Efficacy of Renshen Sanqi Chuanxiong formula for preventing vascular aging

OBJECTIVE:To evaluate the efficacy of Renshen Sanqi Chuanxiong formula(RSCF)for preventing vascular aging,and to investigate the possible molecular mechanism underlying the actions of ***:Potentially active components and their relatively direct targets were identified by combining drug-likeness(DL)screening using a target identification *** aging-associated targets for RSCF were obtained by selecting common genes not only from potential targets but also from human vascular aging-associated *** 3.2.1 software was employed to visualize the complex compound-target and target-function *** process and molecular function were assessed,and the Kyoto Encyclopedia of Genes and Genomes and pathway enrichment analyses were performed using Clue *** directly associated with vascular aging were integrated into a"vascular aging-related"***:Altogether,122 potentially active components of RSCF were identified through DL screening,and their corresponding 692 direct targets were retrieved via target prediction and *** identified 49 vascular aging-associated targets for RSCF by overlapping the 692 potential targets with 146 human vascular aging-associated *** results from the compound-target network indicated that most components acted on common targets and displayed synergistic action,which showed that the magnifying effects of RSCF were based on these common *** target-function network revealed that each target was involved in multiple function modules,suggesting that RSCF was multi-functional during treatment of vascular *** results of the Clue GO analysis indicated that most of the targets were associated with the hypoxia-inducible factor 1(HIF-1)signaling *** results from the pathway analysis also indicated that an integrative vascular aging-related pathway mainly included an angiogenesis regulation module,cell-survival module,and oxidative stress-resistance ***

Evidence for a therapeutic effect of Braintone on ischemic brain damage

This study used a novel combination of in vivo and in vitro experiments to show that Braintone had neuroprotective effects and clarified the molecular mechanisms underlying its efficacy. The Chinese herbal extract Braintone is composed of Radix Rhodiolase Essence, Radix notoginseng Essence, Fofium Ginkgo Essence and Rhizoma Chuanxiong. In vivo experiments showed that cerebral infarction volume was reduced, hemispheric water content decreased, and neurological deficits were alleviated in a rat model of permanent middle cerebral artery occlusion after administration of 87.5, 175 or 350 mg/kg Braintone for 7 consecutive days. Western blot analysis showed that Braintone enhanced the expression of hypoxia-inducible factor la, heme oxygenase-1 and vascular endothe- lial growth factor in the ischemic cortex of these rats. The 350 mg/kg dose of Braintone produced the most dramatic effects. For the in vitro experiments, prior to oxygen-glucose deprivation, rats were intragastrically injected with 440, 880 or 1 760 mg/kg Braintone to prepare a Braintone-co ntaining serum, which was used to pre-treat human umbilical vein endothelial cells for 24 hours. Human umbilical vein endothelial cell injury was alleviated with this pre-treatment. Western blot and real-time PCR analysis showed that the Braintone-containing serum increased the levels of hy- poxia-inducible factor la mRNA and protein, heine oxygenase-1 protein and vascular endothelial growth factor mRNA in oxygen-glucose deprived human umbilical vein endothelial cells. The 1 760 mg/kg dose produced the greatest increases in expression. Collectively, these experimental findings suggest that Braintone has neuroprotective effects on ischemia-induced brain damage via the up-regulation of hypoxia-inducible factor la, heme oxygenase-1 and vascular endothelial growth factor expression in vascular endothelial cells.

Current Status and Future Perspective in the Globalization of Traditional Chinese Medicines

Globalization of traditional Chinese medicines started around 1996, which was initiated by the Chinese government. However, substantial progress was only achieved in recent years including the adoption of TCM quality monographs in the western pharmacopoeias(United States Pharmacopoeia and European Pharmacopoeia) and registration in main stream drug regulatory agencies such as US Food and Drug Administration(FDA) and European Medicines Agency(EMA). So far, several TCM herbal quality monographs were adopted by the United States Pharmacopoeia including Chinese Salvia, Ganoderma lucidum and panax notoginseng, etc. Over 45 TCM quality monographs were recorded in the European Pharmacopoeia with 20 more in progress. After the successful registration of the first TCM product named Diao Xin Xue Kang as traditional medicine via the Medicines Evaluation Board of the Netherlands, several other TCM herbal products are in the registration process in several European member states. So far, there has been still not any TCM product authorized as a drug by the FDA regardless of a few TCM products in phase III or phase II clinical trials. This review summarizes the progress made in the globalization of traditional Chinese medicines in recent years and future issues in this regard.

