This study reports on the novel and simple green method involving the use of apple (Malus domestica) and tomato (Solanum lycopersicum) extracts in the synthesis of electroactive layers of silver nanoparticles|graphene oxide (AgNPs|GO) and zinc oxide nanoparticles|graphene oxide (ZnONPs|GO). The surface morphology of the green synthesized nanocomposites was studied using High-Resolution Transmission Electron Microscopy (HRTEM), High-Resolution Scanning Electron Microscopy (HRSEM) while the elemental analysis was studied using Fourier Transform Infrared Spectroscopy (FTIR), Raman spectroscopy and X-Ray diffraction (XRD) and their optical properties were further characterised using Ultraviolet Spectroscopy (UV-vis). The electrochemical studies of these nanocomposites were achieved using cyclic voltammetry (CV) where an increase in electron conductivity of the AgNPs|GO and ZnONPs|GO nanocomposite was observed. Comparatively, the silver nanoparticulate-based platforms were observed to have superior electrochemical properties as opposed to the zinc oxide-based platform. The observed electrochemical activities of the synthesized nanocomposites are a good indication of their suitability as electroactive platforms towards the development of electrochemical sensors. Electrochemical sensors are popular in the Electrochemistry field because they may be developed using different methods in order to suit their intended analytes. As such, the synthesis of a variety of electrochemical platforms provides researchers with a vast range of options to select from for the detection of analytes.

Background: Ischemia-reperfusion injury of organ transplantation activates several mediators which may link the innate to the adaptive immune response. Down the cascade of TLRs, we selected to study the expression of Interferon Regulatory Factors (IRF)-3 and -7 inside human Kidney Transplanted (KTx) organs and the synthesis of IFNα, the main growth factor induced by them, in KTx aspiration biopsy cultures. Simultaneously, we tested their robustness in diagnosing Acute Rejection (AR). Methods: Fine-needle aspiration biopsies (F-nab) were performed either on day 7 or 14 post-KTx among stable patients or on the day of AR diagnosis. On Fnab cytopreparations, we studied IRF3 and IRF7 by the enzymatic avidin-biotin complex staining, and in a different group of cases we quantified IFNα by ELISA in 48 hours Fnab culture supernatants. Results: AR group showed a significantly up-regulated expression for IRF3 and IRF7, reaching Positive Predictive Values (PPV) of 0.824 and 0.8, respectively, as well as Negative Predictive Values (NPV) above 0.9 for both;IFNα presented a PPV = 1.0 and a NPV = 0.9. A variation in the results was noticed according to different immunosuppressive therapies. Conclusions: Our findings suggest that IRF3 and IRF7, and IFNα which they promote, may be very important players in the early days post-KTx, linking the innate with an adaptive response and triggering acute rejection. These differences were very clear-cut, lending consistency to our speculation. It would be important to scrutinize for other potential effects derived from these IRFs up-regulation which could be of clinical relevance.

Jatropha curcas is a species with a variety of uses. It is grown primarily for oil for biodiesel, but also has agronomic and medicinal applications. Two methods were evaluated for cryopreservation of seeds and zygotic embryos of J. curcas: desiccation followed by rapid immersion of seeds and embryos in liquid nitrogen (LN, -196°C), and vitrification of zygotic embryos. Prior to cryo-preservation, seeds were manually scarified and the moisture content (MC) of seeds and embryos was determined. Explants were disinfected after cryopreservation. Seed germination after LN exposure was 100%. Plantlet development was better in sand substrate than that in vitro. Survival of zygotic embryos after cryopreservation was also 100%, without significant differences between treatments. Optimal development (100%) and plantlet length (51.77 mm) were observed with embryos dried for 60 min to 9.4% MC under laminar flow prior to cryopreservation. Zygotic embryos subjected to the vitrification procedure did not withstand LN exposure. Survival data for non-cryopreserved embryos after each step of the vitrification procedure provided information about embryo tolerance to cryoprotectants.

Efficient technologies for gene silencing would be important to carry out functional analysis with P. brasiliensis genes, as well as for a better understanding of the biology and pathogenesis of this pathogenic fungus. Due to the fact that homologous recombination is unusual in P. brasiliensis, the development of knockout isolates is currently non-feasible. The goal of this work was to assess RNA interference (RNAi) technology as an alternative tool for gene silencing previously employed successfully in H. capsulatum. For this purpose, we built different inverted repeat transgenic hairpin constructs to down-regulate the PbGP43 and PbP27 genes known to codify for two fungal immunogenic proteins that elicit a strong immune response during experimental paracoccidioidomycosis. Using the RNAi strategy, a reduction in the mRNA levels of the PbGP43 and PbP27 genes was observed during the first 20 days after selection;however, in the transformed yeast cells, the gene silencing status proved non-stable through the assay. We demonstrated that electrotransformation was suitable to transform P. brasiliensis yeast cells and integrate the hairpin constructions;nonetheless, gene silencing was not stable along the experimental time. A detailed analysis of the underlying molecular RNAi machinery may provide further insights into the intracellular mechanism that governs this reverse genetic tool.

