Effects of rhamnolipids on bacterial communities in a dioxin-contaminated soil and the gut of earthworms added to the soil

The biosurfactants rhamnolipids and the “soil ecosystem engineers” earthworms are often used to remediate contaminated soils. However, the effects of rhamnolipids on earthworm intestinal flora and microbial community in soil containing earthworms are not clearly understood. In our study, a 21-d microcosm experiment was carried out to reveal the effects of rhamnolipids on microbial abundance, composition, and metabolism, as well as contaminant degradation capacity. Both rhamnolipids and earthworms had positive effects on soil bacteria. Rhamnolipid-amended soil(RT) showed higher bacterial abundance and metabolic activity than earthworm-amended soil(ET), while the improvement in bacterial composition and contaminant degradation capacity by rhamnolipids was lower than that by earthworms. Notably, these effects were further amplified by the combined treatment of rhamnolipids and earthworms(RET). Specifically, the bacterial abundance(log-transferred) increased from 9.5 copies g-1in the control with no addition to 10.3, 10.6, and 11.1 copies g-1in ET, RT, and RET, respectively. Compared to ET, the relative abundance of the dominant phylum, Proteobacteria, increased from 41.66% to 51.67% in RET,and more pollutant-degrading bacteria were also enriched in RET. Therefore, the increases in bacterial abundance and contaminant-degrading bacteria led to the following ranking of soil dioxin removal rate: RET(77.28%) > ET(59.83%) > RT(24.65%) > control(4.71%). Moreover, the addition of rhamnolipids enhanced the abundance of bacterial functional genes involved in metabolism and environmental information processing. In addition, the composition and diversity of bacteria in the gut of earthworms were conspicuously affected by rhamnolipids, and the relative abundance of Microbacterium and Shewanella increased significantly(P < 0.05). Umbelliferone ameliorates renal function in diabetic nephropathy rats through regulating inflammation and TLR/NF-κB pathway

Diabetic nephropathy(DN) is a leading cause of renal failure, contributing to severe morbidity and mortality in diabetic patients. Umbelliferae(Umb) has been well characterized to exert protective effects in diabetes. However, the action and mechanism of Umb in DN remains unclear. In this work, we studied the effect of Umb in a streptozotocin(STZ)-induced DN rat model and explore its underlying mechanism. DN rats were treated with Umb(20, 40 mg·kg^(-1)) or irbesartan(15 mg·kg^(-1)) for 4 weeks. Levels of serum glucose, insulin, blood uric acid, creatinine, triglycerides(TG) and total cholesterol(TC) were measured by commercial assay kits,respectively. Histopathological changes and inflammatory cytokine levels including IL-6, IL-1β and TNF-α in the kidney were also evaluated. Alterations in the expression of podocin, CD2 AP and TLR/NF-κB were assessed by western blotting. Our results showed that Umb reduced renal injury in DN rat model, as evidenced by the decrease in blood glucose, 24 h urinary protein, serum creatinine,and blood uric acid. Umb also significantly ameliorated the renal histopathological alteration, and down-regulated the expression of epithelial-to-mesenchymal transition-related molecular markers podocin and CD2AP. Moreover, Umb inhibited TLR2, TLR4, MyD88 expressions, NF-κB activation and considerably reduced levels of other downstream inflammatory molecules(TNF-α, IL-6, IL-1β).These findings indicated that Umb improved renal function through regulating inflammation and TLR/NF-κB pathway, suggesting the potential efficacy of Umb in DN treatment.

Adsorption of 2-mercaptobenzothiazole from aqueous solution by organo-bentonite

The adsorption behavior of 2-mercaptobenzothiazole onto organo-bentonite was investigated. Natural bentonite from Gaozhou in Guangdong province, China was collected. Organo-bentonite was prepared by intercalation of cetyltrimethyl ammonium bromide into the natural bentonite. The physicochemical properties of the prepared organo-bentonite were characterized by X-ray diffraction, N2 adsorption-desorption isotherm and Fourier transform infrared spectroscopy. The results showed that montmorillonite is the main component of the natural bentonite. The basal spacing of the natural bentonite is 1.47 nm, which increased to 1.98 nm on intercalation with cetyltrimethyl ammonium bromide. Moreover, both the surface area and pore volume increased with intercalation. Clear CH2 stretching （3000-2800 cm-1） and scissoring （1480-1450 cm-1） modes of the intercalated surfactants were observed for organo- bentonite. Compared with the pseudo first-order kinetic model, the pseudo second-order kinetic model is more suitable to describe the adsorption kinetics of 2-mercaptobenzothiazole onto organo-bentonite. The adsorption capacity of 2-mercaptobenzothiazole onto organo-bentonite increased with increasing initial concentration of 2-mercaptobenzothiazole, but decreased with increasing adsorbent dosage. The adsorption isotherm of 2-mercaptobenzothiazole onto organo-bentonite fits well with the Langmuir model. The maximum adsorption capacity of organo-bentonite for 2-mercaptobenzothiazole was 33.61 mg/g, indicating that organo-bentonite is a promising adsorbent for 2-mercaptobenzothiazole.

