Targeted metabolomics reveals the aberrant energy status in diabetic peripheral neuropathy and the neuroprotective mechanism of traditional chinese medicine JinMaiTong

Diabetic peripheral neuropathy (DPN) is a common and devastating complication of diabetes, for which effective therapies are currently lacking. Disturbed energy status plays a crucial role in DPN pathogenesis. However, the integrated profile of energy metabolism, especially the central carbohydrate metabolism, remains unclear in DPN. Here, we developed a metabolomics approach by targeting 56 metabolites using high-performance ion chromatography-tandem mass spectrometry (HPIC-MS/MS) to illustrate the integrative characteristics of central carbohydrate metabolism in patients with DPN and streptozotocin-induced DPN rats. Furthermore, JinMaiTong (JMT), a traditional chinese medicine (TCM) formula, was found to be effective for DPN, improving the peripheral neurological function and alleviating the neuropathology of DPN rats even after demyelination and axonal degeneration. JMT ameliorated DPN by regulating the aberrant energy balance and mitochondrial functions, including excessive glycolysis restoration, tricarboxylic acid cycle improvement, and increased adenosine triphosphate (ATP) generation. Bioenergetic profile was aberrant in cultured rat Schwann cells under high-glucose conditions, which was remarkably corrected by JMT treatment. In-vivo and in-vitro studies revealed that these effects of JMT were mainly attributed to the activation of adenosine monophosphate (AMP)-activated protein kinase (AMPK) and downstream peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α). Our results expand the therapeutic framework for DPN and suggest the integrative modulation of energy metabolism using TCMs, such as JMT, as an effective strategy for its treatment.

Three New Hydroxytetradecenals from Amomum tsao-ko with Protein Tyrosine Phosphatase 1B and Glycogen Phosphorylase Inhibitory Activity

Direct conversion of syngas to aromatics over metal oxide and zeolite(OXZEO) composite catalysts is ***,the selectivity of more valuable products such as benzene,toluene and xylenes(BTX) is limited due to undesired secondary methylation of ***,we report that the introduction of SAPO-34 into MnCrO_(x)-ZSM-5 catalyst enhances significantly the formation of BTX without sacrificing the aromatics *** optimized conditions,the fraction of BTX in aromatics reaches 64.7% versus 28.9% over the catalyst without SAPO-34.A number of model reaction tests and characterizations reveal that SAPO-34 consumes partially the intermediates such as ketene,by converting them to light ***,the methylation of BTX by ketene to heavy aromatics is inhibited over the external acid sites of ZSM-5,leading to an enhanced BTX selectivity in the *** hybrid catalyst provides an efficient method for highly selective synthesis of BTX from syngas.

The Completeness of Accreting Neutron Star Binary Candidates from the chinese Space Station Telescope

A neutron star(NS)has many extreme physical conditions,and one may obtain some important information about an NS via accreting neutron star binary(ANSB)*** upcoming chinese Space Station Telescope(CSST)provides an opportunity to search for a large sample of ANSB *** goal is to check the completeness of the potential ANSB samples from CSST *** this paper,we generate some ANSBs and normal binaries under the CSST photometric system by binary evolution and binary population synthesis method and use a machine learning method to train a classification *** the Precision(94.56%)of our machine learning model is as high as before study,the Recall is only about 63.29%.The Precision/Recall is mainly determined by the mass transfer rate between the NSs and their *** addition,we also find that the completeness of ANSB samples from CSST photometric data by the machine learning method also depends on the companion mass and the age of the *** candidates with a low initial mass companion star(0.1 M_(⊙)to 1 M_(⊙))have a relatively high Precision(94.94%)and high Recall(86.32%),whereas ANSB candidates with a higher initial mass companion star(1.1 M_(⊙)to 3 M_(⊙))have similar Precision(93.88%)and quite low Recall(42.67%).Our results indicate that although the machine learning method may obtain a relatively pure sample of ANSBs,a completeness correction is necessary for one to obtain a complete sample.

Efficacy and safety of chinese herbal medicine as adjunctive therapy in sepsis patients with bloodstream infection:a propensity-matched analysis

OBJECTIVE:To investigate the efficacy and safety of chinese herbal medicine in treating sepsis patients with bloodstream ***:A 6-year retrospective study was carried out at a university hospital in *** sepsis patients with bloodstream infection were *** primary outcome was 28-day mortality after *** score method was used to adjust for possible confounding.28-day mortality was estimated by KaplanMeier analysis and compared using the log-rank *** regression analysis was carried out to identify factors impacting in-hospital mortality ***:Following the application of the propensity score method,a total of 176 patients were *** all-cause 28-day mortality in the control group and chinese herbal medicine group was 21.6%and 14.8%,***-Meier survival analysis showed that chinese herbal medicine was associated with a lower hazard ratio(HR)in all-cause 28-day death compared with the control group[HR=0.44,95%CI(0.22,0.90),P0.05).Blood-activating and stasis-eliminating herb administration was associated with reduced inhospital mortality among sepsis patients with bloodstream infection[HR=0.54,95%CI(0.34,0.94),Pchinese herbal medicine,especially the blood-activating and stasis-eliminating herb,might have certain efficacy and safety in treating sepsis patients with bloodstream *** should prescribe blood-activating and stasis-eliminating herb in treating these two coalescent critical diseases as long as no contraindications ***,further studies are needed to validate our results.
