The Florida Museum of Natural History＇s McGuire Center for Lepidoptera and biodiversity, on the University of Florida campus in Gainesville, Florida, has become one of the world＇s largest institutions for research on butterflies and moths, and an important research facility for insect science. The facility was constructed by combining the staff and merging the Lepidoptera holdings from the Allyn Museum of Entomology, the Florida State Collection of Arthropods and other University of Florida collections, and now includes over ten million specimens from all over the world, rivaling some of the largest Lepidoptera research collections globally. The facility includes a team of domestic and international researchers studying many areas of lepidopterology, including behavior, biodiversity, biogeography, ecology, genomics, physiology, systematics and taxonomy. In this paper, we introduce the McGuire Center, its staff, and the many research activities for researchers across entomological disciplines.

Genes and proteins expressed at different life cycle stages in the model protist Euplotes vannus revealed by both transcriptomic and proteomic approaches

Sexual reproduction first appeared in unicellular protists and has continued to be an essential biological process in almost all *** protists, which contain both germline and somatic genomes within a single cell, have evolved a special form of sexual reproduction called conjugation that involves mitosis, meiosis, fertilization, nuclear differentiation, genome rearrangement, and the development of unique cellular structures. The molecular basis and mechanisms of conjugation vary dramatically among ciliates, and many details of the process and its regulation are still largely unknown. In order to better comprehend these processes and mechanisms from an evolutionary perspective, this study provides the first comprehensive overview of the transcriptome and proteome profiles during the entire life cycle of the newly-established marine model ciliate Euplotes vannus. Transcriptome analyses from 14 life cycle stages(three vegetative stages and 11 sexual stages) revealed over 26,000 genes that are specifically expressed at different stages, many of which are related to DNA replication,transcription, translation, mitosis, meiosis, nuclear differentiation, and/or genome rearrangement. Quantitative proteomic analyses identified 338 proteins with homologs associated with conjugation and/or somatic nuclear development in other ciliates, including dicer-like proteins, Hsp90 proteins, RNA polymerase Ⅱ and transcription elongation factors, ribosomal-associated proteins, and ubiquitin-related proteins. Four of these homologs belong to the PIWI family, each with different expression patterns identified and confirmed by RT-qPCR,which may function in small RNA-mediated genome rearrangement. Proteins involved in the nonhomologous end-joining pathway are induced early during meiosis and accumulate in the developing new somatic nucleus, where more than 80% of the germline sequences are eliminated from the somatic genome. Whole-genome duplication(WGD) events are widespread across eukaryotes and have played a significant role in moulding the genetic architectures of diverse organisms. In the present study, the newly sequenced genome of a giant ciliated protist, Stentor roeselii, provides an opportunity for the analysis of the collinearity and retention of reciprocal best-hit genes between two Stentor species. As a main result, we have unveiled a previously undetected ancient WGD event shaping the genome of its congener, Stentor coeruleus, a model protist used in cytological and evolutionary studies. Genomes of two congeners, S. coeruleus and S. roeselii, are compared and analyzed, revealing that:(i)the former exhibits a much higher retention rate of colinear-gene pairs(28%) than does S. roeselii, and in S. coeruleus, 75% of genes that have a RBH hit in S. roeselii, have paralogs with high amino-acid identity, consistent with a WGD event in the lineage leading to S. coeruleus;(ii) the S. roeselii genome possesses extremely short intergenic regions, implying that the lengths of intergenic regions are under strong selection;(iii) the unique characteristics of introns may have been shaped in the common ancestor of heterotrichs;(iv) gene families that play a role in activities of multiple protein kinases and voltage-gated ion channels expanded rapidly in the ancestor of both taxa, possibly relating to the remarkable regenerative ability in Stentor. This study offers new insights into the evolutionary dynamics of ciliate genomes,with implications for understanding of the processes underlying the evolution of genomic complexity.

