Nowadays,presynaptic dopaminergic positron emission tomography,which assesses deficiencies in dopamine synthesis,storage,and transport,is widely utilized for early diagnosis and differential diagnosis of *** review provides a comprehensive summary of the latest developments in the application of presynaptic dopaminergic positron emission tomography imaging in disorders that manifest *** conducted a thorough literature search using reputable databases such as PubMed and Web of *** criteria involved identifying peer-reviewed articles published within the last 5 years,with emphasis on their relevance to clinical *** findings from these studies highlight that presynaptic dopaminergic positron emission tomography has demonstrated potential not only in diagnosing and differentiating various Parkinsonian conditions but also in assessing disease severity and predicting ***,when employed in conjunction with other imaging modalities and advanced analytical methods,presynaptic dopaminergic positron emission tomography has been validated as a reliable in vivo *** validation extends to screening and exploring potential neuropathological mechanisms associated with dopaminergic *** summary,the insights gained from interpreting these studies are crucial for enhancing the effectiveness of preclinical investigations and clinical trials,ultimately advancing toward the goals of neuroregeneration in parkinsonian disorders.

Role of the globus pallidus in motor and non-motor symptoms of Parkinson's disease

The globus pallidus plays a pivotal role in the basal ganglia circuit. Parkinson's disease is characterized by degeneration of dopamine-producing cells in the substantia nigra, which leads to dopamine deficiency in the brain that subsequently manifests as various motor and non-motor symptoms. This review aims to summarize the involvement of the globus pallidus in both motor and non-motor manifestations of Parkinson's disease. The firing activities of parvalbumin neurons in the medial globus pallidus, including both the firing rate and pattern, exhibit strong correlations with the bradykinesia and rigidity associated with Parkinson's disease. Increased beta oscillations, which are highly correlated with bradykinesia and rigidity, are regulated by the lateral globus pallidus. Furthermore,bradykinesia and rigidity are strongly linked to the loss of dopaminergic projections within the cortical-basal ganglia-thalamocortical loop. Resting tremors are attributed to the transmission of pathological signals from the basal ganglia through the motor cortex to the cerebellum-ventral intermediate nucleus circuit. The cortico–striato–pallidal loop is responsible for mediating pallidi-associated sleep disorders. Medication and deep brain stimulation are the primary therapeutic strategies addressing the globus pallidus in Parkinson's disease. Medication is the primary treatment for motor symptoms in the early stages of Parkinson's disease, while deep brain stimulation has been clinically proven to be effective in alleviating symptoms in patients with advanced Parkinson's disease,particularly for the movement disorders caused by levodopa. Deep brain stimulation targeting the globus pallidus internus can improve motor function in patients with tremordominant and non-tremor-dominant Parkinson's disease, while deep brain stimulation targeting the globus pallidus externus can alter the temporal pattern of neural activity throughout the basal ganglia–thalamus network. AZD1775 synergizes with SLC7A11 inhibition to promote ferroptosis

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. Considering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced compensatory upregulation of SLC7A11, which conferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the combination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

CapOX as neoadjuvant chemotherapy for locally advanced operable colon cancer patients: a prospective single-arm phase Ⅱ trial

Objective： The aim of this prospective, single-arm phase II trial was to confirm the safety and efficacy of neoadjuvant chemotherapy（NAC） using oxaliplatin plus capecitabine（Cap OX） for patients with operable locally advanced colon cancer（CC）.Methods： Patients with computed tomography-defined T4 or lymph node-positive CCs were enrolled. After radiological staging, patients were treated with at least 2 cycles of NAC consisting of 130 mg/m2 oxaliplatin on d 1,plus 1,000 mg/m2 capecitabine twice daily for 14 d every 3 weeks, followed by surgery, and then with the rest cycles of adjuvant chemotherapy. Radiological response was evaluated after 2 cycles of NAC. Tumor response, treatment toxicity, and surgical complications were recorded. The pathological response to therapy was evaluated according to the tumor regression grade（TRG） score. The primary endpoint was pathologic tumor response. This trial is registered in Clinical ***（No： NCT02415829）.Results： Forty-seven patients were enrolled in the study. Forty-two patients completed the planned *** total radiological response rate was 68%（32/47）, including complete and partial response rates of 2%（1/47）and 66%（31/47）, respectively. Stable disease was observed in 32%（15/47） and progressive disease was observed in none. Complete pathologic response, major regression, and at least moderate regression were achieved in 1（2%）, 2（4%）, and 29（62%） patients, respectively. Four patients developed grade 3 treatment toxicities. One patient with wound infection occurred after operation（1/47, 2%）. There was no treatment-related ***： Our results suggest that NAC with Cap OX is an effective and safe treatment option for patients with locally advanced CCs.