The Qiangtang Basin（QB）, located in the central Tibetan Plateau, is a Jurassic marine basin and one of the most important prospective salt resource belts in China. In recent decades, many outcrops of gypsiferous bed have been found in the Jurassic marine strata in the basin. Salt springs with abnormally high sodium（Na-＋） contents had been identified in the Late Jurassic Xiali Formation（Fm.） in the basin in the last years. However, to date, no potash or halite deposits have been identified in the QB. Gypsum outcrops and salt springs are very important signs in the investigation of halite and potash deposits. Therefore, the Xiali Fm. is a potentially valuable layer to evaluate for the possible presence of halite and potash deposits in the basin. However, few studies have explored the formation conditions of evaporites in the unit. Here, we present detailed geochemical records from the Yanshiping section related to the study of the formation conditions of evaporites in the Xiali Fm. of the QB. Climate proxies based on the obviously increased anion concentrations of SO4（2-） and Cl-- and the significant correlation coefficients of Ca2＋-SO42-（R = 0.985） and Na-＋-Cl--（R = 0.8974） reveal that the upper member of the Xiali Fm.（the upper Xiali Fm.） formed under an arid climate and evolved into the sulfate phase or early chloride phase. Provenance proxies based on the obviously increased K-＋ and Na-＋ ion concentrations and the significant correlation coefficient of Na-＋-Cl--（R = 0.8974） suggest that the upper Xiali Fm. featured optimal provenance conditions for the possible formation of halite deposits. The regression and the semi-closed tidal flat environment in the upper Xiali Fm. were favorable for the formation of potash and halite deposits. The low Mg-（2＋） /Ca-（2＋） values（mean value = 1.82） and significant Na-＋-Cl-- correlation coefficient（R = 0.8974） also suggest that the upper Xiali Fm. is the layer most likely to contain potential halite deposits. The balance of acidity and redox capability over modified CeO_2 catalyst for the selective catalytic reduction of NO with NH_3

The effect of acidity and redox capability over sulfuric acid-modified CeO_2 catalysts were studied for the selective catalytic reduction of NO_x with NH_3(NH_3-SCR). The deposition of sulfate significantly enhanced the catalytic performance over CeO_2. NO_x conversion over4H_2SO_4/CeO_2 at 230–440 °C was higher than 90%. The strong redox capability of CeO_2 could result in unselective NH_3 oxidation and decrease high temperatures catalytic activity and N_2 selectivity. The deposition of sulfate increased the acidity and weakened the redox capability, and then increased the high temperature NO_x conversion and N_2 selectivity. An appropriate level of acidity also promoted the activity at 190–250 °C over ceria-based catalysts, and with further increase in the acidity, the SCR activity decreased slightly. Weak redox capability lowered the low-temperature catalytic activity. Excellent SCR activity requires a balance of acidity and redox capability on the catalysts.

To obtain the P8 protein of Rice gall dwarf virus (RGDV) with biological activity,its outer coat protein gene S8 was expressed in Spodoptera frugiperda (Sf9) insect cells using the baculovirus expression *** S8 gene was subcloned into the pFastBacTM1 vector,to produce the recombinant baculovirus transfer vector *** transformation,pFB-S8 was introduced into the competent cells (*** DH10Bac) containing a shuttle vector,Bacmid,generating the recombinant bacmid *** being infected by recombinant baculovirus rvpFB-S8 at different multiplicities of infection,Sf9 cells were collected at different times and analyzed by SDS-PAGE,Western blotting and immunofluorescence *** expression level of the P8 protein was highest between 48-72 h after transfection of Sf9 *** microscopy showed that P8 protein of RGDV formed punctate structures in the cytoplasm of Sf9 cells.

Potential of Ulva prolifera in phytoremediation of seawater polluted by cesium and cobalt: an experimental study on the biosorption and kinetics

Radionuclides accidentally released from nuclear power plants,such as ^(137)Cs and ^(60)Co,can lead to severe contamination of marine *** macroalgae are effi cient in absorbing metal elements from seawater.A 10-day simulation was conducted to assess the potential of scavenging cesium(Cs)and cobalt(Co)with live U lva prolifera that was cultured in seawater medium containing Cs and Co in diff erent concentrations(0(the control),10,20,50,and 100 mg/L).In the experiment,5 kg(fresh weight)of *** was cultured in natural seawater in 90-L *** showed that after the experiment,the average bioconcentration factors(BCFs)of the control group were 247.2 and 1126.4 for Cs and Co,*** absolute absorption quantity of *** increased and the BCFs decreased with the increase in Cs and Co *** biosorption of the two metals on the fi rst day well fi t the pseudo-second-order equation(R^(2)>0.95),indicating that adsorption is the rate-limiting step in the total biosorption *** of both metal ions declined signifi cantly in the fi rst hour and decreased by 25.2%and 15.5%in 48 h,***,live *** is effi cient at scavenging Cs and Co in seawater,providing potential applications for the phytoremediation of radionuclides contaminated seawaters.

