Dual roles ofα1,4-galactosyltransferase 1 in spermatogenesis of Drosophila melanogaster

Agrocybe cylindracea Polysaccharides repair the intestinal mucosal barrier in DSS-induced colitis by modulating the gut microbiota -macrophage axis

The Agrocybe cylindracea polysaccharides (ACP) have been shown to possess various health benefits. However， the anti-inflammatory effects of ACP and the mechanisms underlying these effects remain poorly understood. In this study， we delved into the potential of ACP in mitigating colitis induced by dextrose sodium sulfate (DSS) in C57BL/6J mice and explored the associated mechanisms. ACP demonstrated notable anti-inflammatory effects in the intestines， as evidenced by the restoration of various indicators such as increased body weight and colon length， decreased DAI score， and alleviation of pathological damage in the colon. Additionally， ACP upregulated the expression of ZO-1， Occludin， and Muc2， suggesting a restoration of the mechanical and mucus barriers of the intestinal mucosa. Furthermore， ACP facilitated the polarization of M2 macrophages and preserved intestinal immune balance. The impact of ACP on the biological barrier was further scrutinized. ACP ameliorated disruptions in the gut microbiota， particularly by enhancing the relative abundance of bacteria that produce short-chain fatty acids (Anaerostipes， Romboutsia， and Butyricicoccus). Remarkably， the ameliorative effects of ACP on colitis were compromised upon the removal of the microbial community， but were restored post fecal microbiota transplantation， indicating that the efficacy of ACP in alleviating colitis is dependent on the gut microbiota. Intriguingly， the regulatory influence of ACP on macrophages also hinged on the gut microbiota. In essence， ACP exhibits the potential to modulate the composition of the gut microbiota， fostering the growth of beneficial bacteria， which in turn promotes the polarization of colonic M2 macrophages and facilitates the repair of the intestinal mucosal barrier in DSS-induced colitis. Extrachromosomal DNA(ecDNA)is a form of circular DNA mostly found in tumor *** the typical chromosomal DNA,ecDNA is circular,self-replicating,and carries complete or partial gene *** ecDNA occurrence remains a rare event in cancer,recent studies have shown that oncogene amplification on ecDNA is widespread throughout many types of cancer,implying that ecDNA plays a central role in accelerating tumor *** has also been associated with increased tumor mutation burden,chromosomal instability,and even tumor microenvironment *** may be crucial in influencing tumor heterogeneity,drug sensitivity,oncogenic senescence,and tumor immunogenicity,leading to a worsening prognosis for tumor *** this way,several clinical trials have been conducted to investigate the importance of ecDNA in clinical *** this review,we summarize the biogenesis,characteristics,and current research methods of ecDNA,discuss the vital role of ecDNA-caused tumor heterogeneity in cancers,and highlight the potential role of ecDNA in cancer therapy.

Prophylactic hyperthermic intraperitoneal chemotherapy in patients with locally advanced gastric cancer after curative surgery:Final results of a phase Ⅱ trial

Objective: The trial was designed to evaluate the efficacy of prophylactic hyperthermic intraperitoneal chemotherapy(HIPEC) with cisplatin for patients with locally advanced gastric cancer(LAGC).Methods: Between March 2015 and November 2016, a phase II clinical trial was performed. Fifty consecutive patients with LAGC were randomly assigned to two groups: the experimental group(radical gastrectomy + HIPEC with cisplatin + adjuvant chemotherapy) and the control group(radical gastrectomy + adjuvant chemotherapy).Survival rates were closely ***: The 5-year overall survival(OS) rate of all patients was 80.0%. The 5-year OS rate in the experimental group was lower than that in the control group, at 75.8% and 88.2%, respectively, with no statistical *** addition, 5-year recurrence-free survival(RFS) rates of patients who underwent HIPEC or not were also 75.8%and 88.2%, respectively. In the multivariate analysis, only pT stage [risk ratio(RR)=7.079, P=0.018] was significantly associated with prognosis. The most common recurrence pattern was peritoneal recurrence in both groups. The experimental group had a lower incidence of peritoneal recurrence than the control group with no statistical ***: This trial clearly revealed that prophylactic HIPEC with cisplatin neither decrease the risk of peritoneal recurrence nor improve the prognosis of patients with LAGC. Thus, HIPEC with cisplatin is not recommended as a prophylactic treatment for peritoneal recurrence of LAGC after radical gastrectomy.

Characterization of core maize volatiles induced by Spodoptera frugiperda that alter the mating-mediated approach–avoidance behaviors of Mythimna separata

The fall armyworm(Spodoptera frugiperda) is an invasive species and a destructive pest of maize, which significantly impacts native species and communities via complex mechanisms like competition for resources. However, the interaction between S. frugiperda and local pests remains unclear. In this study, we determined that Oriental armyworm(Mythimna separata) females with different mating status displayed different approach–avoidance behaviors towards maize which was infested by S. frugiperda larvae. The virgin M. separata females were repelled, while the mated females were attracted by the S. frugiperda-infested maize. To further understand the olfactory mechanism of this phenomenon, seven volatiles induced by S. frugiperda in maize were characterized by gas chromatography and mass spectrometry(GC-MS), including trans-2-hexenal, linalool, trans-β-farnesene, cis-3-hexenyl acetate, β-caryophyllene, trans-α-bergamotene, and isopentyl acetate. Additionally, electrophysiological and behavioral assays of the seven compounds were performed using both virgin and mated females of M. separata. We determined that virgin and mated females displayed different responses to the herbivore-induced plant volatile(HIPV).Trans-β-farnesene was the core compound for repelling virgin females, and trans-2-hexenal was the key attractant for oviposition in mated female M. separata individuals. These findings help our comprehension of the relationships between maize pests and offer new possibilities for controlling them by olfactory-based strategies.