Biosynthesis is up-regulated in tumors and thus the demand for anabolic intermediates is increased. The metabolic routes providing the building blocks for macromolecules are thus a very attractive target as they are not normally up-regulated in a normal quiescent cell. Some routes for glycolysis-derived intermediates production have been identified, but these do not constitute the whole pool of biosynthetic molecules in the cell, as many of these derive from mitochondria in the Krebs cycle. Indeed, this metabolic pathway is considered a “biosynthetic hub” from which anabolism is fed. If a metabolite efflux is indeed occurring, anaplerotic reactions must keep a steady supply of substrates. In spite of this obvious relevance of anaplerosis, it has been poorly characterized in the malignant cell context. Glutaminolysis and and pyruvate carboxylation are two pathways that function in an anaplerotic fashion. In spite of the increasing evidence implicating these two processes in cancer metabolism their role as intermediate providers is overlooked. In this review we analyze the implications of an active anaplerosis in cancer and we discuss experimental evidence showing the relevance of these metabolic routes in tumor physiology.

Fault diagnosis technology has been widely applied and is an important part of ensuring the safe operation of mechanical *** response to the problem of frequent faults in rolling bearings,this paper designs a rolling bearing fault diagnosis method based on convolutional capsule network(CCN).More specifically,the original vibration signal is converted into a two-dimensional time–frequency image using continuous wavelet transform(CWT),and the feature extraction is performed on the two-dimensional time–frequency image using the convolution layer at the front end of the network,and the extracted features are input into the capsule *** capsule network converts the extracted features into vector neurons,and the dynamic routing algorithm is used to achieve feature transfer and output the results of fault *** different datasets are used to compare with other traditional deep learning models to verify the fault diagnosis capability of the *** results show that the CCN has good diagnostic capability under different working conditions,even in the presence of noise and insufficient samples,compared to other *** method contributes to the safe and reliable operation of mechanical equipment and is suitable for other rotating scenarios.

The limestone with smaller particle size provides rapid correction of soil acidity. However, the limestone with large particles can promote bigger root growth, and may still have a residual effect on the soil. The aim of this study was to evaluate the residual effect of the application and incorporation of different limestone with a particle size bigger than 0.30 mm in root growth, nutrition and yield of maize in Cerrado soil. A randomized completed block utilized in this experiment, with five treatments and four replications. The grain sizes of dolomitic limestone incorporated into the Oxisol were: 0 (control, no-lime), 2.00-0.82 mm, 0.82-0.56 mm, 0.56-0.30 mm, 0.30-0.20 mm, in doses of 6.5 t·ha-1, 3.9 t·ha-1, 2.6 t·ha-1 and 1.3 t·ha-1 respectively. The limestone applied in introduction no-tillage. The maize were evaluated of root attributes, nutrition and grain yield in two years 2009 and 2010. The use of limestone particle size of 2.00-0.82 mm, and the quantity of 6.5 t·ha-1 applied to the soil and incorporated showed elongation and root growth of maize thin depth of 20-40 cm in the soil. In that, same proposed management of the limestone was the highest maize grain yield in the first year. The rooting characteristics evaluated correlated with leaf Ca and Cu and maizegrain yield.

Thorn scrub vegetation in Mexico is distributed over 50 million ha, where native tree species are the source of forage, timber, firewood and charcoal. Research describing wood durability of species from this vegetation type has not been fully determined, nor classified according to international standards. Thus, the aim of this study was to determine and classify the natural durability of ten woody species. Their natural durability was determined according to the European Pre-Norm 807, the loss of dynamic modulus of elasticity （MOEdyo）（MPa） was determined and wood mass loss （g） after being exposed to Trametes versicolor and Coniophora puteana fungi. Wood durability was classified accord- ing to the European Norm 350-1. Highly significant differences （p 〈 0.001） were found between the durability of woody species. The more durable species with lower MOEdyn lost were Condalia hooked （57.5% ± 0.6%）, Havardia pallens （58.2% ± 0.4%） and Acacia schaffneri （58.9% ±6.3%）. Species with lower mass loss after exposed to Coniophora puteana were Ebenopsis ebano （6.3% ±1.9%）, Condalia hooked （8.6% ±2.3%） and Cordia boissieri （11.8% ±2.3%）. E. ebano （7.1% ±2.4%）, Condalia hooked （8.2% ± 2.5%） and Cordia boissieri （11.5% ± 3.1%） showed the lower mass lost after exposed to T. versicolor. According to European Norm 350-1, three woody species were classified as very durable and durable species.

Molecular Modeling of Potential Dual Inhibitors of HIV Reverse Transcriptase and Integrase

With the goal of suggesting dual inhibitors of HIV reverse transcriptase (RT) and integrase (IN), herein we report the molecular docking of an initial set of 556 compounds related to the pyridinone class. Docking with multiple crystallographic structures of HIV-1 RT led to 160 potential binders of RT interacting with key amino acid residues at the enzyme’s allosteric site. Compounds selected from the docking with RT were further docked with a crystallographic structure of HIV-1 IN. A total of 31 structures had the potential to make contacts with Mg2+ ions located in a small space between DNA and IN. Interactions with Mg2+ ions are relevant because they participate in the stabilization of the IN-DNA complex. In conclusion, 31 compounds synthetically accessible are proposed as dual inhibitors of RT and IN. It is hypothesized that the suggested compounds will inhibit RT by occupying the allosteric site for NNRTIs and will inhibit the catalytic activity of IN by destabilizing the IN-DNA complex. The main perspective of this work is the synthesis and biological testing of the candidate molecules.