The Southwest Australian Floristic Region(SWAFR) is a global biodiversity hotspot with high plant diversity and endemism and a broad range of threatening processes. An outcome of this is a high proportion of rare and threatened plant species. Ongoing discovery and taxonomic description of new species, many of which are rare, increases the challenges for recovery of threatened species and prioritisation of conservation actions. Current conservation of this diverse flora is based on integrated and scientific evidence-based management. Here we present an overview of current approaches to the conservation of threatened flora in the SWAFR with a focus on active management through recovery and restoration that is integrated with targeted research. Key threats include disease, fragmentation, invasive weeds, altered fire regimes, grazing, altered hydro-ecology and climate change. We highlight the integrated approach to management of threats and recovery of species with four case studies of threatened flora recovery projects that illustrate the breadth of interventions ranging from In situ management to conservation reintroductions and restoration of threatened species habitats. Our review and case studies emphasise that despite the scale of the challenge, a scientific understanding of threats and their impacts enables effective conservation actions to arrest decline and enhance recovery of threatened species and habitats.

Performance of Hybrids between Weedy Rice and Insect-resistant Transgenic Rice under Field Experiments: Implication for Environmental Biosafety Assessment

Transgene escape from genetically modified （GM） rice into weedy rice via gene flow may cause undesired environmental consequences. Estimating the field performance of crop-weed hybrids will facilitate our understanding of potential introgression of crop genes （including transgenes） into weedy rice populations, allowing for effective biosafety assessment. Comparative studies of three weedy rice strains and their hybrids with two GM rice lines containing different insect- resistance transgenes （CpTI or Bt/CpTI） indicated an enhanced relative performance of the crop-weed hybrids, with taller plants, more tillers, panicles, and spikelets per plant, as well as higher 1000-seed weight, compared with the weedy rice parents, although the hybrids produced less filled seeds per plant than their weedy parents. Seeds from the F1 hybrids had higher germination rates and produced more seedlings than the weedy parents, which correlated positively with 1000-seed weight. The crop-weed hybrids demonstrated a generally enhanced relative performance than their weedy rice parents in our field experiments. These findings indicate that transgenes from GM rice can persist to and introgress into weedy rice populations through recurrent crop-to-weed gene flow with the aid of slightly increased relative fitness in F1 hybrids.

Distribution and Selective Effects of Vrn-A1,Vrn-B1,and Vrn-D1 Genes in Derivative Varieties from Four Cornerstone Breeding Parents of Wheat in China

Vernalization, the process of a long exposure to low temperature to induce flowering in plants, is essential for plants to adapt to cold winters. The presence of vernalization genes Vrn-A1, -B1, and -D1 in four cornerstone breeding parents of wheat in China （Funo, Mentana, Yanda 1817, and Bima 4） and 322 derivative varieties （mostly winter wheat） from these parents were determined using PCR based molecular method. The frequencies of the VRN-1 genes in these derivative varieties were in order of Vrn-Dl（67.1%）〉 Vrn-Bl（19.6%）〉 Vrn-Al（5.3%）, which are similar as the former conclusion that Vrn-D1 is associated with the latest heading time, Vrn-A1 the earliest, and Vrn-B1 intermediate values. The frequencies of Vrn-Al and Vrn-B1 loci in the derivative varieties from winter wheat zones were higher than that from spring winter zones. Based on the wheat breeding history in China and the fact of non-random distribution of Vrn-A1 and Vrn-B1 in the derivative varieties from the four parents, there could be a strong selective effect on VRN-1 genes in different regions where the derivative varieties were cultivated.

Taxodiorneria peizhongii Z. J. Ye, J. J. Zhang et S. H. Pan was regarded as a new Intergenerlc hybrid between Taxodlum mucronatum Tenore （as the female donor） and Cryptomeria fortunei Hoolbrenk ex Otto et Dletr （as the male donor）. To confirm the authenticity of the intergeneric hybrid, we analyzed the rbcL gene and the Internal transcribed spacer （ITS） of 26S-18S ribosomal RNA gene of the three species using polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP） and arbitrarily primed PCR （AP-PCR）, and ob- tained the following results：i） Taxodiomeria peizhongii had the same RFLP maps of the rbcL gene and the ITS as Taxodlum mucronatum, but was different from C. fortunei; II） a 311-bp PCR amplification product was obtained In C. fortunei by AP-PCR of ITS, but was not found in Taxodiomeria peizhongii. Our results have demonstrated that C. fortunei did not provide any genome for Taxodiomeria peizhongii, Implying that T. peizhongii Is not an Intergenerlc hybrid between the two species.

