cell mutation with sequential clonal and subclonal expansion of somatic mutation acquisition during pathogenesis,thus exhibiting a Darwinian evolutionary process(Gerstung et al.,2020;Nik-Zainal et al.,2012).Through next-generation sequencing of tumor tissue,this evolutionary process can be characterized by statistical modelling,which can identify the clonal state,somatic mutation order,and evolutionary process(Gerstung et al.,2020;Mcgranahan&Swanton,2017).Inference of clonal and subclonal structure from bulk or single-cell tumor genomic sequencing data has a huge impact on studying cancer *** state and mutation order can provide detailed insight into tumor origin and future *** the past decade,various methods for subclonal reconstruction using bulk tumor sequencing data have been ***,these methods had different programming languages and data input formats,which limited their use and ***,we established a web server for Clonal and Subclonal Structure Inference and Evolution(COSINE)of cancer genomic data,which incorporated twelve popular subclonal reconstruction *** deconstructed each method to provide a detailed workflow of single processing steps with a user-friendly *** the best of our knowledge,this is the first web server providing online subclonal inference based on the integration of most popular subclonal reconstruction *** is freely accessible at ***.

The effects of repeated propofol anesthesia on spatial memory and long-term potentiation in infant rats under hypoxic conditions

Propofol is widely used as an intravenous drug for induction and maintenance in general *** is a common complication during *** want to know the effect of propofol on spatial memory and LTP(Long-term potentiation)under hypoxic *** this study,84 seven-day-old SpragueeDawley rats were randomly assigned into six groups(n Z 14)-four control groups:lipid emulsion solvent t 50%oxygen(CO),lipid emulsion solvent t room air(CA),lipid emulsion solvent t 18%oxygen(CH),and propofol t 50%oxygen(propofoleoxygen,PO);and two experiment groups:propofol t room air(propofol eair,PA),and propofol t 18%oxygen(propofolehypoxia,PH).After receiving propofol(50 mg/kg)or the same volume of intralipid intraperitoneal(5.0 ml/kg),injected once per day for seven consecutive days,the rats were exposed to 18%oxygen,50%oxygen and air,until recovery of the righting *** found that the apoptotic index and activated caspase-3 increased in the PH group(P<0.05)compared with the PA group,fEPSP(field excitatory postsynaptic)potential and success induction rate of LTP reduced in all propofol groups(P<0.05).Compared with the PO group,the fEPSP and success induction rate of LTP reduced significantly in the PA and PH groups(P<0.05).Moreover,compared with CH group,the average time of escape latency was longer,and the number of platform location crossings was significantly reduced in the PH group(P<0.05).Thus,we believe that adequate oxygen is very important during propofol anesthesia.