Anti‑inflammatory and Cytotoxic Triterpenes from the Rot Roots of panax notoginseng

Four new protopanaxatriol-type triterpenes(1-2)and glucosides(3-4),were isolated from the rot roots of panax notoginseng(Burk.)Chen,along with four known ones(5-8).Their structures were elucidated on the basis of extensive spectroscopic analysis(HRESIMS,NMR,UV,IR,and OR)and acidic *** possible transformation pathway of these compounds were also speculated from ginsenoside Rg_(1).Compound 1,with a uniqueα,β-unsaturated ketene in its side chain,showed significant inhibitory effects against NO production on Murine macrophage cells(IC_(50)=4.12±0.20μM)and comparable cytotoxicities against five human cancer cell lines(myeloid leukemia HL-60,lung cancer A-549 cells,hepatocellular carcinoma SMMC7721,breast cancer MCF-7,and colon cancer SW480)to positive control,cisplatin(DDP).

Alzheimer’s disease is a complex neurodegenerative ailment accompanied by cognitive problems and progressive memory *** main causes of the disease are indistinguishable even though aging is said to be the key risk factor,along with the hypotheses such asβ-amyloid plaques,tau-proteins,neuroinflammation,and oxidative ***,Alzheimer’s disease is not curable,and the available medicines are only used to provide symptom *** and researchers have developed numerous models to study Alzheimer’s disease,such as in silico methods,in vitro models of cultured rat cortical neurons,in vivo rodent models involving mice and rats,and non-rodent models as *** review provides an insight into the modality of Alzheimer’s disease treatment using medicinal plants that are natural sources with abundant bioactive substances,and traditional formulations such as an extract from notoginseng Radix et Rhizoma,Chinese herbal formulations(i.e.,traditional Chinese medicine kidney-nourishing formula,supplements such as coenzyme Q10,alpha-lipoic acid,Ginkgo Biloba,omega-3’s,and acetyl-L-carnitine).In addition,we have discussed the evaluation parameters for experimental Alzheimer’s disease,which are used to assess the mechanism of memory and pharmacological effects of molecules for drug discovery and *** common experimental analysis methods for Alzheimer’s disease include behavioral,biochemical,and histological *** work will allow researchers to access an informative reference of traditional medicines along with experimental analysis methods in one place for Alzheimer’s disease research.

STUDIES ON THE ANTI-FATIGUE EFFECT OF THE SAPONINS OF STEMS AND LEAVES OF panax GINSENG(SSLG)

Effects of Raphani Semen on anti-fatigue and pharmacokinetics of panax ginseng

Objective: To explain the phenomenon that panax ginseng is not compatible with Raphani Semen based on pharmacodynamics and ***: The forced swimming time and biochemical parameters such as blood lactate(BLA), serum urea nitrogen(SUN), and hepatic glycogen(GLU) were determined for anti-fatigue experiment. The UPLCMS/MS was used to analyze the pharmacokinetics of Rg1, Re, Rb1, and Rd after orally administration of P. ginseng and P. ginseng combined with Raphani Semen to rats. Pharmacokinetic differences of four ginsenosides between single uses of P. ginseng and combined with Raphani Semen were ***: The results showed that Raphani Semen tended to significantly reduce the anti-fatigue activity of P. ginseng. Co-administration of P. ginseng and Raphani Semen had significant effects on the pharmacokinetics of the four ginsenosides in rats compared to that observed with P. ginseng extract alone. The AUC0–12 hvalues of the four ginsenosides in PG group were higher than the corresponding values in the PR group. It can be inferred that Raphani Semen decreased the blood exposure of the four ginsenosides in rats when it combined with P. ***: The anti-fatigue activity and pharmacokinetic results showed that Raphani Semen may reduce the pharmacological actions of P. ginseng.

Advances on hormone-like activity of panax ginseng and ginsenosides

Traditional Chinese medicine(TCM)has been paid much attentions due to the prevention and treatment of steroid hormone ***,the root of panax ginseng ***(Araliaceae),is one of the most valuable herbs in complementary and alternative medicines around the world.A series of dammarane triterpenoid saponins,also known as phytosteroids,were reported as the primary ingredients of Ginseng,and indicated broad spectral pharmacological actions,including anti-cancer,anti-inflammation and *** skeletons of the dammarane triterpenoid aglycone are structurally similar to the steroid *** in vitro and in vivo studies showed that Ginseng and its active ingredients have beneficial hormone-like role in hormonal *** review thus summarizes the structural similarities between hormones and dammarane ginsenosides and integrates the analogous effect of Ginseng and ginsenosides on prevention and treatment of hormonal disorders published in recent twenty years(1998–2018).The review may provide convenience for anticipate structure-function relationship between saponins structure and hormone-like effect.