We examine the intersection of the FAIR principles(Findable,Accessible,Interoperable and Reusable),the challenges and opportunities presented by the aggregation of widely distributed and heterogeneous data about biological and geological specimens,and the use of the Digital Object Architecture(DOA)data model and components as an approach to solving those challenges that offers adherence to the FAIR principles as an integral *** approach will be prototyped in the Distributed System of Scientific Collections(DiSSCo)project,the pan-European Research Infrastructure which aims to unify over 110 natural science collections across 21 *** take each of the FAIR principles,discuss them as requirements in the creation of a seamless virtual collection of bio/geo specimen data,and map those requirements to Digital Object components and facilities such as persistent identification,extended data typing,and the use of an additional level of abstraction to normalize existing heterogeneous data *** FAIR principles inform and motivate the work and the DO Architecture provides the technical vision to create the seamless virtual collection vitally needed to address scientific questions of societal importance.

Intercropping of rice varieties increases the eff iciency of blast control through reduced disease occurrence and variability

Creating a crop-heterogeneous system by intraspecific mixtures of different rice varieties can substantially reduce blast diseases. Such variety mixtures provide an ecological approach for effective disease control, maintaining high yields with the minimum fungicide applications. Whether such an approach is universally applicable for random rice variety combinations and what is the variation pattern of the diseases under intercropping still remains unclear. We conducted two-year large-scale field experiments involving 47 rice varieties/lines and 98 variety-combinations to compare the occurrence of rice blast in monoculture and intercropping plots at multiple sites. In the experiments, the plant height of the selected tradi- tional varieties was about 30 cm taller, and their life cycle was 10 days longer, than that of the improved rice varieties. The monoculture included either traditional or modern rice varieties grown in separate plots. The intercropping included both traditional and modern rice varieties planted together in the same plots. Results from the field experiments under natural disease conditions demonstrated significant reduction for rice blast disease in intercropping plots, compared with that in monoculture plots. For traditional varieties, the average blast incidence reduced from -26% in monoculture to -10% in intercropping, and the disease severity reduced from -17 in monoculture to -5 in intercropping. For modern varieties, the average blast incidence reduced from -19% in monoculture to -10% in intercropping, and the severity from -10 in mono- culture to -4 in intercropping. Traditional rice varieties （-72%） had a much greater increase in the efficiency of disease control than modern varieties （-60%）. In addition, substantially lower values of variance in the blast incidence and severity was detected among the variety combinations in intercropping plots than in monoculture plots. An efficient molecular method for the accurate and efficient identification of indica and japonica rice was created based on the poly-morphisms of insertion/deletion(InDel) DNA fragments obtained from the basic local alignment search tool(BLAST) to the entire genomic sequences of indica(93-11) and japonica rice(Nipponbare).The 45 InDel loci were validated experimentally by the polymerase chain reaction(PCR) and polyacrylamide gel electrophoresis(PAGE) in 44 typical indica and japonica rice varieties,including 93-11 and Nipponbare.A neutrality test of the data matrix generated from electrophoretic banding patterns of various InDel loci indicated that 34 InDel loci were strongly associated with the differentiation of indica and japonica *** extensive analyses involving cultivated rice varieties from 11 Asian countries,and 12 wild Oryza species with various origins confirmed that indica and japonica characteristics could accurately be determined via calculating the average frequency of indica-or japonica-specific alleles on different InDel loci across the rice *** method was named as the ’’InDel molecular index" that combines molecular and statistical methods in determining the indica and japonica characteristics of rice *** with the traditional methods based essentially on morphology,the InDel molecular index provides a very accurate,rapid,simple,and efficient method for identifying indica and japonica *** addition,the InDel index can be used to determine indica or japonica characteristics of wild Oryza species,which largely extends the utility of this *** InDel molecular index provides a new tool for the effective selection of appropriate indica or japonica rice germplasm in rice *** also offers a novel model for the study of the origin,evolution,and genetic differentiation of indica and japonica rice adapted to various environmental changes.