Uncertainty of an automatic system for counting alpha tracks on CR-39

For accurate counting of alpha tracks on the polyallyl diglycol carbonate of CR-39-type track detectors,the size distributions of both artifact tracks and alpha tracks were investigated with an automatic counting system. At the same temperature and etchant concentration, the numbers and sizes of alpha tracks changed significantly with the etching time, and the artifact track changes were smaller. At the etching time of 5 h, the sizes of alpha tracks were evidently larger than those of the artifact tracks, and the deviation of its size distribution was much smaller than those of longer etching time. Based on the size distribution of alpha tracks etched for 5 h, the overlap effect and uncertainty of overlap correction were studied by the Monte Carlo simulations for different track densities. It was found that the counting uncertainty of the system could be less than 6% in a density range of 10–160 tracks mm^(-2) after taking the overlap correction into account.

Semaphorin3B modulates radiosensitivity of human glioma U-87MG cells

This study was to determine the Semaphorin3B(SEMA3B) role in glioma cells responding to irradiation. Two glioma cell lines,which were used here was wild-type p53(U-87MG),and the other was harboring mutated p53 (U-251).The SEMA3B mRNA could be detected in the two cell *** expression level of SEMA3B mRNA was higher in U-87MG cells than in U-251 cells,and increased with time in U-87MG cells after *** of SEMA3B expression by shRNA decreased the radiosensitivity of U-87MG cells,this may be associated with the increased G2 accumulation after *** addition,G2 accumulation after irradiation was enhanced in SEMA3B low-expressing U-87MG *** results showed that the SEMA3B was implicated in glioma cells responding to irradiation.

A new sampler for simulating aerosol deposition in the respiratory tract

For estimation of the deposition fractions of radon progeny in different regions of the respiratory tract, a new sampler consisting of three different configurations of sampling heads was developed. The deposition fractions of aerosols on the wire screens inside the sampling heads were calculated with the fan model of filtration theory. The deposition fractions of aerosols in different regions of the respiratory tract were calculated with the lung dose evaluation program (LUDEP)developed by National Radiological Protection Board (NRPB) as references. In general indoor and mine environments, the deviation between the deposition fractions of attached aerosol on the wire screens designed in this study and its reference values in the respiratory tract is less than 5%. It is possible to accurately estimate the deposition fractions of radon progeny in different regions of the respiratory tract through mimic measurements of radon progeny collected with the new sampler.

Comparisons of dosimetric properties between GD-300 series of radiophotoluminescent glass detectors and GR-200 series of thermoluminescent detectors

In order to compare the dosimetric properties of two kinds of solid state detectors used for monitoring ex- ternal exposures,experiments were performed for the GD-300 series of radiophotoluminescent glass detectors (RPLGD)and the GR-200 series of thermoluminescent detectors(TLD).X-rays and ^(137)Cs and ^(60)Coγ-rays were used to irradiate the RPLGDs and TLDs,their dose linearity,uniformity/reproducibility of signal,fading effect and energy dependence were *** kinds of the detectors have generally good dosimetric *** dose linearity and energy dependence of the GD-351 dosimeters and the TLDs are nearly the same,but the RPLGD is much better than the TLD in terms of the uniformity and fading effect.