Inactivated Sendai Virus Suppresses Murine Melanoma Growth by Inducing Host Immune Responses and Down-regulating β-catenin Expression

Abstract Objective This paper aims to investigate the anti-tumor mechanism of inactivated Sendai virus （Hemagglutinating virus of Japan envelope, HVJ-E） for murine melanoma （B16F10）. Methods The murine dendritic cells （DCs） were treated with HVJ-E, and then the cytokines secreted from DCs and costimulation-related molecules on DCs were measured. Meanwhile, the expression of 13-catenin in HVJ-E treated murine melanoma cells was detected. In addition, HVJ-E was intratumorally injected into the melanoma on C57BL/6 mice, and the immune cells, CTL response and tumor volume were analyzed. Results HVJ-E injected into B16F10 melanoma obviously inhibited the growth of the tumor and prolonged the survival time of the tumor-bearing mice. Profiles of cytokines secreted by dendritic cells （DCs） after HVJ-E stimulation showed that the number of cytokines released was significantly higher than that elicited by PBS （P〈O.05）. The co-stimulation-related molecules on DCs were comparable to those stimulated by LPS. Immunohistochemical examinations demonstrated the repression of 13-catenin in B16F10 melanoma cells after HVJ-E treatment. Meanwhile, real-time reverse transcription PCR revealed that HVJ-E induced a remarkable infiltration of CDllc positive cells, chemokine ligand 10 （CXCL10） molecules, interleukin-2 （IL-2） molecule, CD4^＋ and CD8^＋ T cells into HVJ-E injected tumors. Furthermore, the mRNA expression level of 13-catenin in the HVJ-E injected tumors was also down-regulated. In addition, B16F10-specific CTLs were induced significantly after HVJ-E was injected into the tumor-bearing mice. Conclusion This is the first report to show the effective inhibition of melanoma tumors by HVJ-E alone and the mechanism through which it induces antitumor immune responses and regulates important signal pathways for melanoma invasion. Therefore, HVJ-E shows its prospect as a novel therapeutic for melanoma therapy.

A new method for the retrieval of ocean wave parameters from SAR imagery is developed,based on the shape-from-shading(SFS)***,the SFS technique has been used in the reconstruction of 3D landform information from SAR images,in order to generate elevation maps of topography for land ***,in order to retrieve ocean wave characteristics,we apply the SFS methodology,together with a method to orient the angular measurements of the azimuth slope and range slope,in the measurement of ocean surface *** method is applied to high resolution fine-quad polarization mode(HH,VV,VH and HV)C-band RADARSAT-2 SAR imagery,in order to retrieve ocean wave spectra and extract wave *** in situ buoy measurements are used to validate the reliability of this *** show that the method can reliably estimate wave height,dominant wave period,dominant wave length and dominant wave direction from C-band SAR *** advantage of this method is that it does not depend on modulation transfer functions(MTFs),in order to measure ocean surface *** method can be used in monitoring ocean surface wave propagation through open water areas into ice-covered areas,especially the marginal ice zone(MIZ)in polar oceans.

Adjuvant Effects of Recombinant Plasmids of Porcine IL-4 and IFN-γ to Cysticercosis cellulosae Vaccines in Mice and Pigs

To investigate the adjuvant potential of porcine IL-4 and IFN-γ in mice and pigs, the genes of porcine IL-4 and IFN-γ were cloned and the recombinant mammalian expression plasmids were constructed for in vivo expression of the cytokines. Adjuvant effects of recombinant expression plasmids of IL-4 and IFN-γ （pcDNA-IL-4, pcDNA-IFN-γ） co-administrated with Cysticercus cellulosae crude antigen or TSOL18 recombinant protein antigen have been carried out in mice and pigs, respectively. We have demonstrated that recombinant plasmids of the cytokines as an adjuvant could induce stronger immune response in mice and pigs. With the C. cellulosae parasite antigen, porcine pcDNA-IL-4 induced higher specific antibody of immunized mice than pcDNA-IFN-γ. But pcDNA-IFN-γ is significantly stronger than that of no adjuvant or empty plasmids with the antigen control group. For the TSOL18 recombinant protein antigen vaccine, pcDNA-IL-4 still had a stronger ability to enhance specific antibody in swine than pcDNA-IFN-γ （P 〈 0.01）, but the immune protective rate was lower in challenged pigs （only 68.7%）. Although pcDNA-IFN-γ showed lower specific antibody, the protection rate was very high （91%） than other group （P 〈 0.01）. This study indicated that the recombinant expression plasmids of porcine IL-4 and IFN-γ display stronger adjuvant effects to C. cellulosae vaccine, further research should be carried out for understanding of the interaction mechanism.
